Introduction: Periodontitis is an inflammatory reaction caused by specific microorganisms that cause


damage to the alveolar bone. The bacteria can cause a severe damage, and one of them is known as
Aggregatibacter actinomycetemcomitans. Bone damage can be caused of osteoclast activity.
Osteoclast biomarker in blood serum was named a Crosslink C-Terminal Telopeptide (CTx).Green tea
(Camellia sinensis) contains active ingredient epigallocatechin gallate (EGCG) which was proven to
regenerate bone by increasing bone cell proliferation and differentiation using periodontitis wistar
rats. Methods: This study was divided into 4 groups: The first group was a negative control, the
second was a positive control, the third and the fourth were treatment groups. The negative control
group did not receive any treatment. The positive control group was given LPS Aa 5 μg/3μl PBS and
Camellia sinensisinduction with each dose 150 mg / 100 grBB and 200 mg / 100 grBB on days 21-36
days. The level of CTx was examined by using ELISA method to observe osteoclast activity. The
number of osteoclast were stained by HE method. Results: data analysis showed there was a very
strong correlation between administration of Camellia sinensis extract and an increase in osteocyte
count (p <0.05, with correlation coefficient = 0.883), whereas CTx levels showed that there were no
significantly different (p>0,05) between positive groups with treatment caches one and two.
ConclusionThis study revealed that was a Camellia sinensis could increase the number of osteocytes
in periodontitis inducing LPS Aa on rat wistar.

BACKGROUND

Periodontitis is a periodontal disease characterized


by bone damage. that caused by specific
microorganisms, one of which is Aggregatibacter
actinomycetemcomitans (Aa). LPS which is endotoxin
from Aa has been shown to be involved in the
pathogenesis of alveolar bone resorption. Bone
remodeling requires coordination of three cell types
namely osteocytes,osteoblasts, and osteoclasts.

Green tea (Camellia sinensis) contains active ingredient

Based on these problems, the researchers wanted to
find out more about the effect of green tea extract
(Camellia sinensis) to increase the number of
osteocytes and CTx levels in Rattusnorvegicus male
Wistar strain induced by LPS Aa
Purpose : to know the effect of Green Tea Extract (Camellia sinensis) for C-Terminal Telopeptide
Crosslinks (CTx) and Osteocyte count in rats (Rattus norvegicus) Induced by LIPOPOLICAKARIDA (LPS)
Aggregatibacter actinomycetemcomitans

METODE
Gambar tikus (28 rats which were divided into 4 groups)
Gambar tikus+spuit+bakteri (Injection LPS Aa 5 μg/3μl PBS)
Gambar tikus+spuit+daun teh (Injection Camellia sinensis with each
dose 150 mg / 100 grBB and 200 mg /100 grBB on days 21-36 days.)

DISCUSSION
The lowest number of osteocytes was found in the
negative control group. This is due to LPS Aa being
endotoxin that are able to activate the immune
system to secrete proinflammatory cytokines
which can lead bone destruction. The conclusion
from the pearson correlation test found a very
strong relationship between the administration of
Camellia sinensis extract with the number of
osteocyte cells after LPS Aa induction.
Green tea contains EGCG (67%)
which can stimulate osteocyte
formation

Conclussion
1. there was a very strong correlation between administration of Camellia sinensis extract and an
increase in osteocyte count
2. Ctx level showed that there were no significantly different between positive
groups with treatment group
