Escherichia coli, is not inhibited by bile salts and crystal violet and is a gram-
negative bacterium. The pink color of the bacterial growth indicates E. coli is
able to ferment lactose.
Salmonella typhimurium, is not inhibited by bile salts and crystal violet and is a
gram-negative bacterium. The absence of color in the bacterial growth indicates
S. typhimurium is unable to ferment lactose.
OBSERVATION
Pseudomonas aeruginosa: Green colonies with typical matted surface and rough
periphery, non-fermentative.
Escherichia coli: Opaque yellow colonies with a slightly deeper yellow center,
indicating fermentative.
OBSERVATION
V. cholera and its biotype Eltor ferment sucrose, which results in a pH shift and
production of yellow-brown colonies.
OBSERVATION
Colonies of Salmonella typhi are typically with pinpoint black centres.
Salmonella spp. that produce H2S appear as blue-green colonies with black
centers. H2S producers form black-centered colonies in the presence of ferric
ammonium citrate and sodium thiosulfate.
Salmonella typically produce clear colonies with distinct black centres (hydrogen
sulphide: H2S +) on HE.
OBSERVATION
S. aures: Beta Hemolysis, complete lysis of Red Blood Cells, causing a clearing of
blood from the medium under and surrounding the colonies.
OBSERVATION
S. aures: positive result with yellowing of red/pink medium, growth indicated.
OBSERVATION
Haemophilus influenzae: Non hemolytic, opaque cream-to-gray colonies
(accompanying Sheep blood agar shows no growth)
