PROPOSAL SUMMARY

1. Proposal Summary [Provide a brief one paragraph overview of the proposal, i.e. the idea and the problem it may solve and
brief project plan.]
The Omega 3 fatty acids from chlorella are predominantly Ecosapentanoic acids EPA and meet the fish Omega 3 fatty acids.
Chlorella can be cultivated with simpler cultivation methods and can accumulate very biomass as well as lipids with simpler
techniques of induction. Unlike Spirulina and other species of microalgae, the digestibility of Chlorella biomass is very high and
provides a very rich source of crude protein which can be used as feed.
Aquaculture is the fast growing sector which has doubled its production in the past few decades. Aquaculture feed and supplements
are always a challenge in the sector due to their high cost. Microalgae play a major role in the aquaculture as they are a part of the
food chain. Similarly, the derivatives of the microalgae i.e. lipids and pigments can be used as feed supplements in aquaculture.
Along with the aquaculture, these omega 3 fatty acids can be used as feed supplements for poultry and large animals also.
The current project aims in the production of EPA and DHA from microalgae by using nitrogen rich cost effective medium in closed
race way system and its applicability as the aquaculture, poultry and large animals feed supplement.

2. Briefly state the Objectives and Proposed Approach

The description should cover the following points:
1). Strategy and/or methodology of work.

In the current scenario, polyunsaturated fatty acids viz. omega 3 fatty acids are well needed in the human diet. As per the World
Health Organization, a person should consume approx. 200 -500 mg of omega 3 fatty acids. Similarly, omega 3 fatty acids have wide
applicability in the fields of pharmaceuticals, aquaculture and veterinary also. In Aquaculture and Veterinary fields, omega 3 fatty
acids can be used as feed which shows the huge scope of PUFA. Though conventional method of extracting EPA ad DHA from
marine fishes is available, several disadvantages have been noticed. When compared to the fishes, microalgae are easy to grow and
posses good PUFA content. The current project proposal is designed on the following objectives:
* The microalgae is identified by using morphological and molecular sequencing studies.
1. Cost effective medium optimization studies for high yield production of EPA and DHA.
** Production optimization is carried out by taking one factor at a time.
2. Lab trial by using the novel medium composition.
3. Development novel extraction process for achieving high Omega 3 fatty acids yield
4. Qualitative and Quantitative estimation of EPA and DHA content
*** The omega 3 fatty acids obtained needs to be tested qualitatively and quantitatively by using HPLC, GCMS etc.
5. In-vivo growth enhancement study
6. Designing of cost effective closed race way system which can be a substitute to photo bioreactors.
7. Large scale production of microalgae, extraction of EPA and DHA and QC check
8. Formulation of the EPA and DHA
9. Usage of left over microalgal biomass a fodder for large animals.

The project proposal laid down has several novel aspects covering and trying to rectify all the drawbacks in the production of
microalgae. This project targets mainly on the novel production strategies which can reduce the production cost with high yield
enhancement. Following are the novel strategies intended to be implemented in the project:\r\n1. Thermostable potent native isolate
of microalgae with high capacity of EPA and DHA production.\r\n2. Both macro and micronutrients play a major role in the high yield
production of EPA and DHA. The conventional medium BG11 used is very costly and it needs to replace with a cost effective medium.
In the current scenario, the nutrients of the BG11 medium will be replaced with the nitrogen, phosphorous and potash fertilizers which
is cost effective.\r\n3. Development of closed race way system with the polythene sheets which can be a substitute for the high cost
photo bioreactors and conventional raceways.\r\n4. Novel extraction process to achieve high EPA and DHA content.\r\n5. Utilization

Market summary:
• As a wholesome food ingredient Chlorella sorokina has good demand in Japan and Europe.
• The market for Omega 3 oils is increasing and lower cost of production provides an opportunity to exploit this technology on
a commercial scale.
• As a source of easily digestible protein, vitamins and antioxidants Chlorella can be a replacement to soy and other algal
products. It can be incorporated to meals for undernourished children, women and old aged people.
• On the basis of ingredient, fishmeal represents the largest segment, followed by fish oil, wheat flour, soybean meal and
others. Chlorella can widely replace these ingredients as a feed stock for animal nutrition.
• Based on additives, the market is segregated as vitamin and protein, fatty acid, antibiotics, antioxidants and feed enzyme.

