1 INTRODUCTION
This technical bulletin applies to MIURA systems, both Single and Double ARM.
As announced with our previous presentation “Miura Single Sampler ARM and Double Sampler ARM
– New Software Version 2.2.0.10” issued at the launch of the last and newest Miura software version
by our Marketing office, it gives you evidence that next Miura systems will be released in the new
configuration that includes the new software.

2 INTRODUCTION
These new configurations will be on the systems from the following serial numbers:
 Double sampler ARM: from s/n A1294.
 Single sampler ARM: from s/n A1278.

3 GENERAL INFORMATION ON THE NEW SYSTEMS


The new MIURAs are now available basically in two models:
 SINGLE Sampler ARM (M1) – previous version commercially named Miura 200
 DOUBLE Sampler ARM (M2) – previous version commercially named Miura (300 or 500),
which are substantially differing in throughput up to 300 mono-reagent test/hr plus ISE (electrolytes).
All of the systems are however available for bench-top or for floor-standing installation.

The monitor of the PC is now on-board, fixed on an appropriate jointed arm integrated with
the instrument cabinet.
Both single and double sampler arm models can be supplied in two main sample tray
configurations:

 59-sample positions – no more dilution cups used.


 79-sample positions – no more dilution cups used.
The basic versions of the system are:
 M1-59, single-sampler ARM, 59-sample positions,
 M1-79, single-sampler ARM, 79-sample positions,
 M2-59, double-sampler ARM, 59-sample positions,
 M2-79, double-sampler ARM, 79-sample positions.
ISE Module can be included in any of the versions above.

The new sample tray configurations together with the new software features give the system the
main following advantages:
 Small “dilution cups” no longer in use,
 Dynamic sample positioning (as in Miura One system),
 Sample dilution process improved,
 Automatic sample pre-dilution by method,
 Automatic sample repetition with post-dilution,
 More sample positions,
 Fully compatible with MS Windows 7 ™,

 Possibility of upgrading older 50-sample position systems on the field and complete software
compatibility.

New MIURA systems, single and double sampler Arm, adopt a new cuvette Washing Station.
The purpose of this modification is to allow the system to use of a new Cleaner Solution cheaper
and easier to ship. This modification gives the operator the following main advantages:
 Larger aspiration channels.
 Higher aspiration power per channel.
 All needles have self-recovering position (given by springs).
 First aspiration channel has separate waste path.
 Possibility to use the New Cleaner Solution which is more effective.
 Foam formation limiting.

New User Manual and new Quick Start Guide have been issued and the following link is for
downloading the PDF versions; they are valid for any instrument versions:

Note: all new features and performances have been detailed into the new User Manual. Refer to
that publication for detailed descriptions on how to use.

4 MONITOR OF PC IS ON BOARD
The monitor of the PC has been integrated on-board to save space on the
working bench.
It has been fixed onto an appropriate support integrated with the instrument
cabinet, so it doesn’t need to be located on the lab bench anymore.
The cabinet is provided with a hub where to fix the monitor support (a jointed
arm).
The monitor must be fixed on the support through the standard VESA fixings.
The joints of the support can be adjusted as preferred by the user.
This support gives full adjustment margin.

Note: the new system packing list will include this new jointed arm – for mounting instructions refer
to the last Miura User Manual.

5 NEW SAMPLE TRAY CONFIGURATIONS


The system Miura 59-sample positions, single and double ARM, can only use the following supports
indifferently for the samples / standard and calibrators / QCs:
 Sample tubes diameter from 12mm to 16mm and height from 75mm to 100mm,
 3mL sample cups with special adapter

The system Miura 79-sample positions, single and double ARM, can only use the following supports
indifferently for the samples / standard and calibrators / QCs:
 Sample tubes diameter from 12mm to 13mm and height from 75mm to 100mm,
 3mL sample cups without any special adapter.

6 SMALL DILUTIONS CUPS NO MORE USED


Any Standard or Calibrator, any Quality Control or Samples will be introduced in the system only
into standard “sample tubes”, ranging from 12mm to 13mm output diameter, or into 3mL
sample cups. This means that:
a) Dilution is performed directly in the reading cuvettes: being eliminated dilution
cups the operator needs one consumable less and the dilution is directly
performed into the incubation cuvette. The cost of dilutions gets lower as reading
cuvettes will be automatically washed and reused by the system. No more
dilution cups wasted.
b) Any dilution is prepared just before its use and not upon the start of the routine
together with all of the others (old sw). This prevents eventual diluted sample
evaporation and alteration.
c) Standard/Calibrators, QCs and Samples will be processed in the same support type.
d) Higher flexibility of the system in sample positioning on the tray. Software dynamic sample
positions assignment. No more pre-assigned and limited positions.

