I/ INTRODUCTION

Brief introduction about the experiment

Romanowsky stains are such types of stains that are universally employed for the staining of blood cell
with the property is making distinctions in shades of staining, and of granules differentially, depends on
two components: Methylene blue and azure B (trimethylthionine) or eosin Y (tetrabromofluorescein). In
this test, Giemsa stain is used. It differentially stains the cells and appeared colored with shape
respectively.

Objectives of this labwork experiment

✓ Identify different types of leukocytes in the blood.

✓ Determine the relative numbers of the different types of leukocytes by performing a differential
count.

Other techniques used for the same reason.

Romanowsky staining is a prototypical staining technique that was the forerunner of several distinct but
similar methods, including Giemsa, Jenner, Wright, Field, and Leishman stains.

III/ RESULTS AND CONCLUSION

Our group can observe all difference type of white cell that stained by Giemsa stain solution.

In other group, they cannot see Eosinophils.

In our group, the number of Basophils is lowest (1/100)

Both groups examine 100 different white cells and have the same results is that Neutrophils have the
highest number.

Overall, there are a bit of difference in the percentage of number of cells that is given but it is maybe the
total number of cells that our group examined is not enough to ensure anything.

IV/ DISCUSSION

1/ Is it possible to tell T lymphocytes from B lymphocytes using this stain? Explain.

In this test, we cannot observe specific T cells and B cells it is easy to see lymphocytes. It is because it is
too small to observe with normal microscope.

5/ Instructor lectured slowly, so it is easy to follow. The great thing is that lecturer allows us to submit
reports without plastic cover.
