The Effect of soybean Extract on hampering Decreasing Bone Density in Long-term Corticosteroid Therapy in Male White Rat {Rattus norvegicus)
Abstrak : Soybean extract containing phytoestrogen, a group of plant non-steroid mixture, has esfrogenic capability but has less side effect than estrogen when it is given in long-term therapy. It has a role in cell with same way as estrogen to decrease bone resorption with the result that be able to prohibit loosening bone mass, known as antiresorptive agent. This antiresorptive mechanism is believed have a role in hampering decreasing bone density in long-term corticosteroid therapy. The objective of this study was demonstrating the effect of soybean extract on hampering decreasing bone density in long-term corticosteroid therapy. Experimental animals used in this study were three month old male wistar strain white rats. These rats were randomly divided into six groups. They consist of a control group, soybean extract group with different dose. After 100 daysn, the bone density was measured at metaphyseal part of femoral bone of all the experimental animal using ultrasound densitometry DBM Sonic 1200 and serum alkali phosphatase using optimalized standar method according Deutsche Gessellschaft for Chronic recommendation. The mean of bone density and serum alkali phosphatase level were influenced by administering soybean extract and corticosteroid (prednisolon) with p < 0.05. Pairwise comparisons showed a significant difference in mean bone density between control group compare to soyben extract Soup (p < 0.05) and to corticosteroid group (p < 0.05). No significant differences between control group compare to three groups which were administered corticosteroid and soybean extract with different dose (p > 0.05), and there was decreasing the value of p among them. There was a significant difference in mean bone density between soybean extract group and corticosteroid group (p < 0.05). No significant difference between soybean extract compare to three groups which were administered corticosteroid and soybean extract with different dose (p > 0.05). A significant difference also in mean bone density between corticosteroid group compare to three groups which were administered corticosteroid and soybean extract with different dose (p < 0.05). No significant differences in the level of serum alkali phosphatase in almost all of groups (p > 0.05). By using linier regression correlation test was proven that there was a correlation between the raising dose of soybean extract administration with the increasing on hampering decreasing bone density and the increasing serum alkali phosphatase level with each p were 0.000 and 0.000 (.p < 0.05). This study concluded that administration of soybean extract in long-term corticosteroid therapy could hamper decreasing bone density, and could also increasing serum alkali phosphatase level but could not be used as the marker of increasing osteoblast activity. Increasing dose of the soybean extract administraticn in long-tenn corticosteroid therapy had a tendency be able to increase hampering decreasing bone density.