Major opportunity:

Human Functional Foods market:

Chlorella algae is used as a functional food and food ingredient due to its taste profile. The present market for Chlorella as whole
powder is 20000 Tonnes per year and presently the worldwide production is limited to 2000 Tonnes per year. Omega 3 rich chlorella
is expected to have a market of USD 30 Millions in India, Japan and Europe. The market is growing at an AAGR of 12.7 according to
an US report.

In Aquaculture:

Omega 3 fatty acid has huge market potential in the various fields of pharmaceutical, aquaculture and veterinary. In the Indian
Shrimp feed market is projected to reach 1.8 million tonnes by 2024 registering a CAGR of 10 during 2019-2024. Above data shows
huge potential of omega 3 fatty acids i.e. approximately 1,00,000 tonnes.

In Veterinary:
Taking account of 5 states in our country i.e. Tamilnadu, Andhra Pradesh, Maharashtra, West Bengal, Hariyana and Punjab there is
a huge requirement of poultry feed which is around 12 million tonnes for both boilers and layers. Indian animal feed market is
expected to touch INR 898.5 Billion growing at a CAGR of 14.3 during 2019-2024.In this huge market value, the proposed project has
a good capacity to grab the share.

5. Challenges or risk factors associated with the project

The major challenge faced in the proposed project is to develop best cost effective extraction process to achieve high purity product
which is stable and consistent. The cost of technologies such as supercritical CO2 extraction is prohibitive although it provides a very
good quality product. Solvent extraction is a economical alternative but solvent recovery and removal of trace solvents is a
technological challenge. Stability of the product post purification and improving the thermal stability on oxygen sensitivity of the
product are challenges to be overcome. These challenges will be tackled and solutions will be developed by the end of the project.

6. Has any preliminary work been carried out? Give status of work done
If no, please provide the background details.
Native strains of Chlorella species were isolated from Aquaculture tanks, fresh water tanks and river Godavari in Andhra Pradesh and
Telangana states. The strains were cultivated in shakeflasks and screened for their ability to accumulate lipids. The strains having the
ability of producing high biomass above 25 g/L in shake flasks were selected. The PUFA content and Lipid content of the biomass
were analysed and a thermotolerant strain of green unicellular alga Chlorella sorokiana has been isolated from aquaculture water
samples. The isolate was further grown in an optimized medium containing Urea, Ammonium nitrae and Monnoammonium
Phosphates as Nitrogen sources. The role of metal ions like calcium, magnesium and manganese on the biomass production and
lipid accumulation in the cells was investigated. A medium consisting of Triple nitrogen source viz Urea, Ammonium Nitrate and MAP
at 0.1 w/v of culture broth was found to be optimum for highest biomass accumulation 37G/L with a 52 total lipid content on dry weight
basis. A laboratory scale raceway system enclosed with Polypropylene sheet and PVC lining was developed and the efficiency of the
developed raceway was tested. It was found that the lowcost raceway developed could support the growth of chlorella upto 35G/L
biomass. Further work on the enhancement of biomass production in the developed raceway system is being carried out. The low
cost enclosed raceway however is feasible to grow the algae with high cell density and high lipid content. The proposed project
envisages the improvement of process and engineering parameters for a more economical, high yield production of Omega 3 fatty
acids acids through Chlorella sorokiana.

BY Analyzing the potential competitors and possible customers there is huge market requirement for PUFA in market.However we
initially concentrate on aquaculture and animal nutrition. We initially concentrate on feed pellets manufacturing companies who needs
huge quantities of PUFA for manufacturing in their feed formulations.As we already mentioned nearly 1 lakh tons fat is needed in
Indian feed formulations for aquaculture. It is mainly in the form of fish oil. As fish oil has its limiting factors like low content of EPA
and DHA ,odour and limited cultivation space and production cost. Crude fish oil is Rs.120/kg in Indian market. with our technology
we can produce it for Rs.50. So there is huge demand we need to met with.
