1. Safety
1.1 Instrument Safety
1. Always follow basic safety precautions when using this product to reduce the
risk of injury, fire or electrical shock.
2. Read and understand all information in the IFU. Failure to read, understand
and follow the instructions may result in damage to the product, injury to
operating personnel or poor instrument performance.
3. Observe all STOP and CAUTION statements in the IFU.
4. Never open Washer 430 while the instrument is plugged into a power
source.
5. Observe proper laboratory safety precautions, such as wearing protective
clothing and using approved laboratory safety procedures.

Note
Suitable Compact Microplates
The Washer 430 allows to process flat-bottom or round-bottom
microplates in 96-well format typically used for EIA, conforming to the
standard defined by the Society of Biomolecular Screening (see
chapter 2.2.1 Microplates). Other plate types in 96-well format such as
deep-well plates, PCR-plates and filtration plates cannot be used with
the Washer 430

Note
Suitable Strip-plates
The Washer 430 allows to process flat-bottom or round-bottom strip-
well plates in 96-well format, typically used for EIAs, showing a similar
height and footprint as the 96-well microplates listed above.
To achieve proper wash-performance with strip-well plates ensure
that:
• all strips have been properly inserted into the frame of the strip-
plate to give a level surface
• the actual position of the strips within a strip-plate matches the
position of the strips as defined in program used for
processing. See chapter 4.4.2 Procedure to Wash Individually
Selected Strips.

Caution
Be careful when using strip-plates that the strips are positioned
in the microplate as defined in the program used for processing,
otherwise spilling can occur and the instrument may become
contaminated. (see chapter 7.1 Cleaning Procedures).

WARNING
ADJUSTMENT OF WASHER 430 TO A CORRESPONDING MICROPLATE
TO ENSURE PROPER WASH PERFORMANCE IT IS MANDATORY TO
ADJUST THE WASHER 430 TO THE TYPE / MANUFACTURER OF
MICROPLATE OR STRIP-PLATE USED. THIS ALSO APPLIES FOR ANY
PRE-DEFINED PLATE FILE, THAT WILL ALWAYS CONTAIN AVERAGE
PLATE PARAMETERS ONLY, THAT HAVE TO BE VERIFIED WITH THE
CORRESPONDING PLATE TYPE AND IF NECESSARY CORRECTED
BEFORE PUTTING THE WASHER 430 INTO USE.
IF THIS ADJUSTMENT PROCEDURE IS NOT PERFORMED PROPERLY,
THIS MIGHT RESULT IN HIGH LEVELS OF RESIDUAL VOLUME LEFT PER
WELL, AS WELL AS IN INSUFFICIENT WASHING OF THE WELLS AND
MAY SERIOUSLY AFFECT ASSAY PERFORMANCE.
FOR DETAILS ON HOW TO ADJUST THE WASHER 430 TO THE TYPE OF
MICROPLATE OR STRIP-PLATE USED, SEE CHAPTER 6. ADJUST MENU
IN THIS DOCUMENT.

WARNING
WASTE BOTTLE - FILLING LEVEL
MAKE SURE THAT THE FILLING LEVEL OF THE WASTE BOTTLE IS
ALWAYS KEPT BELOW THE MAXIMUM LEVEL INDICATED ON THE
BOTTLE TO AVOID POTENTIAL OVERFLOW. AS THE CONTENTS OF
WASTE BOTTLE ARE POTENTIALLY INFECTIOUS, WEAR PROTECTIVE
CLOTHING (GLOVES, LAB COAT AND SAFETY GLASSES) WHEN
EMPTYING / HANDLING A WASTE BOTTLE. FOLLOW LOCAL STANDARD
OPERATING PROCEDURES ON HOW TO TREAT POTENTIALLY
INFECTIOUS MATERIAL FOR DISPOSAL.

Note
For on-line monitoring of liquid levels in wash buffer-bottles, rinse-bottle
and waste-bottle an optional Liquid Level Sensor System
is available for the Washer 430.
See Chapter 2.4 Instrument Options for further information.

