ABSTRACT
Dermatophyte skin mycoses are the most common superficial infections in humans and animals. They
have become a problem of great interest in public health, as it not only generates high cost in
treatments, recurrent medical visits, but also can become a chronic disease of difficult management
despite having antifungal drugs with very good spectrum of action such as terbinafine. The aim of this
study was to evaluate the in vitro antifungal activity of terbinafine against three filamentous fungi
Trichophyton rubrum, Trichophyton mentagrophytes and Microsporum gypseum, by means of
minimum inhibitory concentration (MIC 50), minimum fungicidal concentration (MIC 90), and percent
inhibition radial mycelium. After the experiments, terbinafine was determined to have a MIC 50
between 0.625 and 1.25μg / ml for Trichophyton mentagrophytes, which coincides with the minimum
fungicidal concentration of 0.625μg / ml. In addition, it was evidenced that there was a radial inhibition
of the mycelium in all concentrations of terbinafine tested in an experiment with Trichophyton rubrum
and decrease of the melanocytic pigment in greater proportion to a concentration of 300, 64.9, 11.1
μg / ml respectively. However, it is important to keep in mind for future experiments that the test
should be performed in triplicate, controlling for confounding variables such as incubation time and
conidia contamination of the same strain in petri dishes. We can conclude that these three methods
are complementary in the study of antifungal susceptibility for dermatophytes.
RESÚMEN
Las micosis cutáneas por dermatofitos son las infecciones superficiales más comunes en humanos y
animales. Se han convertido en un problema de gran interés en salud pública, ya que no solo genera
alto costo en tratamientos, visitas médicas recurrentes, sino que además pueden convertirse en una
enfermedad crónica de difícil manejo a pesar de contar con medicamentos antifúngicos con muy
buen espectro de acción como la terbinafina. En este estudio se pretendió evaluar la actividad
antifúngica in vitro de terbinafina contra tres hongos filamentosos Trichophyton rubrum, Trichophyton
mentagrophytes y Microsporum gypseum, por medio de la concentración mínima inhibitoria (MIC 50),
concentración fungicida mínima (MIC 90), y porcentaje de inhibición radial del micelio. Después de
los experimentos se determinó que la terbinafina presenta una MIC 50 entre 0.625 y 1.25 μg/ml para
Trichophyton mentagrophytes , lo cual coincide con la concentración fungicida mínima que fue de
0.625μg/ml. Además se evidenció que hubo una inhibición radial del micelio en todas las
concentraciones de terbinafina ensayadas en un experimento con Trichophyton rubrum y disminución
del pigmento melanótico en mayor proporción a una concentración de 300, 64.9, 11.1μg/ml
respectivamente. Sin embargo, es importante tener en cuenta para futuros experimentos que la
prueba debe realizarse por triplicado, controlando variables de confusión como el tiempo de
incubación y la contaminación por conidios de la misma cepa en las caja de petri. Podemos concluir
que estos tres métodos son complementarios en el estudio de susceptibilidad antifúngica para
dermatofitos.
