AIM: Isolation of DNA

Requirements:
Papaya

Procedure:
1. The isopropyl alcohol is needed later on but it needs to be ice cold, so first 200 ml of the isopropyl
alcohol is measured out and kept in the freezer after covering the top of the container.

2. A soapy salt solution was prepared.


3. The soapy solution was prepared by mixing 180 ml of water, 20 ml of the vim gel , and 1 tsp of the non-
iodized salt. This solution is called as the extraction solution that will be used to extract the DNA from the
cells.
4. In the next step the papaya is mashed up by blending it in a blender with warm water.

5. Next the papaya solution is mixed with the Extraction solution (soapy solution) that we prepared earlier
for the extraction of the DNA from the cell.

6. After few minutes of squishing the papaya, we filtered out the solution to get rid of the bigger particles
using a strainer.

7. Then the filtered out solution was filtered again using a tissues paper to get a more finer solution.
8. Once everything is filtered out, in the next step we isolated the DNA by pouring the 200 ml chilled
isopropyl alcohol in the filtered solution and let it settle for a few seconds and very quickly white stuff
appeared to be floating on the top. This was identified as the DNA.
9. The entire DNA content was then filtered out using a tissue paper and washed with isopropyl alcohol
several times and left it to dry for the next day.

10. The next day, the filtered out DNA turned hard and completely brown in colour.