WARNING
WITH SPECIFIC WASH BUFFERS THAT SHOW A STRONG TENDENCY
OF FOAMING, EMPTY THE WASTE BOTTLE AS SOON AS THE FOAM-
LEVEL HAS REACHED THE MAXIMUM FILLING LEVEL INDICATED ON
THE WASTE BOTTLE. ADDITIONALLY ADD A COMMERCIALLY
AVAILABLE ANTI-FOAMING AGENT (SUCH AS SILICONE OIL) TO THE
EMPTY WASTE BOTTLE TO HELP REDUCE FOAMING.
IF FOAMING CONTINUES TO BE A PROBLEM WE RECOMMEND TO
SWITCH TO A LARGER WASTE BOTTLE (NOT PROVIDED WITH THE
WASHER 430) AND ADDITIONALLY INCREASE THE CONCENTRATION
OF ANTI-FOAMING AGENT IN THE WASTE BOTTLE. TO HELP
FACILITATE BREAK-DOWN OF THE FOAM IN THE WASTE BOTTLE,
CAREFULLY SWIRL THE WASTE BOTTLE FROM TIME-TO-TIME TO
IMPROVE MIXING BETWEEN FOAM LAYER AND ANTI-FOAMING
AGENT.
REFILL ANTI-FOAMING AGENT AFTER EMPTYING WASTE BOTTLE.
FOR EXAMPLE WHEN USING THE WACKER ANTI-FOAM EMULSION
SE47 (WACKER ARTICLE CODE 21640582) THE RECOMMENDED
CONCENTRATION IS 1ML OF ANTIFOAMING AGENT FOR 1 LITER OF
WASTE SOLUTION.
FOR ANTIFOAMING AGENTS FROM OTHER MANUFACTURERS USE
CONCENTRATIONS AS RECOMMENDED BY CORRESPONDING
MANUFACTURERS.

2. General
2.1 Introduction
The Washer 430 is a microprocessor-controlled washer for strip-wise processing
of microplates in the 96-well format. The Washer 430 is a general purpose
laboratory instrument that is designed for professional use only and should only
be operated by trained personnel.
The instrument is fitted with an 8 way manifold, which is fitted with one aspirating
and one dispensing needle per well.

Note
12 and 16 way manifolds are only available on special request for the
Washer 430.

The instrument can use up to nine software defined plate types that control the
dispensing and aspirating positions for round or flat bottomed plates.
The dispensing and aspirating speeds can also be selected from five set speeds.

Caution
If the instructions given in this document are not correctly performed, the
instrument may be damaged or the procedure may not be correctly
performed and the safety of the instrument cannot be guaranteed.

Caution
The wash results obtained with the Washer 430 are influenced by the correct
use of the instrument, according to the instructions given in this IFU, as well
as the liquid compounds used (reagents, wash buffer, chemistry). The
instructions for use, storage and other manipulations in connection with
samples or reagents have to be strictly followed. Taking this fact into
consideration, results must be interpreted carefully.

Caution
Before the instrument is installed and switched on,
it should be left to stand for at least three hours, so there is
no possibility of condensation causing a short circuit.

Caution
Before washing procedures are started, make sure that the microplate
position A1 is inserted correctly.

compact 96-well microplates and may vary using other 96-well flat
bottom plates or 96-well strip-plates.

2.2.1 Microplates
Only 96-well format microplates (v-shaped, flat and round bottom including
strip-wells) that are in accordance with the ANSI/SBS standards can be used with
the Washer 430. Microplates supplied by bioMérieux are in accordance with the
ANSI/SBS standards.

The Washer 430 is operated via a keyboard and a control panel consisting of a
two row, sixteen digit Liquid Crystal Display (LCD) unit.
The top row of the display unit is used for messages and the bottom row for the
key functions or options.
A Light Emitting Diode (LED) in the center of the letter O of the word POWER
indicates when the washer is switched ON.

2.4 Instrument Options


A trained service engineer can install the instrument with the following options:

That it may not be possible to upgrade to some options on certain

Option A : Upgrade from 1 inlet channel to 4 inlet channels


combined with additional Liquid Sensor System.
The Liquid Sensor System is able to detect when
the solution bottles are empty and when the waste
bottle is full. The weight indicator is inside the bottle
holder. The standard Washer 430 instrument is
equipped with one unit and the additional unit can
be ordered as part of Option A.

Field upgrade for option A is available from serial


number 6520001 onwards.

Option B : Fill Verification option (8 channel manifold only) is


available from serial number 6520070 onwards.
This function allows to actively check during an
overflow wash step, if wash buffer is dispensed into
every single well of a 96 well plate.
It is based on conductivity measurement between
the dispensing needles and aspirating needles
(special manifold required). The conductivity of the
wash buffer has to be within a range of 5 mS / cm
to 30 mS / cm.

Please note that it is possible to combine option A


and B in the instrument.

The Fill Verification Option is possible from firmware level 3.23 onwards.

2.5 Instrument Software Features


The instrument has the following software features:
• 30 user definable washing programs, stored under the numbers 1 to 30.
• Up to 60 processing steps per program, each processing step can be
identical or different from the previous step.
• 3 Washing Modes, Dilution Washing, Bottom Washing and Overflow
Washing.
Set using the Plate type parameters.
• Soak Time selectable from 1 to 999 seconds.
• Software selectable Dispensing Speeds
• Software selectable Aspirating Speeds
• Software selectable Bottom position, for aspiration close to the bottom of
the wells.
• Three Rinse Modes, the different rinse modes can be used to select how the
instrument is rinsed before it is left to stand or switched off.
• Automatic Microplate Centering, the microplate is automatically centered
before the start of the washing program.
• Programmable Strip Selection, the program can be defined so that the
strips to be washed can be selected before starting the washing procedure.

