This document provides specifications for the herb Alfa Alfa, including its macroscopical and microscopical identification, chemical tests by thin layer chromatography, and requirements for loss on drying. Key details identified include that the leaf is trifoliate with serrate margins, the flower is violet, and five spots appear at specific Rf values when a chloroform extract is tested by TLC. The specifications are to ensure quality and accurate identification of the herb.

Tests Acceptance Criteria Reference
Macroscopical Leaf trifoliate, leaflet oblanceolate to narrowly HPI
Identification obovate, 1.5 to 3 cm long; margin serrate, apex acute. Peduncle erect, about equaling the subtending leaves, bearing a subglobose to short- cylindrical head, 1 to 30 cm long. Flower violet, in axillary raceme of 7 to 10 flowers; 3 to 4 cm long; calyx five-toothed; corolla papilionaceous; stamens 1, nine united and one free. Microscopical HPI Identification
Leaflet Epidermal cells with wavy anticlinal walls, upper HPI
epidermal cells 14-25-50-70µ, slightly smaller than the lower epidermal cells which are 20-35-60-100µ.Stomata on both surfaces, oval, 20-30-40µ long and 10-20-25µ wide, either anisocytic or anomocytic. Trichomes Rare on the upper surface, numerous over the mid-rib, HPI on the undersurface, consisting of 2 short basal cells, 10-20µ long and a linear terminal cell 350-1120µ long and 20µ wide, Clavate glandular trichomes 75-100µ long, occur mostly on the undersurface and the calyx, usually consisting of a stalk cell, an intermediate and a terminal cell, 25 wide with a rounded apex and often divided into 2 to 4 cells Lamina Thin, transverse section showing 1-2 layered palisade HPI and spongy parenchyma; the vascular bundles have fibers above and below and the cells of the parenchyma abutting upon the fibers contain prism crystals of calcium oxalate oxalate 8-15 by 4-8 wide. Corolla Epidermis papillose having striated cuticule. Pollen HPI grains spherical, 30-35 in diameter with 3 pores and 3 germinal furrows, exine smooth and contents granular.

Flower Almost square, epidermal cells elongated in axial HPI
direction with two types of epidermal hairs, glandular, 2 or more celled and unicellular trichomes, cortex of rectangular r oval cells containing starch grains and crystals of calcium oxalate. Young Stem Endodermis usually without casparian bands. Outside HPI the endodermis is a zone of chlorenchyma, limited peripherally by a single layer collenchymas except at the angles of stem. Vascular bundles collateral, conjoint and open. Continuous cambial ring is formed which produces continuous zone of lignified xylem. Parenchymatous cells of pih disintegrate to form a hollow. Mature Stem The fluorescence turns greenish on adding a few drops HPI of 5% sodium hydroxide solution. Identification HPI (B y TLC) Carry out Co-TLC of Five spots appear at Rf 0.12, 0.20, 0.30, 0.40, and 0.54 chloroform extract of Mother respectively. Tincture on silica gel 'G' pre- coated plate using n-butanol: acetic acid: water (4: 1:1v/v) as sorvent system. After spraying the plate with ninhydrin reagent and heating the plate for 10 minutes at 110°.
