Tentative sequence of the experiments to be covered

1. Preparation and sterilization of media for animal tissue culture (Complete and
Incomplete).
2. Isolation, identification and primary cell culture of human cheek cells.
3. Estimation of leucocytes (WBC) by Total leukocyte count (TLC) and Differential
leukocyte count (DLC) using permanent slides.
4. Calculation of cell viability in the isolated cells using Trypan Blue assay.
5. To learn calculation of cell concentration (serial dilution technique) in order to set
up various types of assays, using Haemocytometer
6. To analyse and differentiate between suspension and adherent cell lines using
inverted microscope.
7. Subculturing (Splitting and Trypsinization) of suspension and adherent cell-lines.
8. Preparation of growth curve and calculation of doubling time for suspension cell
line.
9. Cryo-preservation and resuscitation of Frozen Cell Lines.
10.Cytotoxicity evaluation of the given compound: MTT assay.
11.Detection of fungal contaminants in cell culture
12.Preparation of cell lysate for various biological assays
13.Determination of antioxidant status of cultured cells: SOD activity under the effect
of a given drug/compound.
14.Determination of oxidant status of cultured cells: NO enzyme activity under the
effect of a given drug/compound.
15.To asses membrane integrity in cell line: lactate dehydrogenase (LDH) assay