2.6 Washing Sequence Modes


The instrument can wash a microplate using the following sequence modes:
Plate Mode Each process step is performed on all of the strips sequentially,
before proceeding to the next process step. The entire plate is
processed for the defined soaking time (as opposed to the Strip
Mode).
Strip Mode The entire Wash program is performed on a strip before
proceeding to the next strip. Each individual strip is processed for
the defined soaking time, before proceeding to the next strip.

2.6.1 Washing Modes


The instrument can use the following washing modes to wash a microplate:
Overflow Washing Overflow washing consists of a simultaneous
aspiration and dispense step. It creates a circular
flow of wash buffer in the well.
Bottom Washing Washes only defined segments of the well
according to the selected aspiration position.
Dilution Washing Dilution washing consists of a sequence of
aspiration and dispense steps.

2.6.2 Aspirating Positions


To ensure that all the wash solution is correctly aspirated from the wells, the
manifold must be positioned so that the aspirating needles are correctly
positioned in the wells for round-bottom, v-shaped bottom or flat-bottom well
microplates.
All settings are defined in the Plate Type setting option of the Adjust menu.
With round or v-shaped bottom wells, the aspirating needles are placed in the
middle of the wells.

With flat bottom wells, the instrument performs crosswise aspiration.
The aspirating needles are set at two positions on the bottom of the wells (front
edge and back edge).

3.1 Unpacking Procedure


1. Visually inspect the container for damage before it is opened.
Report any damage immediately.
2. Select a location for the instrument that is flat, level, vibration free, away
from direct sunlight, and free from dust, solvents and acid vapors. Allow 10
cm distance between the back of the instrument and the wall or any other
equipment.
3. Place the carton in an upright position and open it.
4. Lift the instrument out of the carton and place it in the selected location.
5. Visually inspect the instrument for loose, bent or broken parts.
Report any damage immediately.
6. Compare the serial number on the rear panel of the instrument with the
serial number on the packing slip.
Report any discrepancy immediately.
7. Check the instrument accessories against the packing list.
8. Save packing materials for future use.

3.2 Power Requirements


The instrument is designed to operate at either 100 - 120 volt or 220 - 240 volt.
No voltage setting is required as the washer automatically senses the supplied
voltage.
The washer requires 2 x T 1.6 A / 250 V (slow blow) fuses.

WARNING
TO PREVENT THE RISK OF FIRE, THE MAIN FUSES SHOULD ONLY BE
REPLACED WITH THE SAME TYPE AND RATING OF FUSE.

Before the instrument is installed and switched on,
it should be left to stand for at least three hours, so there is
no possibility of condensation causing a short circuit.

3.3.1 Installing the Instrument


• Unpack the instrument and check the contents.

• Connect the solution tubes to the inlet connectors.
See below 3.3.2 Connecting the Solution Tubes.
• Setup instrument options if included. See 2.4 Instrument Options.

3.3.2 Connecting the Solution Tubes


The solution tubes are connected onto the inlet connectors on the back panel of
the instrument. The instrument is supplied with four meters of tubing that can be
cut to the required length (maximum 1 meter).

Connect the detector unit cable into the appropriate socket in the back panel of
the instrument. The liquid sensor system can be extended to control up to four
different liquid bottles and one waste bottle.

4.1 Switching the Instrument On
Ensure the instrument has been correctly installed and the power cable is
connected into the power cable socket in the back panel of the instrument.
Switch on the instrument, using the on/off switch in the back panel of the
instrument.
The initialization procedure is performed and, depending on the washer type, one
of the following initialization messages is displayed:

Version V X.x is the instrument's software version.
If the Rinse: Night rinsing procedure was not performed before the instrument
was switched off, the following message is displayed:

Caution
The instrument must be primed before it can be used. Please ensure that the
dispensing and aspiration pumps are not run for longer than a few minutes

distilled water to ensure the proper performance of the Washer 430 and to
prevent needles from becoming blocked, for details see 7.1 Cleaning
Procedures.

WARNING
BEFORE THE INSTRUMENT CAN BE USED, ALL DISPENSING
CHANNELS NEEDED FOR THE WASH STEP MUST BE PRIMED TO FILL
THE DISPENSE SYSTEM WITH THE REQUIRED SOLUTION.
IF THE PRIMING PROCEDURE IS NOT PROPERLY PERFORMED, THIS
MAY RESULT IN INSUFFICIENT WASHING OF THE WELLS AND MAY
SERIOUSLY AFFECT ASSAY PERFORMANCE.
ENSURE THAT THE BOTTLE OF PRIMING SOLUTION IS ALWAYS FULL
AT THE BEGINNING OF THE PRIMING PROCEDURE AND CHECK THAT
THE INLET FILTERS IN THE LIQUID TUBES ARE CLEAR.

