ABSTRACT
This monograph for Lactose Monohydrate provides, in addition to common physical constants, a general description
including typical appearance, applications, change in state (approximate), and aqueous solubility. The monograph also
details the following specifications and corresponding tests for verifying that a substance meets ACS Reagent Grade
specifications including: Water, Insoluble Matter, Residue after Ignition, Dextrose, Sucrose, Heavy Metals, and Iron.

TESTS
Water
[Part 2: Titrimetric Methods; Water by the Karl Fischer Method; Volumetric Procedure; Volumetric Procedure for Samples
Using Karl Fischer Reagent, Method 1]. Use 2.0 g of the sample.

Insoluble Matter
[Part 2: Gravimetric Methods; Insoluble Matter]. Use 20 g dissolved in 150 mL of water.

Residue after Ignition

[Part 2: Gravimetric Methods; Residue after Ignition]. In a tared, preconditioned crucible, ignite 5.0 g. Moisten the char
with 2 mL of sulfuric acid.

Dextrose
To a 10 g, finely powdered sample, add 50 mL of 70% ethyl alcohol. Shake frequently for 30 min, and filter through a small,
dry filter paper. Evaporate 10 mL of the filtrate to dryness on a hot plate (~100 °C), and reserve the remainder of the filtrate
for the test for sucrose. Dissolve the residue in 5 mL of water, filter if necessary, and transfer the filtrate to a test tube. Add
5 mL of cupric acetate solution (described below), mix, immerse the test tube in a boiling water bath for 3 min, and allow
the solution to stand at room temperature for 25 min. No red precipitate should appear in the test tube.

For the Determination of Dextrose

C u p r i c A c e t a t e S o l u t i o n . Dissolve 13.3 g of cupric acetate monohydrate in a mixture of 195 mL of water and 5 mL of

glacial acetic acid.

Sucrose
Evaporate 25 mL of the filtrate reserved from the test for dextrose to dryness on a hot plate (~100 °C), and dissolve the
residue in 9 mL of water. Add 1 mL of dilute hydrochloric acid (20:8) and 100 mg of resorcinol, transfer the mixture to a
small test tube, and immerse the tube in a boiling water bath for 8 min. The solution should remain colorless or acquire not
more than a slight yellow coloration.

Heavy Metals
[Part 2: Colorimetry and Turbidimetry; Heavy Metals (as Lead); Procedure for Heavy Metals, Method 1]. Dissolve 6.0 g in
about 20 mL of water, and dilute with water to 30 mL. Use 25 mL to prepare the sample solution, and use the remaining 5.0
mL to prepare the control solution.
