Bacterial culturing on agar plates

Introduction:

Bacteria are an essential part of the ecosystem. They are crucial to our health and the
environment, have an important role in food production and provide bioengineers with tools to
harness their properties and manufacture compounds.

Material and method:

1.Swab
2. Culture
3.alcohol
4.Bensen burners
5.Loop
6.The bacterial sample(throat sample)
7. Blood agar
8.Lab coat
9.Pen(for labeling)
10.Incubator
11.Test tube that contained the sample
12. Petridish

Procedure:

1. First of all we have to sterilize all the method or material that we have
2. We use the inoculation loop to take our sample by burning loop with a bunsen burners , the
loop should be cooled before putting it into test tube we take bacterial colonies and placed into
the test tube after that vigorously shaking the tube to collect all the bacteria and you can see
the turbidity
3. then we take a sample from test tube to growth medium
4. after we finish adding the sample,we put the sample in the for 24h at 37c degree.
Result:

Size:small and medium


Shape:irregular
Color:gray and yellow
Texture:dry and viscid colony
Margin:filamentous,wavy(undulate),entire(smooth)
Height:flat and convex
Beta hemolysis cause disease

Discussion:

My sample differs from the other sample in the color and shape of the colony because of the
different sample or maybe making a mistake during the work and doing less or more
streaking.that’s why these differences in the properties between the sample occurred.

Conclusion:

From this experiment I learned how to grow bacteria in the appropriate condition by using
growth medium and incubator also other material and methods. For diagnosing type and size of
the bacteria that cause the disease,also how to know prevent or treating this disease that can
cause with this type of bacteria.