Caution
Ensure that the dispensing and aspiration pumps are not run for longer
than a few minutes without liquid or they will be damaged.

4.4.1 Performing a Wash Procedure
Insert the microplate to be washed into the plate carrier and ensure that the
microplate is correctly oriented for the type of manifold fitted.

The operating procedure is dependent on the instrument options and the program
settings.

Caution
Before washing procedures are started, make sure that
the microplate position A1 is inserted correctly.

Note
Adjustment of Washer 430 to a corresponding microplate
To ensure proper wash performance it is mandatory to adjust the Washer 430 to
the type / manufacturer of microplate or strip-plate used. This also applies for
any pre-defined plate file, that will always contain average plate parameters
only, that have to be verified with the corresponding plate type and if necessary
corrected before putting the Washer 430 into use.
If this adjustment procedure is not performed properly, this might result in high
levels of residual volume left per well, as well as in insufficient washing of the
wells and may seriously affect assay performance.
For details on how to adjust the Washer 430 to the type of microplate or strip-
plate used, see chapter 6. Adjust Menu of this document.

Suitable Compact Microplates
The Washer 430 allows to process flat-bottom or round-bottom
microplates in 96-well format typically used for EIA, conforming to the
standard defined by the Society of Biomolecular Screening
(see chapter 2.2.1 Microplates). Other plate types in 96-well format such as
deep-well plates, PCR-plates and filtration plates cannot be used with the
Washer 430

Suitable Strip-plates
The Washer 430 allows to process flat-bottom or round-bottom strip-well
plates in 96-well format, typically used for EIAs, showing a similar height
and footprint as the 96-well microplates listed above.
To achieve proper wash-performance with strip-well plates ensure that:
• all strips have been properly inserted into the frame of the strip-
plate to give a level surface
• the actual position of the strips within a strip-plate matches the
position of the strips as defined in program used for
processing. See chapter 4.4.2 Procedure to Wash Individually
Selected Strips.

Caution
Be careful when using strip-plates that the strips are positioned
in the microplate as selected in the program, otherwise spilling
can occur and the instrument may become contaminated.
(see chapter 7.1 Cleaning Procedures).

When the instrument is busy, do not touch the manifold.
After the instrument has been used the manifold may be infectious!

Fill Verification
The Fill Verification option is a method of checking the filling of the wells in a
microplate. Possible errors with the dispensing system as well as the aspiration
system can be recognized.
The Fill Verification only works with the OVERFLOW wash procedure.
The wash buffer forms an electrical connection between the dispensing needles
and the aspirating needles. As soon as the wash liquid touches the aspirating
needles the electric circuit is closed.

The following errors can be detected using this method.


• Fill Dispense Error
• Fill Aspiration Error
• Fill Function Error
Fill Dispense Error
The error messages Inadequate wash, Check disp path are displayed.
These error messages appear if some of the needles are dispensing no or too
little solution. Possible errors could be a result of blocked dispensing needles,
bubbles in the dispensing channels, empty liquid bottles or the dispensing pump
no longer functions.
If no connection between the dispensing needles and the relevant aspirating
needles is made at least 30 µl before the end of the dispensing step, the wash
program will be interrupted.
Fill Aspiration Error
The error message Fill Asp. Err. is displayed.
In order to prevent the wells from overflowing, the instrument checks when the
individual aspirating needles come into contact with the wash solution.
If this happens before 200 µl has been dispensed into the wells the program is

Fill Function Error


The error message Fill Funct. Err is displayed.
Before each wash procedure and each row the program checks whether there is
an electrical connection between the dispensing and aspirating needle(s). If a
connection exists, the dispense and aspiration errors can not be detected and the
program is interrupted immediately or will not even be started
The underside of the manifold must then be cleaned, as the deposits from the
wash buffer can produce an electrical connection between the needles, which
could prevent the Fill Verification option from functioning correctly.
Test Conditions
The following conditions are required for the Fill Verification to function flawlessly.
• The conductance of the wash buffer must be between 5 mS/cm and
30 mS/cm.
• The amount of wash liquid dispensed must produce at least 100 µl overflow.
• At least 450 µl should be dispensed with an Overflow Position slightly above
the upper well limit.

Conductance of some Standard Wash Buffers


Most of the commercial wash buffers lie in the mentioned conductance ranges.
The conductance is proportional to the wash buffer concentration.

