About the FLEXMAP 3D
The FLEXMAP 3D is an enhancement of our current flow cytometric-based platform, the Luminex
100 and 200. The new system can acquire samples twice as fast as the Luminex 200 for High
Throughput Screening applications and can read a combination of 500 multiplexed microspheres. In
combination with your analytes of interest this translates into a 500 plex assay.
The FLEXMAP 3D also incorporates numerous usability enhancements:

The FLEXMAP 3D system combines differentially dyed fluorescent microsphere sets with an innovative
instrument design to take bead-based multiplexing technology to the next dimension. Dual sampling
channels, increased injection rate and 384-well plate compatibility deliver faster read times, while other
features such as automated probe adjustment, straightforward routine maintenance operations and an
intuitive automation and LIS-compatible software interface simplifies operation.

The combination of these features make FLEXMAP 3D the platform of choice for flexible, simple
high-throughput multiplex testing.

384 Well Plate Compatibility


The FLEXMAP 3D is compatible with 96 and 384-well plate formats and offers sample plate heating
control for thermally sensitive applications. These features combined with the foil piercing sampling
probe provide reliable temperature control over a range of 35C to 60C.

Automation/LIS Compatibility
The new xPONENT 4.0 software controls the FLEXMAP 3D system and offers interfacing options for
laboratory information systems (LIS) as well as automation. An optional swivel base for the system
further simplifies integration with third-party liquid handlers.

Simplified Maintenance
The FLEXMAP 3D system has multiple features which simplify start-up, shutdown and maintenance
operations and greatly enhance usability for HTS applications.
• Automated probe height adjustment
• Automatic calibration for MicroPlex™ and MagPlex™ microspheres, and enhanced PMT settings
• Calibration and verification kits with color coded reagents and CD with importable reagent target values
• System initialization routine combines system warmup, washes, calibration and verification into
one simple daily startup routine

How did Luminex build a 500 plex?
The FLEXMAP 3D system works in the same basic manner as the LX200 analyzer. Mixtures of
internally dyed xMAP microspheres are used in an assay, each having been prepared to bind to
a specific analyte of interest. After the assay has been performed, the mixture of microspheres is
aspirated into the system and each microsphere is individually excited by a red and green laser.
The xMAP microspheres, impregnated with three dyes,excite at the same red laser wavelength and
fluoresce at three seperate wavelengths. Monitoring the relative intensity of the three signals allows
the system to discriminate up to 500 different microsphere sets. The green laser is then used to
excite any surface associated reporter molecules, normally in the form of phycoerythrin, and the
resulting fluorescence is measured to determine the amount of analyte present.

What types of Microspheres can be used?


All current Luminex microsphere products are compatible with the FLEXMAP 3D easing conversion
of current Luminex microsphere based research products to the new system. For commercially
available assay kits, check with the assay kit manufacturer to confirm assay compatibility and
available support with the FLEXMAP 3D system. If running an assay using xMAP Technology as an
in vitro diagnostic, the assay must be run according to the instructions provided with the assay.

What does the new bead map look like?


The new three-dimensional bead map is a matrix of the emitted fluorescence of the three internal
dyes on the x, y, and z axis. It is a 3D graph with ellipsoid targets for each region. The original 1-100
bead map is the base of the 500 plex bead map with the new regions emerging from it as the third
dye is added in increasing amounts.

How do the new microsphere sets compare to the
previous sets?
Each xMAP microsphere line, such as the MicroPlex Microspheres and MagPlex Microspheres,
is sourced from highly controlled progenitor microsphere lots to ensure homology of microsphere
functionality and surface chemistry. The same progenitor microsphere formulations and equivelant
dyeing proceses are used to create regions for the expanded FLEXMAP 3D map. Microsphere
integrity is maintained within the expanded map as the combinations of internal dyes do not affect
microsphere functionality or surface chemistry.

Warranty for the FLEXMAP 3D System


The FLEXMAP 3D system comes with a one year warranty. After the warranty year we strongly
suggest that end-users purchase a service agreement to cover system maintenance and repairs.
About nine months after the instrument purchase a Luminex Representative will contact you to initiate
this discussion. Please see the “Luminex Maintenance and Service Agreements Options” sheet for
more information about service contracts on the platform.

Upon initial installation, the xPONENT 4.0 software will open with a 90 day trial license. Prior to the
end of this 90 day period, you must contact Technical Support to setup a permanent access key for
the modules of software purchased with your system.

In order to assign your license key, Technical Support will need information about your Luminex
system. Please have the following information available:
• The serial number of your Luminex system

Additionally, Technical Support will need information provided in the xPONENT licensing export file.
To create this file:
• Open the Luminex xPONENT software.
• Select “Admin” from the toolbar.
• Select the “Licensing” tab.
• Click the “Export to File” button, which creates a text attachment that can be saved wherever is
convenient for you to retrieve it.
• Email this attachment to support@luminexcorp.com.

