ACTIVITY NAME: Preparing Permanent Slides


OBJECTIVES:
You have two objectives to fulfill by the end of this lab:
1. Focus specimens using the compound microscope.
2. Create permanent slides.

MATERIALS:
1. Microscope 8. colorless nail polish
2. Laboratory gown 9. facial tissue or cloth
3. bottle for the specimens 10. alcohol (70 percent, isopropyl)
4. glass slide, cover slips 11. toothpick
5. forceps or tweezers 12. slide box or Microscope Plastic Slide
6. specimens Mailer
7. rag (round)

Specimen:
1. Pollen (From any flower)
2. Wing/s of a fly
3. Human hair strand (white)
4. Letter e (printed) – font size “5”, font style “arial”

REMINDERS:
1. Wear your laboratory gown during the activity. For girls, don’t forget to tie your hair.
2. Clean your work stations after the said activity.
3. Manage your wastes properly as discussed by your teacher. (ex. Broken slides etc.)
4. Return all the microscopes to your teacher or to the laboratory where you got it from.

PROCEDURE:
1. Prepare your specimen.
2. Make sure that your microscopes (specially the lenses – eyepiece and objectives), glass slides and cover slips are
free from dust. Use the smooth cloth or facial tissue to clean these things.
3. Place the slide on a flat surface.
4. Use tweezers or a forceps to place the sample/specimen on the slide.

5. Use the colorless nail polish to seal the specimen with cover slip. Gently place the cover slip on top of the
specimen with colorless nail polish. (See image, the red color is the colorless nail polish.) That’s how you will be
placing it in the slide. Next is to place a cover slip. Gently place the cover slip which will cover the entire center
in which the specimen and the polish are placed.

6. Let it dry for few minutes or an hour. Check the slide for bubbles using the microscope.
7. Use the scanner and the low power objective to view the specimen.

SUBMISSION DATE: December 2 (LUNCH TIME) (4 slides only) Make sure to place it in a slide box or in a slide mailer. You
can use one slide box for the entire section. Just don’t forget to label all your slides like this.

Sample final slide: Of course, with cover slip. Sample label:


Indicate the name of the

who created the slide.
Ex. (Jobelle Suñga will
become J.Suñga.)

Also don’t forget to


place your section.
(ex. 8A for 8 Andromeda)