The following values were measured using a normal dilution:


Wash buffer SIGMA: 1 tablet in 200ml - 14.7 mS/cm
Phosphate buffered saline Tablets:

This message may also be displayed, if the Fill Verification or Manifold Detection
options are enabled but the required parts are not fitted to the instrument. In this

Transport Error
If the instrument cannot move the plate carrier, the following message is
displayed:

Waste bottle - Foaming
With specific wash buffers that show a strong tendency of foaming, it is
recommended to add a commercially available anti-foaming agent (such
as silicone oil) to the empty waste bottle to help reduce foaming.
If foaming continues to be a problem we recommend to additionally
increase the concentration of anti-foaming agent in the waste bottle.
To help facilitate break-down of the foam in the waste bottle, carefully
swirl the waste bottle from time-to-time to improve mixing between foam
layer and anti-foaming agent.
Refill anti-foaming agent after emptying waste bottle. For example when
using the Wacker Anti-foam emulsion SE47 (Wacker article code
21640582) the recommended concentration is 1ml of antifoaming agent
for 1 liter of waste solution.
For antifoaming agents from other manufacturers use concentrations as
recommended by corresponding manufacturers.

Liquid Bottle Empty


If the liquid bottle is empty before a program is started, the following message is
displayed:

Switch off the instrument to clear the display. Ensure that the small ribbon cable
and the interface cable (from the mainboard) are connected correctly to the Fill
Verification board. Switch the instrument on again.
Contact the service engineer to disable the Fill Verification option in the
Service menu and to correct the problem.

If the fill verification board needle status can not be deleted, the following error
message is displayed:

The problem is caused by an electrical conducting liquid connection from the
dispensing to the aspirating needles on the manifold. The underside of the
manifold must then be cleaned using, for example, distilled water.

Service menu and to correct the problem.
Fill Aspiration Error
When the well is not correctly aspirated the following error is displayed:

The problem is caused by a blocked aspirating needle / tube or the aspirating
pump may not be functioning correctly. Use the cleaning needles to clean the
aspirating needles (see 7. Maintenance and Cleaning) or replace the pump.

4.6 End of Operation


At the end of operation, the instrument can be either left switched on with the
manifold in the rinsing solution or switched off.
Instrument Left Switched On
Use the following procedure to leave the instrument switched on with the manifold
soaking in the distilled water at the end of operation:
Place the wash solution tube into a bottle containing distilled water.
Use the Rinse: Night rinsing procedure to rinse the wash system.
(see 7. Maintenance and Cleaning).
When the rinsing procedure is completed the following message is displayed:

Stop

If the manifold is to be soaked overnight, do not switch the instrument off.


Instrument Switched Off
If the instrument is to be switched off at the end of operation the Rinse: Day
rinsing procedure should be performed to prevent the needles becoming blocked.
Place the wash solution tube into a bottle containing distilled water.
Use the Rinse: Day rinsing procedure to rinse the wash system.
When the rinsing procedure is completed, switch the instrument off.

To edit a defined program, proceed as described in the Defining a New Program
section and edit the parameters as required. If the program is locked it can not be
edited with the onboard software and the following error message is displayed:

microplates in 96-well format typically used for EIA, conforming to the
standard defined by the Society of Biomolecular Screening. Other plate
types in 96-well format such as deep-well plates, PCR-plates and
filtration plates cannot be used with the Washer 430

The Washer 430 allows to process flat-bottom or round-bottom strip-
well plates in 96-well format, typically used for EIAs, showing a similar
height and footprint as the 96-well microplates listed above.

6.3 Adjust Liquid Sensors


Note
Every Washer 430 is delivered with
the liquid sensor system pre-calibrated.
Calibration is only necessary if Option A is refitted in the instrument.

Caution
This procedure must be performed when Option A is
refitted in the instrument.

This procedure allows a warning level to be determined. This informs the


user when the level of liquid in the bottles is too low or too high.
When the level of liquid in the liquid bottles 1 & 2 (and optionally 3 & 4) is below
600ml and the level of the liquid in the waste bottle is above 2000ml the
instrument will emit an acoustic signal to alert the user.
Fill the bottles before performing the adjustment procedure. The bottles marked
green (wash solution) and gray (rinse solution) should be filled with 600ml of
water and the bottle marked red (waste) should be filled with 2000ml of water. For
Option A and option A & B use similar liquid volumes (600 ml) for the purple and
blue bottles. Place all bottles on the corresponding colored bottle holders. Go to
the Adjust menu and select Adj. Liq. Sens.

7.1 Cleaning Procedures
WARNING
ALL PARTS OF THE INSTRUMENT THAT COME INTO CONTACT WITH
POTENTIALLY INFECTIOUS MATERIAL MUST BE TREATED AS
POTENTIALLY INFECTIOUS AREAS. IT IS ADVISABLE TO WEAR
PROTECTIVE POWDER-FREE GLOVES WHEN PERFORMING THE
CLEANING PROCEDURE AND ALSO WHEN MAKING ADJUSTMENTS TO
THE INSTRUMENT.

