BIOCHEM FORMAL REPORT

CARLA

Abstract

This experiment aims to prepare different buffer solutions, determine the pH of each buffer through colorimetry and
electrometry, and calculate the buffer capacity of prepared buffer solution. The group was assigned to develop a 0.50
M phosphate buffer with a pH of 7. Disodium phosphate as weak acid and sodium hydroxide as conjugate base was
incorporated as the buffers reagents. Moreover, to obtain the exact pH of the buffer solution, this was adjusted by
using 0.1 M HCl and NaOH and closely monitoring it through the pH meter. The buffers prepared with varying pH
were, then, subjected in colorimetric determination using different acid-base indicators namely bromophenol blue,
bromocresol green, bromocresol purple, phenol red, methyl red, methyl orange, phenolphthalein, and thymol blue.
The results should have shown the colors light green, blue, yellow, yellow, pink, orange, colorless, and yellow-
orange respectively.
