MATERIALS

Petri Dish (ID=23mm) Cathode Carbon Cloth

Anode

5 grams of Standard Plate Count Agar was dissolved in 200ml disstiled in 250 ml dilution bottles and auto claved
for______. Then added with 5 grams glucose as additional microbe substrate.

Salt Bridge

5 grams of Standard Plate Count Agar was dissolved in 200ml disstiled in 250 ml dilution bottles and auto claved
for______. Then added with 20 grams sodium chloride.

Assembly and inoculation

1g of wastewater and 10g of agar was poured into the petri dish after drying carbon cloth was placed on top and was
inoculated directly with 1g of wastewater and another 10g of agar. After drying, 5g of salt bridge was poured directly on
top until brim full. The cathode was place on top of petri dish.
REMARKS

 Standard Plate Count Agar was used due to availability


 For preliminary experiments, assembly and inoculation was performed under non-sterile conditions
 Carbon Cloth was not cut perfectly
 Electrical connection of the cathode was checked using ammeter and multimeter.
 Wastewater was checked for microbial count using petrifilm aerobic plate count to verify microbial colonies.
 After 24 hours, it was observed that there was a formation of gas in the anode pushing the electrodes out of the
set-up. According to our microbiologist this can be caused by the coliform present in the wastewater sample.
