S1 Figure: Steady state population distributions for the four types of tristabilities in the DNFL-B
network are plotted together with the contours of the potential energy and 1-p bifurcation diagrams.
The distributions were obtained from stochastic simulations of the model using Gillespie’s stochastic
simulation algorithm. At each S an ensemble of 10000 simulations were run with random initial values
between 0 and 1000. The intensity of the color indicates the density of the population. Low and high
density are represented by yellow and green, respectively.

of each parameter (sample space of 10000) for the four networks. We performed local parameter
sensitivity analysis of all four 2-C motifs to understand the role of individual parameters in tristability.
We began with a parameter set that resulted in tristability, then we counted the number of times the
model generated tristable bifurcation diagrams when a particular parameter was varied keeping other
parameters fixed.