Note: the new system packing list will NOT include old dilution cups.

7 DYNAMIC SAMPLE POSITIONING AND MORE POSITIONS


The new software gives the system much higher flexibility than the older one in terms of positioning
of samples, of standard and calibrators, of QCs on the tray.
The software manages dynamically sample positions and automatically assigns the first free
sample positions to any item to process. This means that the operator can choose any of the
positions indifferently for samples, calibrators and quality controls without any limitation.
Moreover, also the sample cups can be located anywhere on the tray.
No longer dilution positions are needed on the tray – as said before the dilution is performed into
the incubation and reading cuvette just before the analysis.
The new systems with the new software give the possibility to have more sample positions.
New systems can now be supplied with 59-sample position or 79-sample positions tray that are quite
higher than the 50 positions provided in the previous ones.

8 SAMPLE DILUTION PROCESS IMPROVED


The sample dilution process has been improved both for standard and calibrators when creating
“calibration curves” or when sample dilution is required.

Part of the process related to the generation of a dilution starting from a concentrated has been
changed:
 The sample aspiration timing, when aspirating from concentrated sample, has been
optimized. By avoid quicker depressions a higher precision in bigger volume aspiration of
thicker fluids has been reached: substantially it has been given longer time for sample
aspiration.
 Higher diluted volume percentage mixed twice during preparation of the dilution in
cuvette.
Possibility of probe’s outer surface cleaning (in washing sink – w/systemic solution) after aspiration of
the diluted sample from the cuvette for analysis (this feature requires new washing sink tips).

9 AUTOMATIC SAMPLE PRE- AND POST-DILUTIONS


With the new software both sample pre-dilutions and sample post-dilutions have been automated.
Sample pre-dilutions are now selectable for each application in each method parameters menu
and they are valid for all samples and can be different for each sample type.
They are automatically performed in the incubation and reading cuvette by the system.
Sample post-dilutions can be now automatically run by the system or on operator’s request. This
means: repetitions of the sample in auto-post-dilutions are now selectable for each application in
each method parameters menu and can be different for each sample type. This applies in case of
specific events that can be selected or disabled (i.e.: result out of normal range, result out of
linearity, result out of substrate depletion, …).
To set pre- and post-dilutions enter the menu Chemistry>Methods>Dilution by samples.
Repetitions of the sample at “desired” dilution ratio “on operator request” are always possible as in
the previous sw version.

10 MS WINDOWS 7™ COMPATIBILITY
The new software requires a new installer. The new installer has been designed to be fully
compatible with MS Windows 7™ and it has also been optimized in terms of memory used and
operating timing.

11 THE NEW WASHING STATION


With respect to the previous instrument model, changes have been made at different levels. Below
the main list.

1. Washing station head – aspiration needles:


 Aspiration needles sizes have been increased if compared to the previous ones and springs
have been added on all positions to help self-alignment on the cuvette bottom.
Consequently the fixing system for needles have been changed and much improved: now
a metallic screw cap holds the spring and the needles assembly.
 Aspiration needle tubing internal diameters have also been increased.
In this way the possibility to get aspiration needle block has been reduced significantly and the
flow have been increased.

2. Washing station head – 1st aspiration channel:


 The first aspiration needle and channel has been separated by the others; it is connected to
a single peristaltic pump (120ml/min flow) entirely dedicated to the reaction liquid
aspiration when the cuvette, that has completed analysis, is approaching the washing
station. The waste of the first aspiration needle is directly channeled to a dedicated waste
tubing into the same external waste tank through an additional output connector (the
waste tank wiring is new and includes two tubes and two connectors). The command of this

pump is the same controlling the aspiration needle pump and it has been derived through
a wiring jumper.
This choice allows the possibility to treat foamy reagents that, containing detergents, could lead to
the formation of foam that limits the performance.
Note: the new system packing list will include the new Waste Tank wiring.

