Part I: Production Rates

For a 1,3 Propanediol (C3H8O2) – PDO – production organism (C1H1.8O0.5N0.2) a first


chemostat experiment is performed and measurements are made when the chemostat is at
steady state. PDO is produced aerobically using glucose (C6H12O6) as a substrate and NH4+ as
N-source. The chemostat feed Fin in L/h contains glucose and (NH4)2SO4 dissolved in water.
Air sparging, with rate FN,in in mol/h, is used to supply O2 to the organism and to remove
CO2. The dry sparged air contains much more CO2 than normal and its composition is 21%
O2; 0.5% CO2 and 78.5% N2. The offgas is passed through a cooling system, to make the gas
as dry as possible before analysing with a coupled gas chromatograph-mass spectrometer.
