Method & Principle: (Real Time PCR)

In this assay patient’s extracted RNA along with standards & controls was run with master mix including specific primers, Taqman
probes, & enzymes etc. in state of the art real time PCR system, which by defining threshold cycles automatically interprets the
results.

Comments & Suggestion:

1. Please note that the titer of SARS-CoV-2 in body fluctuates in accordance with the virus load, duration and effect of
treatment, sampling time etc. Variable results may occur in different phases of viral infection; therefore clinical correlation is
very important.

2. False negative results may occur due to low concentrations of virus and mutations in target sequence of 2019-nCoV.

3. False positive results may occur due to cross contamination of specimen, infection of other Coronaviruses etc.

4. It is possible that variable results of SARS-CoV-2 PCR may occur during convalescent time (2-4 Weeks) due to
intermittent low-level shedding of viral RNA (Chen et al., 2020; Xing et al., 2020).

5. Lab. to Lab. variation may also occur due to difference in sensitivity, specificity and reproducibility of different assay
techniques used.

6. All the results should be correlated with other clinical & laboratory findings.
