transformant plants, crude mitochondrial extracts. Oxygen uptake was analyzed with a Clark-
type electrode. For each assay, about 200 μg crude mitochondrial extract was resuspended in 2
mL reaction medium containing 25 mM TES TES, 10 mM KCl,
KCl 2 mM MgSO4, 5 mM KH2PO4, 0.3 03
M sucrose, 0.1% BSA (pH 7.5), and applied to the electrode in a sealed glass chamber, under
continuous stirring in the dark. Oxygen uptake was initiated by 10 mM glycine and measured in
the presence or absence of rotenone (50 μM).