The most important cleaning procedure for this instrument is to rinse the liquid
system with distilled water before the instrument is left to stand or switched off at
the end of each day.
The manifold should be removed and thoroughly cleaned at least once a month
or if one or more of the needles are blocked.

Caution
If the manifold is not rinsed, the needles will become blocked.
This will result in the manifold either needing expensive repairs
or it could become completely ruined.

7.1.1 Outer Surface of the Instrument Housing


The outer surface of Washer 430 may be cleaned periodically using a tissue
moistened with a mild detergent solution.

7.1.2 Display Window Cover


The display window cover on the alphanumeric display may be cleaned
periodically using a tissue moistened with a mild detergent.

7.1.3 Cleaning Pumps and Tubes


To clean the pumps and tubes perform Rinse-Night on a daily basis. (See chapter
7.5 Rinse Modes).

7.1.4 Manifold Cleaning


The most important cleaning procedure for this instrument is to rinse the wash
system before the instrument is left to stand or switched off at the end of each
day.

Caution
If the manifold is not rinsed, the needles will become blocked.
This will result in the manifold either needing expensive repairs
or it could become completely ruined.

WARNING
All parts of the instrument that come into contact with potentially
infectious material must be treated as potentially infectious areas. It is
advisable to wear protective powder-free gloves when performing the
cleaning procedure and also when making adjustments to the instrument.

The manifold can be cleaned using:


• The supplied cleaning needles (accessory box)
Carefully push the cleaning needles into the aspirating and dispensing
needles.
The small cleaning needle is for the dispensing needles and the large
cleaning needle is for the aspirating needles.
Rinse the manifold block with distilled water to ensure that all particles have
been removed.
• A gentle ultrasonic bath of warm distilled water for 15 minutes.
• Autoclaving (max. 130 °C , max. five times)

Note
Autoclaving is only intended for use with a manifold that is not fitted with
the Fill Verification option. Manifolds with the Fill Verification option
should only be cleaned in an ultrasonic bath. If additional cleaning is
required, use a small brush. Do not use autoclaving.

Note
For instruments fitted with the Fill Verification Option, the underside of the
manifold must also be cleaned daily to avoid Fill Function Errors (see
chapter 4.5.4 Instrument Fitted with Fill Verification (Option B).

Should it become necessary to take the manifold apart (possibly infectious!) take
care to insert all seals properly when reassembling the manifold.
Twisted or missing seals may result in a leaking manifold!
After the manifold has been cleaned, reconnect the tubing and replace it into the
instrument. Tighten the manifold into place using the two screws, ensure that the
screws are tight.
When the manifold has been refitted, switch the instrument on and perform the
priming procedure using distilled water.

7.1.5 Cleaning Waste Bottles

WARNING
BEFORE CLEANING THE WASTE BOTTLES EMPTY THEM ACCORDING
TO DISPOSAL REGULATIONS (SEE CHAPTER 7.7.3 DISPOSAL OF
OPERATING MATERIAL).
THE BOTTLES MUST BE CLEANED REGULARLY DEPENDING ON THE
APPLICATIONS, USING A MILD DETERGENT.

Note
It is advisable to wear disposable powder-free gloves
when handling waste bottles.

7.1.6 Liquid or Foam Spills


Spilling of liquid or foam may occur when the Washer 430 is operated improperly
such as:
• Used microplate not matching installed manifold
• Position of the strips in the strip-plate do not match the positions defined in
the program used for processing
• Plate parameters not properly adjusted
• Waste bottle not emptied when liquid level or foam level reaches maximum
filling level
• No anti-foaming agent used with wash buffers showing strong tendency to
foam
WARNING
WASTE BOTTLE - FILLING LEVEL
MAKE SURE THAT THE LIQUID LEVEL OF THE WASTE BOTTLE IS
ALWAYS KEPT BELOW THE MAXIMUM LEVEL INDICATED ON THE
BOTTLE TO AVOID POTENTIAL OVERFLOW. AS THE CONTENTS OF
WASTE BOTTLE ARE POTENTIALLY INFECTIOUS, WEAR PROTECTIVE
CLOTHING (GLOVES, LAB COAT AND SAFETY GLASSES) WHEN
EMPTYING / HANDLING A WASTE BOTTLE. FOLLOW LOCAL
STANDARD OPERATING PROCEDURES ON HOW TO TREAT
POTENTIALLY INFECTIOUS MATERIAL FOR DISPOSAL.

WARNING
ALWAYS SWITCH-OFF WASHER 430 BEFORE REMOVING ANY KIND
SPILLS ON THE INSTRUMENT. IF THE SPILL OCCURS IN THE
INSTRUMENT A SERVICE TECHNICIAN IS REQUIRED.