Once you have purchased your FLEXMAP 3D, you can later upgrade any of the optional modules by
contacting your Luminex partner or Luminex sales representative or by contacting Luminex Technical
Support.

Warranty for the Dell PC that is included with the
FLEXMAP 3D System
Dell provides a limited warranty with PCs shipped from Luminex. Upon receipt of the Dell PC that is
associated with your Luminex System, please complete the warranty transfer described below. This
will ensure the newly acquired PC is in your name and that you will receive technical support from
Dell for the remainder of your warranty period.

Required Items: Service Tag Number and Express Service Code (ESC) located directly on the Dell PC.

1. Complete the Current Customer Information by placing an X in any *REQUIRED field, except the
Service Tag and ESC fields.
2. Enter the Service Tag and ESC into the appropriate fields.

New Customer Information

1. Complete all *REQUIRED fields in this section with YOUR information. This is how your system
will be registered with Dell.
2. Once all *REQUIRED fields are completed, hit “Submit”. *Please ensure that you enter a correct
e-mail address. If, for any reason, your request fails to process immediately, you will be notified
at the e-mail address entered in the New Customer Information when Dell has completed the
request.
3. After submission, a successful ownership transfer will result in a confirmation message listing the
Service Tag numbers transferred.

4. If your transfer request fails to process automatically, you will receive a failure message. All
failures are automatically routed to the appropriate group within Dell for research and response.
You will be notified when your request has been completed or if additional information is needed.

purchased at the time of sale. You will have the option of later upgrading with additional software
modules should the need arise.

The System Requirements and Upgrade Checklist will assist in determining that your system
components are upgrade ready and that your customized xPONENT 4.0 Upgrade Kit contains your
required items.

Instrument Quick Guide
The FlexMAP 3D instrument that contains fluidics, optics, sheath fluid, and an platform for microtiter
plate movement. See the User’s Manual for information. Refer to the FLEXMAP 3D Hardware User’s
Manual, xPONENT 4.0 Software User’s Manual and reagent package inserts for complete instructions
and safety information, including electrical, mechanical, radiation, biohazard and chemical safety
information.

Use the below diagram as guidance for part accessibility:
System Preparation - Fluidics and Probe Height
1. Power FLEXMAP 3D system on using toggle switch on right side of instrument.
2. Ensure system has adequate sheath fluid in storage container and waste lines drain into a waste
location which is not full. Make sure sheath fluid container is at or below instrument level.
3. Click the xPONENT shortcut to start the software. Login to software using username and
password.
4. Click Eject on the System Status bar to eject the XY platform.

5. Add a 96 or 384 plate to the XY platform. For a filter plate a add large, flat disk to make sure
the probe does not puncture the well bottom. Otherwise no probe height adjustment tools are
needed.
6. From the Home page click Probe and Heater. On the Probe and Heater page, select the Plate
Type (96 well or 384 well) and enter a plate name.
7. Select the appropriate wells to calibrate probe height. This could be within the plate, the reservoir,
or the strip well locations in the off-plate reagent area. Make sure NO LIQUID is in the wells.
8. Click Auto Adjust Height. The probe will check heights in each well designated.

9. Click Save to save the plate settings. The plate name will appear when building assay protocols.
10. Readjust the probe height whenever using new plate types, after system maintenance, or when
troubleshooting an acquisition problem. Probe Height is the MOST COMMON error encounted.
Daily System Start-up
1. On the Home page, click System Initialization. This opens the Auto Maint tab on the Maintenance
page.
2. On the Admin, System Setup tab there are three options listed below that can be used for system
initialization. Calibration is required weekly , however Performance Verification should be done
daily to check system integrity and insure calibration is valid.

These instructions demonstrate option “a” above.
3. Make sure the Calibration Kit and Performance Verification Kit information has been imported into
the software using the CDs provided with the kits.
4. Select the correct kit lot numbers for your Calibration and Verification kit using pull down menus.
5. Eject the XY tray using the System Status bar.
6. Add two strip wells into the off-plate reagent area.
7. Notice the plate layout in the software which directs color coded reagent locations

8. Add water and 70% isopropanol or 70% ethanol to the correct reservoirs within the Off Plate
Reagent area.
9. Vortex all Calibration and Performance Verification Kit reagents for ten seconds each.
10. Using the color coded plate layout diagram in the Reagents area of the tab as a guide, add F3DCAL1,
F3DeCAL1, F3DCAL2 , and F3DCAL3 to positions SA1, SB1, SC1, and SD1 on the first well strip.
11. Add F3DVER1, F3DeVER1, F3DVER2, Fluidics 1, and Fluidics2 to SA2, SB2, SC2, SD2, and SE2
on the second well strip as shown in the plate layout.
12. Click the Run button. The run should take 10 to 45 minutes depending on which system initialization
option is selected and if the system is
already warmed up.
13. Once complete click Report to view the
Calibration and enhanced Performance
Verification report.
Notes:
• You can import individual CAL and VER
information from the Lot Management tab.
• You can run individual reagents on the
Cmds & Routines tab; however, the
Performance Verification reports will not be
generated. You can only run Fluidics as part
of the Performance Verification Routine.
• You can create custom routines on the
Cmds & Routines tab; however, they will
not generate the Performance Verification report in this manner.
• Causes for failing calibration or verification may be that the reagents are not vortexed, reagents in
wrong well locations, or selecting wrong kit lot values.

