EPIGENETICS AND DNA REPAIR

• DNA damage triggers many

histones epigenetic modifications

• Those modifications direct the

recruitment of proteins involved in
DNA DAMAGE SENSING
AND REPAIR PROCESSES

The preventive modulation of histones acetylation through HDAC INHIBITORS

(Histone Deacetylase) Results in PHOTOPROTECTION
CONCEPT
EPIGENETICS… AT THE ROOT
• Epigenetic modifications are changes that alter the activity of a gene without
changing the nucleotide sequence. These phenomena are heritable at cellular level
but not at trans-generational level.

• The mutations of the histone proteins associated with the DNA are epigenetic
modifications and consist in covalently addition or removal of chemical moieties.

• Histones modifications can change within minutes, allowing rapid response to

external stimuli.

Me=methylation, Ac=acetylation, P=phosphorylation, Ub= ubiquitination

Histones acetylation and deacetylation is balanced by two enzymes, HAT

and HDAC. Therefore, HDAC inhibitors strongly influence gene expression
CONCEPT

HEALTHY SPROUTS AND RADISHES

TAPROOTS
Edible root rich in nutrients, polyphenols and
vitamins (ex.: carrot, turnip, beet)

It comprises three big families. Among them,

raphanus taproot belongs to the Brassicaceae
genus

Healthy brassicaceae plants are a main source

of glucosinolates (GLs),
sulfur-rich secondary metabolites

RAPHANUS RADISH SPROUTS ARE EXTREMELY

RICH IN GLUCOSINOLATES
CONCEPT
FROM GLUCOSINOLATE TO ISOTHIOCYANATE
GLUCOSINOLATES CONTRIBUTE TO PLANT DEFENSE AGAINST PESTS AND DISEASES

About 120 different glucosinolate molecules have been identified.

Different structure components:
• sulphonated oxime group
• Sulphur-linked glucose moiety
• variable alkyl side chain (R)

THE MYROSINASE
BIOTRANSFORMATION
Myrosinase is an enzyme naturally occurring in brassicaceae
that, in presence of water, cleaves off the glucosinolate’s
glucose group.
The released molecule quickly converts into
ISOTHIOCYANATE, nitrile or thiocyanate.

These are the effective active molecules that serve AS DEFENSE FOR THE PLANT AND
HAVE POSITIVE HEALTHY EFFECT against various pathologies and chronic diseases in humans
CONCEPT
AKOSKY PINK BEAUTY
ISOTHIOCYANATES
• SULFORAPHENE has being studied for its anticarcinogenic properties

• RAPHASATIN showed antimutagenic, chemopreventive, antifungal,

antibacterial and antioxidant activity

• SULFORAPHANE manifests a variety of properties in cell culture

systems and animal models, including antioxidative, anti-inflammatory,
antiangiogenic and neuroprotective activities

• SULFORAPHANE and RAPHASATIN are known inducers

of detoxification pathway as adaptive responses to environmental stressors

More recent studies underline the ability of

ISOTHIOCYANATES to influence EPIGENETIC ACTIVITY
CONCEPT

ROSE WINTER RADISH

• This radish variety is a distinct species among RAPHANUS genus

• It is an ancient species, evolved directly from the WILD RADISH.

Its characteristic appearance clearly distinguishes it from all other
modern, small and round radishes, commonly found in supermarkets

• It is very RESISTANT to ENVIRONMENTAL STRESS

• It is known as WINTER RADISH as it can withstand a hard frost

without damage

RADISHES
SPROUTS
Sprouts are a potent source of ACTIVE MOLECULES

During sprouting, a substantial increase of SECONDARY METABOLITES is observed

It is known that, in sprouts, free RADICAL SCAVENGING, ANTIOXIDANT PROPERTIES

and UV LIGHT PROTECTION are improved

SPROUTS ARE ONE OF THE

RICHEST SOURCES
OF SECONDARY METABOLITES

AKOSKY® PINK BEAUTY contains:

• Isothiocyanates: RAPHASATIN,
SULFORAPHENE AND SULFORAPHANE

• FLAVONOIDS (luteolin) and POLYPHENOLS

(coumaric acid, ferulic acid)

• various AMINO ACIDS (Proline, Valine, Cysteine,

Leucine)

• VITAMINES (Vitamin C and Biotin)

MECHANISM OF ACTION

DNA PROTECTION
Several DNA damages, caused by aging, exposure to chemicals and UV, drugs and diet, occur daily and some of these
can be critical for cell survival. EPIGENETIC MODIFICATIONS INFLUENCES THE DNA REPAIR.

AKOSKY® PINK BEAUTY modulates the nuclear chromatin conformation

WITH EPIGENETIC MECHANISMS, ALLOWING RAPID DNA SELF-REPAIR.
CLINICAL TEST

POST-IRRADIATION TREATMENT
20 male and female volunteers (age 20-65 years old) were UV-irradiated on their back on three skin sites using a solar simulator to
generate a local erythema (T24h).
24h post-irradiation (T0), one site was treated with a cream containing AKOSKY® PINK BEAUTY at 1%, one site with a placebo
formulation and one site was left untreated. Erythema and Trans Epidermal Water Loss (TEWL) measurements were taken at T -24h,
T0, T1h.

AKOSKY® PINK BEAUTY significantly REDUCES SKIN ERYTHEMA caused by UV

irradiation and gives a substantial SUPPORT TO THE SKIN BARRIER
