Exercise 1- DNA extraction

1. For which reasons the isolated DNA can be used?
- Genetic testing
- Body identification
- Analysis of forensic envidence
2. Name the 4 main steps in DNA extraction.
- Collect cheek cells
- Burst cells open to release DNA
- Separate DNA from proteins and debris
- Isolate concentrated DNA
3. How to collect the check cells?
- Drag the swab over to the inside of the test subject’s mouth to collect a sample.
- place the swab into an Eppendorf tube, look at closely at the swab that it is coverd with
hundreds of tiny cheek cells.
4. What are components of lysis solution?
detergent and an enzyme called proteinase K.
5. What is purpose of using concentrated salt in the 3rd step?
The salt causes proteins and other cellular debris to dump together
6. Name one equipment used in DNA extraction.
- Micro- Centrifuge
Exercise 2 - DNA detective: use DNA evidence (DNA profiling) to solve a crime
BIO1061 - Basic Biology & Labs

1. List three samples you would like to take and bring to lab for DNA analysis? Explain
the reasons you choose these samples?
Three samples I would like to take and bring to lab for DNA analysis:
- Blood cells on the glass
- skin cells on the handle
- Cheek cells from saliva
The reason for my answer is that we all leave DNA behind wherever we go. DNA is a
molecule found in the nucleus of nearly every cell in your body. Therefore, three sample
I given containing cells will contain DNA.
2. Who is crime? Why?
As my observation, the crime is suspect 3 due to DNA of suspect 3 similar to sample
from crime scene.
