CULTURE METHODS
Culture Media:
A liquid or gelatinous substance containing
nutrients in which microorganisms, cells, or
tissues are cultivated for scientific purposes.
History
The original media used by Louis Pasteur – urine or
meat broth
Solid media were first introduced by Robert Koch
incorporating gelatin.
Colony – A microbial colony is defined as a visible
cluster of microorganisms growing on the surface of a
solid medium, presumably cultured from a single cell
Agar
Used for preparing solid medium
Obtained from seaweeds (Red algae)
No nutritive value
Not affected by the growth of the bacteria.
Melts at 95oC & sets/cools at 42oC
2% agar is employed in solid medium
Nutrient broth: Meat extract broth
This is a combination of peptone and meat
extract. Peptone is a mixture of Polypeptides and
Amino acid. It serves as the source of carbon,
nitrogen and energy. Peptone prepared by
digesting meat with trypsin and other proteolytic
enzymes.
Reduction of oxygen
By using reducing agents – 1% glucose, 0.1% Thioglycolate
