1. Give the results and interpretation from the biochemical tests of Escherichia coli.

 Urease Test

 The urease broth looks exactly the same as its original form. There is no color change
which indicates that this is a negative reaction. So this means that the organism
Escherichia coli does not produce urease.

 MIO

 The tube became cloudy and the stab line cannot be seen and this means that the
organism Escherichia coli is motile. It also produced gas because there are bubbles in
the tube. Ornithine was a decarboxylase because there is a bluish gray color in the tube.
The cloudy purplish gray indicates a positive result which is the dark decarboxylation of
the amino acid ornithine. When the Kovacs reagent was added, there is a pink color
forming on top which means that the organism E. coli does deaminate the amino acid
tryptophan which would be produced in the agar itself.

 Citrate Test

 The culture media remained green and that indicates a negative result. A negative
citrate result indicates that the organism E. coli does not utilize citrate as the sole source
of nitrogen and carbon in order for it to grow on the media. So, there is no alkaline
reaction and it keeps the agar neutral which keeps it green.

 Triple Iron Sugar

 The organism E. coli result was acid over acid because it produced a yellow color which
indicates an acidic environment and that there was a fermentation. It also produced gas
which can be seen from the bubbles that formed in the tube but there is no blackening
of the media which means there is no H2S production.
