Extraction of components of Advil PM tablet

Active Ingredients are –


• Ibuprofen= 200mg/tablet
• Diphenylhydramine citrate= 38mg/tablet

Solvents for ibuprofen
➢ For ibuprofen [acidic drug] n-hexane, n-heptane, cyclohexane,
• Crush one tablet dissolve it in 15ml of distilled water. methylcyclohexane, toluene and
isooctane.
• Use filter paper in e. flask to remove binders.
• Check the Ph of tablet it must be 7<.
• In separating funnel take the filtered sample.
• Add 60 ml of n-hexane to it [sample 1:3 hexane].
• Shake it properly approx. for 10 minutes or until get the aqueous layer and organic
layer separated.
• Take out the organic layer [depends on the density of solvent with respect to water.
✓ Solvent with low density must be at the upper side of separating funnel.
• Repeat it one more time .

➢ For diphenylhdramine citrate [basic drug]


• Crush the tablet dissolve it into 15 ml of of distilled water.
• Use filter paper to remove the binders.
• Check the Ph it must be basic otherwise add some base to it like ammonia to make it basic
similar to the ph of our drug of interest.
• In separating funnel take the filtered sample.
• Add sample to separating funnel with 15 ml of chloroform and 60 ml of n-hexane
[chloroform 1:3 n- hexane]
• Shake it until get the two layers properly. Solvents for diphenylhydramine
• Take out the organic layer
n-hexane, n-heptane, cyclohexane,
• Repeat it one more time
methylcyclohexane,
dichloromethane and chloroform

Keep the extracted sample of both the drugs to vaporize until it remains in drops which
are sufficient to apply on TLC Plate.
