Procedure
1. Add 10 grams of ground coffee
2. Pour 4 grams of sodium carbonate on top of of coffee
a) The sodium carbonate will react with a lot of different molecules in the
coffee and make them more water-soluble.
3. Pour the coffee and sodium carbonate in 60 ml of distilled water
4. Place watch glass on the top and bring to a boil
5. Boil for 15 mins.
a) Solution may foam a lot. To prevent foaming you may pick it up and shake it
a little
b) Foaming occurs when you are providing too much heat to the solution
6. Remove from boiling and allow it to cool
7. Remove the coffee grinds from the solution with the use of coffee filter
8. Place a watch glass on top of the filtration to avoid losing heat
9. After filtering, add a little bit of of boiling water to wash the original flask, allow it
to be filtered then further add a 100ml of boiling distilled water and once again place
the watch glass on the top while filtering.
10. Transfer the coffee to the Separatory Funnel.
11. Add 15ml of Dichloromethane to the coffee
12. Shake the separatory funnel gently in order to avoid forming emulsion
13. After seeing layers form, separate the bottom layer by opening the stopcock
14. The bottom layer contains the caffeine
15. Repeat the process with 15ml of Dichloromethane, then repeat again with 10ml of
Dichloromethane
16. After finishing repeating the process. Put the organic solvent extracts back into a
clean separatory funnel
17. If you notice that the extract is a little bit cloudy, you may pour a saturated salt
solution in order to pull the water from the organic solvent.
18. Collect the clear bottom layer by opening the stopcock
19. Add some drying agent or molecular sieves
20. Dry for 20 mins then decanted it to a clean beaker.
21. Wash off the leftover sieves with dichloromethane
22. Boil the organic solvent to remove the Dichloromethane
23. Be care of overheating the beaker and wait for a little clump of crude caffeine to
take form
24. Recrystallize the small amount of caffeine using a 95% ethanol in a test tube
25. Place the test tube in a freezer and the contents will be vacuum filtered
26. The yield would be a 30mg of Caffeine
