• by: use 'by' when talking about a general way to travel.

I go to work by bus.

She goes to the beach by bike.

We go to work by car.

• in: use 'in' when you sit inside the vehicle.

I am in the car.

They are in the helicopter.

He is in the truck.

• on: use 'on' when you can stand up or walk within the vehicle.

I am traveling on the bus.

We just got on the airplane.

They are on the subway.
