ACTIVITY 1 - RQ 2 (TUBES)

 CAPILLIARY TUBE
 PROPER CLEANING
Check niyo itong link guys hehe. Usually kasi ang mga capilliary tubes kay
disposable so wala sya cleaning chuchu. Itong link kay proper cleaning ito ng capilliary tube pero
sa thin layer chromatography sya ginagamit. Hindi lang talaga ako sure if pwede din ito sa blood
capilliary tube.

 STORAGE
After sample collection, microcollection devices should be capped immediately to
prevent exposure to the air, especially if the blood sample will be used for blood gas
analysis.Capped samples should immediately be mixed to prevent clotting.

(After collection storage lang makita ko guys hehe. Usually din kasi room temp lang ang
capilliary tubes kapag walang laman hehehehe)

 WINTROBE TUBE
 PROPER CLEANING
The tubes are cleaned by employing a water-filled 20 cc. syringe fitted with a 5-
inch 13-gauge needle. The needle is inserted to the bottom of the tube and by forcing
water through the syringe, the material contained in the tube is expelled. Any water
remaining in the tube is removed by aspiration. The tubes are then dried in the usual
manner.

 STORAGE
The collected blood should be tested as soon as possible after collection.
Prolonged storage at the room temperature would result in a change in the shape of the RBCs due
to metabolism. After about 6 hours, the chances of hemolysis increase, which would give an
erroneous result. In the macro-hematocrit method, the filing of the Wintrobe hematocrit tube
requires proper care. In the microhematocrit method, the sealing of the capillary tube should be
secure to prevent any leakage. The centrifuge machine should not be opened during the test to
avoid erroneous results. The chances of error in the result will increase if the centrifugation is
interrupted. Immediately after completion of the rotation, the operator should not open the lid
should until after the complete stoppage of the rotation.

 WESTERGREN
 PROPER CLEANING
The Westergren method requires collecting 2 ml of venous blood into a tube
containing 0 .5 ml of sodium citrate. It should be stored no longer than 2 hours at room temperature
or 6 hours at 4 °C. The blood is drawn into a Westergren-Katz tube to the 200 mm mark. The tube
is placed in a rack in a strictly vertical position for 1 hour at room temperature, at which time the
distance from the lowest point of the surface meniscus to the upper limit of the red cell sediment is
measured.

 STORAGE
For use, the tube must be clean and dry. It should be cleaned in an acetone-water
system. Dichromate and the commonly available detergents are not recommended. If the tube is
“disposable” it must be supplied clean and dry and the manufacturer should be expected to
guarantee that the cleaning procedure does not introduce any factor which may affect its use in the
ESR test as compared with glass tubes cleaned by the recommended method.