3. Washing station manifold:


 The first aspiration channel has been separated by the others as described above.
 The single aspiration channel, that previously was grouping all aspiration needles, has now
been doubled for the remaining five aspiration channels (from the 2nd position to the 6th):
those have been combined into two groups, the first one includes aspiration channel #2
and #3 while the second one includes aspiration channel #4, #5 and #6. Each of these two
channels goes directly to one diaphragm pump head that is then discharging to the waste
tank.
Again, with this new configuration the risk to generate foam while aspirating has been
significantly limited.

4. Diaphragm pump for aspiration needles:


 The single-head diaphragm pump has been doubled to allow aspiration by both aspiration
channels. A double head pump have been used and the command of this pump is the
same controlling the 1st aspiration channel peristaltic pump; it has been derived through a
wiring jumper.
 The discharge tubing, coming from both head outlet fittings, are connected together and
channeled to the waste tank through the main waste connector.

11.1 The New Washing Station in Miura single and double sampler arm
The new Washing Station in Miura systems has the following configuration:
 5 couples of needles, each composed by one shorter and smaller needle for dispensing
and by one longer and larger one for aspiration (ASPx/DISx);
 1 aspirating needle, large size (ASP6);
 1 drying tip (white Teflon).
The architecture is shown in the following pictures:

The washing station pumps configuration includes:


 5 peristaltic pumps, for dispensing the washing solution (all peristaltic pumps are placed
behind the front panel of the instrument);
 1 peristaltic pump for aspiration of the reaction in position #1 (placed behind the front
panel of the instrument);
 2 diaphragm pumps, one single-head for the drying tip and one double-head for the
aspiration needles (these pumps are placed behind the back panel of the instrument).

Each of the 5 dispensing peristaltic pumps is directly connected to only one dispensing needle
(from position 1 to position 5 - short ones) of the washing station.
One additional peristaltic pump is on the aspiration needle in position 1; it aspirates reaction fluids
limiting the foam generation (and then the impedance) along the waste path to enhance the
flow.
The double-heads (single motor) diaphragm pump is connected in parallel to 5 of the 6 aspirating
needles (long and smaller ones): one head aspirates in parallel from needle 2 and 3, the other
aspirates from needles 4, 5 and 6. The single-head diaphragm pump is dedicated to the drying tip
only.
During the washing cycle all cuvettes progressively run under each of the needles from position 1
up to position 8, moving one position on at any machine cycle. During the washing cycle,
composed from a descent phase, a waiting phase and a rising phase, the needles, placed in the
different positions, behave as follow:
 Positions 1 (couple of needles): the cuvette is fully discharged of the reaction fluid
and then filled with Systemic Solution; solution level
left is about 450ul;
 Positions 2 (couple of needles): the cuvette is fully discharged and then filled with
Cleaner Solution; solution level left is about 400ul;
 Positions 3÷4 (couple of needles): the cuvette is fully discharged and then filled with
Systemic Solution; solution level left is about 450ul;
 Position 5 (couple of needles): the cuvette is fully discharged and then filled with
Systemic Solution; solution level left is about 450ul. It
is optically measured and checked (transparent or
zero-value updating);
 Position 6 (single needle): the cuvette is fully discharged;
 Position 7 (no needle): no actions;
 Position 8 (tip): the cuvette is dried from residual liquid.

12 THE NEW CLEANER SOLUTION


A new Cleaner Solution has been released to be used with the system including the new Washing
Station.
This new solution is cheaper and easier to ship by maintaining the same effectiveness of the
previous Cleaner Solution. The new Cleaner Solution can be packed and send as concentrate
double 100mL vials kit that is much more compact than the old one packed in 5L containers.
It helps to keep cuvettes clean being really effective against plastic aging and by limiting at the
same time any invasive effect.
The new Cleaner solution can be purchased in kit including 2x100mL vials.
It must be prepared by mixing one 100mL vial of solution into 5L of bi-distilled water (conductivity
below 1.6µS/cm) directly into the 5L blue tank of the system.
The new Cleaner Solution purchasing code is: P3140000282 (Cleaner Solution for Miura 2x100 mL).

Note: The new system packing list will include this new Cleaner Solution.

The previous Cleaner solution can be anyway used without any performance degradation in
systems with the new Washing Station.

NOTE: the new Cleaner Solution can be used also into older systems only after special upgrade of
the instrument. This special retrofit upgrading the system will be described in a forthcoming
dedicated technical bulletin that will be sent soon after this one.