WARNING
WHEN CLEANING SPILLS ALWAYS WEAR DISPOSABLE POWDER-
FREE GLOVES AS WELL AS PROTECTIVE GLASSES AND CLOTHING.
All spills (liquid or foam) have to treated as potentially infectious. Therefore
always wear protective powder-free gloves and protective clothing when cleaning
up a spill. Additionally, all resulting waste has to be treated as potentially
infectious and the disposal has to be performed according to the information
given in chapter 7.7.3 Disposal of Operating Material.
Always remove spills immediately after they have occurred. Use paper tissue to
soak-up spills and wipe-surface dry. Carefully disinfect surface were spill has
occurred according to the disinfection procedure in chapter 7.6.1 Disinfection
Procedure.

7.2 Manifold Removal

WARNING
AFTER THE INSTRUMENT HAS BEEN USED,
THE MANIFOLD MAY BE INFECTIOUS.
BEFORE THE MANIFOLD IS REMOVED IT MUST BE THOROUGHLY
DISINFECTED AND OBSERVE PROPER LABORATORY SAFETY
PRECAUTIONS, SUCH AS WEARING PROTECTIVE CLOTHING AND USING
APPROVED LABORATORY SAFETY PROCEDURES.

The manifold should be removed and thoroughly cleaned at least every 6 months
or if one or more of the needles are blocked.
Lift the manifold arm and remove the two screws that hold the manifold onto the
holder.

7.2.1 Replacing the Manifolds


The Washer 430 is delivered with the manifold already installed.
The instrument can be used with the following types of manifolds:
• Standard (8 way manifold)
• Fill verification Factory installation required

The manifold is fitted to the instrument using the following procedures:


1. Lift the manifold arm.
2. Carefully fit the manifold onto the manifold arm and ensure that the locating
pins are correctly inserted through the holes in the plate.
3. If the manifold detection option is fitted, ensure that the manifold type
indicator is correctly fitted between the manifold type detector switches.
4. Tighten the manifold into place using the two screws.
5. Fit the dispensing tube onto the top connector on the rear of the manifold.
6. Fit the aspirating tubing (marked with red rings) onto the two bottom
connectors on the rear of the manifold.
7. If the Fill Verification option is fitted, connect the small ribbon cable onto
the connector block on the left hand side of the circuit board in the manifold
arm.

7.2.2 Replacing the Plate carrier

The instrument is delivered with the plate carrier already installed. If the plate
carrier is removed, for example: for cleaning, it can be replaced using the
following procedure:
1. Lift the manifold arm
2. Slightly tilt the plate carrier towards the right.

3. Carefully slide the plate carrier with the waste sink at the rear over the
guide bar.

The plate carrier must be slightly tilted so that it goes over the pin that activates
the automatic microplate centering mechanism.
To reinstall the plate carrier follow the procedure listed below:
1. Push the plate carrier so that it is fully inserted into the instrument.
2. Lower the plate carrier.
3. Lower the manifold arm.

7.3 Replacing the Main Fuses

WARNING
TO PREVENT THE RISK OF FIRE, THE MAIN FUSES SHOULD ONLY BE
REPLACED WITH THE SAME TYPE AND RATING OF FUSE.

The following steps must be performed to replace the Main Fuses, which are
located by the power cable connection, in the rear panel of the instrument.
1. Switch off the instrument and unplug the power cord.

right hand side of the compartment. Remove the compartment out and to the

7.4 Preventive Maintenance Plan

7.4.1 Daily
• Perform Rinse Night with distilled water.

• Check the liquid filter(s) in wash bottle(s) for particles and rinse liquid filter(s)
with water.

7.4.3 Every Six Months


• Clean the plate carrier guide bar with 70% ethanol.
• Check the centering mechanism of the plate carrier and clean if necessary
with 70 % ethanol.
• Clean the Manifold aspirating and dispensing needles using the cleaning kit
supplied with the instrument.

7.4.4 Yearly (Service Engineer Required)


• Save the customers specific programs using the WinWash Clinical software.
• Disinfect the instrument. (See 7.6 Instrument Disinfection)
• Replace all aspirating and dispensing tubes.
• Replace the liquid filter(s) in the wash bottle(s).
• Replace the manifold sealing and clean the inner parts of the manifold.
• Check the aspirating pump and replace if necessary.
• Recalibrate the dispensing pump.

7.5 Rinse Modes


There are three rinse modes to flush the liquid system and prevent needle
blockages. They should be carried out when the instrument is left to stand or is
switched off at the end of operation.
The Rinse modes are:

This procedure is used to rinse the wash solution system if the instrument is to be
left standing for a longer time. Alternatively it can be used to rinse the wash

If the instrument is left to stand for 1 hr to 1/2 day, wash solution can be used, for
longer times use distilled water.

