Q1: This experiment is reverse phase chromatography. What does


that tell you about the polarity of the mobile and stationary phases?

We have a polar (aqueous) mobile phase, so hydrophobic molecules
in the polar mobile phase take up to the stationary phase. Hydrophilic
molecules in the mobile phase however will pass through the column
and these are eluted first.

Q2: Which compound do you expect to elude first from the column
and why? (2 pts)
The benzoic acid (unreacted) because it is the least polar

Q3: What is the definition of an isocratic separation? (2 pts)

Isocratic elution is where the composition of the mobile phase don’t


change during the entire elution process.
