HISTOLOGY PRACTICAL HP1: Cell

Aims: 1. To demonstrate how to operate the light microscope properly. 2. To demonstrate the features of the cell and its components. Objectives: At the end of this session the student should be able to: 1. operate the light microscope properly. 2. familiarize with some stains used in histological preparations. 3. recognize various cell shapes and sizes. 4. recognize cell organelles and inclusions that are visible under the light microscope.

57. Liver (H&E) Note, under low power magnification, the arrangement of liver cells in the form of anastomizing cords. Note, under high power magnification, the following features of liver cells: a. Cytoplasm; b. Nucleus; c. Nucleolus; d. Chromatin 22. Spinal Cord (H&E) 23. Spinal Cord (Cresyl violet and Luxol fast blue) Observe, under low power magnification, the motor neurons which are seen as large cells in the anterior horn of the spinal cord. Note the nucleus, nucleolus and chromatin. 5. Epididymis (Silver) The tubular epididymis is seen in cross-section. Note the golgi apparatus which is seen as a black network at the apical region of the cells. The silver stain rendered the golgi apparatus a black colour.

59. Liver (PAS and H) Observe, under high power magnification, the reddish-stained glycogen granules within the cytoplasm. 3. Skin, pigmented (H&E) Observe, under high power magnification, the cells at the deepest layer of the epidermis of the skin. Note the brownish granules of melanin pigments. This brownish colour is the natural colour of the pigments and is not due to the stains.

61. Pancreas (H&E) Observe, under high power magnification, the pancreatic acinar cells. Note: a. The acidophilic zymogen granules at the apical region of the cells. b. The basophilic stain of the basal region of the cells.