7.6 Instrument Disinfection

WARNING
ALL PARTS OF THE INSTRUMENT THAT COME INTO CONTACT WITH
POTENTIALLY INFECTIOUS MATERIAL MUST BE TREATED AS
POTENTIALLY INFECTIOUS AREAS. IT IS ADVISABLE TO WEAR
PROTECTIVE POWDER-FREE GLOVES WHEN PERFORMING THE
CLEANING PROCEDURE AND ALSO WHEN MAKING ADJUSTMENTS TO
THE INSTRUMENT.
It is very important that the instrument is thoroughly disinfected before it is
removed from the laboratory or any servicing is performed on it.
Before the instrument is returned to the distributor for servicing, it must be
disinfected and a disinfection certificate completed by the operating authority.
If a disinfection certificate is not supplied, the instrument may not be accepted by
the servicing center or it may be held by the customs authorities.

7.6.1 Disinfection Procedure

Caution
Before starting the disinfection procedure use a rinse procedure
(Rinse Day) with distilled / deionized water (lab quality) to flush the system.

If the laboratory has no specific disinfection procedure, the following procedure


should be used to disinfect the instrument.
The instrument should be disinfected using one of the following solutions:
Lysetol Manufacturer: Schülke & Mayr GmbH.
Aseptisol Manufacturer: Bode Chemie Hamburg
If neither of these solutions are available 70% ethanol should be used as an
alternative.

WARNING
THE DISINFECTION PROCEDURE SHOULD BE PERFORMED BY
AUTHORIZED TRAINED PERSONNEL IN A WELL VENTILATED ROOM
WEARING DISPOSABLE POWDER-FREE GLOVES AND PROTECTIVE
GLASSES AND CLOTHING.

Please note that the disinfectant can influence the performance of your
instrument if it comes into contact with the electronics!

The following procedure should be used to disinfect the instrument.


1. Wear protective powder-free gloves, protective glasses and protective
clothing.
2. Prepare an autoclaveable bag for all disposables used during the disinfection
procedure, label it with autoclave tape and put it in the autoclave or follow the
procedure defined in chapter 7.7.3 Disposal of Operating Material.
3. Prime the liquid system with disinfectant. Perform the Rinse - Night
procedure with disinfectant solution and stop the procedure after five hours.
4. Switch off the instrument and disconnect the instrument from the mains
power supply.
5. Disconnect the instrument from any accessories that are used for example:
Liquid Sensor System, computer, etc. Accessories that should be shipped
together with the instrument have to be included in the disinfection
procedure.
6. Carefully spray the disinfectant solution (or use a disposable soft tissue
paper towel soaked in the disinfectant) on all outer surfaces of the
instrument.
7. After a minimum contact time of 10 minutes, repeat step 6 of this procedure.
8. Wipe dry the outer surfaces of the instrument.
9. Pack the instrument and its accessories.
10. Disinfect your hands and clean them with a mild detergent.
11. Complete a disinfection certificate and attach it to the outside of the
box so that it is clearly visible. (See below for an example of the
disinfection certificate).
The following label MUST be completed by the operating authority and attached
to the top of the package in which the instrument is returned, before sending the
instrument to the servicing center for repair.

7.7 Disposal of Instrument


7.7.1 Introduction
This chapter gives instructions on how to lawfully dispose of waste material
accumulating in connection with the Washer 430.

Caution
Observe all federal, state and local environmental regulations.

7.7.2 Disposal of Packing Material


According to Directive 94/62/EC on packaging and packaging waste, the
manufacturer is responsible for the disposal of packing material.

Returning Packing Material


If you do not intend to keep the packing material for future use, e.g. for transport
and storage purposes:
Return the packaging of the product, spare parts and options via the field service
engineer to the manufacturer.

7.7.3 Disposal of Operating Material

WARNING
CHEMICAL AND BIOLOGICAL HAZARDS CAN BE ASSOCIATED WITH
THE WASTE MATERIAL (MICROPLATE) OF THE PROCESS RUN ON THE
WASHER 430.
TREAT THE USED MICROPLATE, WASTE BOTTLE, WASTE SINK ON
THE PLATE CARRIER, DISPOSABLES AND ALL SUBSTANCES USED,
IN ACCORDANCE WITH GOOD LABORATORY PRACTICE GUIDELINES.
INQUIRE ABOUT APPROPRIATE COLLECTING POINTS AND APPROVED
METHODS OF DISPOSAL IN YOUR COUNTRY, STATE OR REGION.

Please contact your local bioMérieux service representative before disposing of
the instrument.

Caution
Always disinfect the instrument before disposal.

Method of Disposal Contaminated Waste

DEPENDING ON THE APPLICATIONS, PARTS OF THE WASHER 430
MAY HAVE BEEN IN CONTACT WITH BIOHAZARDOUS MATERIAL.
• MAKE SURE TO TREAT THIS MATERIAL ACCORDING TO THE
APPLICABLE SAFETY STANDARDS AND REGULATIONS.
• ALWAYS DECONTAMINATE ALL PARTS BEFORE DISPOSAL.
