As shown in class, since the Km values for the reactions are the same but the Vmax values are

not, we are facing with a noncompetitive inhibition:

To find KI in this case we use:

𝑉𝑚𝑎𝑥
𝑉𝑚𝑎𝑥𝑎𝑝𝑝 = (1)
[𝐼]
1+
𝐾𝐼
Data (obtained from class):
𝑉𝑚𝑎𝑥𝑎𝑝𝑝 = 25, 𝑉𝑚𝑎𝑥 = 50, [𝐼] = 5 𝑚𝑀
Solving:
[𝐼] ∙ 𝑉𝑚𝑎𝑥𝑎𝑝𝑝 5 ∙ 25
𝐾𝐼 = = = 5 𝑚𝑀
𝑉𝑚𝑎𝑥 − 𝑉𝑚𝑎𝑥𝑎𝑝𝑝 50 − 25

So, 5 mM is the KI for the second inhibitor.