System Shutdown
Perform the System Shutdown routine every day after running the system. This routine sanitizes the
fluidics lines, removes residual buffer, and prevents clogs from forming in the sample probe.
1. On the Home Page, click Shutdown.
2. Add 20% bleach solution and water to the reservoirs designated on the plate layout.

3. Click Run.
Other Suggested Maintenance:
Any time you experience acquisition problems (or once a week if running debris ridden samples) we
suggest the following:
1. Remove the sample probe following instructions that can be found in the FLEXMAP 3D User’s
manual and place it in a sonicator bath for five minutes.
2. Rinse the probe with water.
3. Replace and readjust the probe height.
4. Run the Weekly Maintenance routine on the Maintenance page, Cmds & Routines tab.
5. Calibrate the system and run Performance Verification.

You can run individual commands and routines from Cmds & Routines tab. When running
calibration or performance verification reagents be sure that the selected lot number is the current
active lot on the Lot Management tab.

Collecting Data Without a Previously Created Protocol:
1. On the Home page click Create a New Batch from a new Protocol.
2. On the Settings tab click:
• Name
• Bead Type (MagPlex or MicroPlex)
• Volume to aspirate (20 to 200 ul)
• DD gate (usually 7500 to 15000)
• Reporter Gain (Standard or Enhanced PMT)
• Analysis Type (Quantitative, Qualitative, Allele Call, or none)
• Enter number of Standards and Controls as needed
3. Click Next

4. On the Analytes tab enter:
• Analyte (Bead/Region number)
• Names for each analyte (notice by right clicking you can paste a list)
• Default Analysis Changes (weighting, type of curve fit)
• Click Next
Notice that by right clicking you are given the option of pasting data into the software

5. The Stds & Ctrls tab will only require information if the assay analysis type requires standards or
controls.
• To use standards and controls already in the system, click Apply Std/Ctrl Kit.
• To create a new kit, enter kit information at the top of the screen.
• For each standard or control, enter information in the tables. Notice you can right click to

past values in from a text or spreadsheet.
• Under Admin page, Batch Settings tab you can set up kits so that entry of standard specific
information (name, lot #, exp date) is optional.
• To set up a dilution series, enter the value of the highest standard in the analyte column
and select one of the preset Dilution options (1:2, 1:5 or ½ log). You can also type your
own dilution factor in the box (1:10, 1:100, or 1:0.33 for a lowest to highest dilution curve).
Standards can also be resorted and dilution curves applied as needed.
• Click Next.
6. The Plate Layout tab helps you set up where samples will be placed on the plate.
• To add well commands, highlight the appropriate wells and mark them as Unknown, Standard,
Control, Background, or Wash. You can also delete commands that you’ve added, and
change the starting location on the plate. If you wish to run in replicate, change the Replicate

Count to the appropriate number and the grouping to your preferred grouping method.
• To add maintenance commands, choose the desired command from the drop-down list.
Highlight the well that you want to apply it to, and then choose Before Well Command or After
Well Command. If you are working with more than one plate, choose the Add & Change Plate

• As you add commands to your plate, they appear in the command sequence chart. Here, you
can give each of your wells an ID. You can also import an ID list and move your commands
up and down in the command sequence chart.
a. Click Save to save the batch and run it later.
b. Click Save Pcol to save the protocol and standard and control information to use
later for another batch.

c. Click Run Batch to begin data acquisition.
Collecting Data Using an Imported or Previously Created Protocol:
1. On the Home page, highlight the protocol.
2. Click Create a batch using a previously created protocol.
3. Enter a batch name and confirm the selected protocol.
4. Click Next to open the Std &Ctrl tab. Confirm the standard and control lots selected or choose another.
5. Click Next to open the Plate Layout tab. Add or remove samples as desired.
6. Eject the XY platform and insert the plate. Be sure you have adjusted the probe height for that
particular plate.
7. Click Run Batch.
Notice you can also Save this batch as pending and run at a later time or you can extract the protocol
for use later with other batches by clicking Save Protocol. This is most useful if you changed
protocol defaults during batch setup.

Data Analysis
1. Open the Results page, then open the Saved Batches tab
2. Select the batch of interest by double clicking.
3. Select from Statistic to look at all analyte data at once or choose Analyte to look at individual
analyte data.

Generating Reports
Reports can be generated for collected batch data, calibration results, performance verification
results and system status. To generate a report:
1. On the Results Page, click the Reports tab.
2. Select the report. Batch, Calibration Summary, Performance Verification, Protocol or System Log
reports are available.
3. Select the type and analytes if needed.
4. Click Generate.
