PROTOCOL EM QUANTIFICATION: MITOCHONDRIA SIZE, NUMBER, SHAPE

-Open image1 of animal 1


-Analyze  Tools  Grid 20 microns^2
-Analyze 1 section of every 10 sections and alternate the series of squares (those that
fall into the orange line) we quantify:
Section 1

Section 10

Section 20

We would keep going down in the horizontal rows for section 30, 40, etc
Section 100
We keep going in the vertical columns and then we start over

-Zoom in placing the mouse in the square we are going to quantify


-Draw all the mitocondrial outlines with the “Tool” “Free hand selection”, holding the
shift key

-Press Ctrl+M to obtain the area these mitochondria occupy in the square

-Count the outlines we have drawn to know the number of mitocondria

*Only outline the mitochondria bodies that conform easily distinguishable units and
are inside the limits of the square without touching it (keep same criterion in all the
images)
