What is s-adenosylmethionine (SAMe)?
 S-adenosylmethionine is the manmade form of chemical that occurs naturally in human

body.

 SAMe has been used in alternative medicine as a likely effective aid in reducing the

symptoms of depression, and in treating osteoarthritis.

 SAMe is effective in treating liver disease during pregnancy, improving symptoms in

people with fibromyalgia, and in treating nerve problems in people with AIDS, or

sexual dysfunction in men with depression.

Dose of SAMe:-
 For Depression:- Up to 1600 mg of SAMe daily in 2 divided doses for up to 8 weeks has
been used alone or together with antidepressant medications.

 For osteoarthritis: 600-1200 mg of SAMe daily in up to three divided doses for up to 84


days has been used.

 For fibromyalgia: 800 mg per of SAMe daily in two divided doses for 6 weeks has been
used.

 For conditions in which bile flow from the liver is slow or blocked (Intrahepatic
cholestasis): 500 mg of SAMe taken twice daily until delivery or 1600 mg taken daily in
two divided doses for 2 weeks has been used.

 For sexual dysfunction: 400 mg of SAMe taken twice daily for 2 weeks, then increased to
800 mg twice daily for another 6 weeks, has been used.

Mechanism of Action of SAMe:-
 S-adenosylmethionine is an intermediate metabolite of methionine. Its

involvement in methylation assists in cellular growth and repair, maintains

the phospho-bilipid layer in cell membranes. It also helps in the maintenance

of the action of several hormones and neurotransmitters that affect mood.

 S-Adenosylmethionine (SAMe) is a natural substance present in the cells of the

body. It is a direct metabolite of the essential amino acid L-methionine.

SAMe plays a crucial biochemical role in the body by donating a one-carbon

methyl group in a process called trans methylation.

 SAMe, formed from the reaction of L-methionine and adenosine

triphosphate catalyzed by the enzyme S-adenosylmethionine synthetase. It

is the methyl-group donor in the biosynthesis of both DNA and RNA

nucleic acids, phospholipids, proteins, epinephrine, melatonin, creatine

and other molecules which are further used in body development.

 Absorption:- SAMe is absorbed from the small intestine when taken through

oral route. It is advised to take SAMe on empty stomach.

Advantages:-
 Depression:- According to a study, it was found that The SAM-e clinical data

also are strongly suggestive of antidepressant efficacy. It was found that

SAMe is effectively useful in treatment of depression [1].

 Liver injury:- In a clinical study, it was found that SAMe is effectively

beneficial in the treatment of liver disease by expressing over two genes

named as MAT1A and MAT2A. Those patients who were using SAMe were

found to have improvement in their chronic liver diseases [2].

 Tobacco Dependence:- In a study, around 120 subjects were involved and it was

found that SAMe did not increase smoking abstinence rates. Abstinence and

tobacco withdrawal data from this clinical trial suggest that SAMe holds

promise for the treatment of tobacco dependence [3].

 Osteoarthritis of knee and hip:- A four phase clinical trial was conducted in

which 656 patients were involved and it was found that SAMe on both pain and

function is potentially clinically relevant and, although effects are expected [4].
