1.0 Purpose

As part of our continuous development of the Microflow product range, we have developed
improved calibration software and set up procedure for our SE and ABS Class II safety cabinets.
Good customer service and product safety are our number one priorities. The purpose of this
documented procedure is to briefly detail the method statement to identify the procedural
requirements for the set up, calibration and testing of airflow alarms on the “SE” & “ABS” range of
Biological Safety Cabinets maintained by the service personal of BIOQUELL (UK) Limited,

2.0 Scope
This documented procedure applies to all permanent, temporary and contract staff involved in the
activity of set up, calibration and testing of airflow alarms on the “SE” & “ABS” range of Biological
Safety Cabinets on site at the customer’s premises. The aim of this procedure is to demonstrate
that the service personal of BIOQUELL Limited are focused at providing customer satisfaction
throughout all stages of commissioning and routine maintenance.

3.0 Revision Details

present time there are no revision requirements.

5.0 Instructions

5.1.0 B.S.C Setup Calibration Introduction


This applies to Standard Class II cabinets, where there is a top box fan or remote exhaust fan use
software V2.44 for ducted (exhaust fan assisted units.). 100% exhaust cabinets and cabinets without
any exhaust extraction (Option A and B) should remain calibrated by the original V2.40 calibration
software for recirculating (non-exhaust fan assisted units). The new software V2.44 allows the main
cabinet fan to control the down flow level mid way between the high and low down flow set points, i.e.
if the cabinet is set up with high and low set points of 0.25 and 0.50 m/s, the cabinet will control at the
down flow velocity of 0.375 m/s, the inflow set point is utilized purely as an alarm.

5.1.1 BSC Initial Setup Values


If communication between the computer and controller is not made, click on “Connect” at the set
up screen. It is not unusual to see a dialog box saying “Controller not configured correctly, please
configure”. Disregard this message using the exhaust fan slider, manually set the fan to
approximately the values shown. Adjustment is made by dragging the slider next to “Main fan”
speed slider on the calibrate screen. Alternatively, for small increments, the bar either side of the
slider may be clicked, or for the smallest adjustment, either direction arrow may be clicked. Using
the main fan slider, manually set the main fan slider and exhaust fan slider to zero as shown in the
table below (For 220 to 240 V AC units only)

5.1.2 BS EN12469: 2000 Airflow requirements:


The Mean down flow velocity shall be between 0.25 and 0.50 m/s (49 to 98 ft/min). No individual
measurement shall differ from the mean by more than ± 20% when measured 100 mm above the
top edge of the working aperture. The inward velocity through the working aperture shall not be
less than 0.4m/s (79 ft min).

5.1.3 Airflow Alarms Set Point Requirements:


The Low Down flow alarm shall bet set to 0.25 m/s ± 0.02 m/s (49 ±4 ft/min)
The High Down flow alarm shall be set to 0.50 m/s ± 0.02 m/s (98 ±4 ft/min)

Re circulating (non-exhaust fan assisted units) V2.40 (BS EN12469: 2000)


The Low Exhaust alarm set point shall be set to 0.4 m/s ± 0.02 m/s (79 ±4 ft/min)

Ducted (exhaust fan assisted units.) V2.44 – Manufacturer’s specification


The Low Exhaust alarm set point shall be set to 0.5 m/s ± 0.02 m/s (98 ±4 ft/min)

5.1.4 Mean Down flow Velocity Determination:


Using a 100mm vane anemometer, carry out a survey of the down flow airflow’s and note all
readings and determine the average down flow velocity, this should be around 0.375 m/s ±0.02
m/s. Once the average down flow has been calculated the location of the point closest to the
average down flow should be used for all further down flow measurement for the following steps.

See table below for the number of readings to be taken to determine the average down flow
velocity the lateral positions shall be ¼ distance from the front and back of the work area:

5.1.5: Down Flow Calibration Procedure


The down flow calibration process may now be started. Ensure that the front screen is in the normal
operating position and with the exhaust fan slider set to zero

NOTE: The “Calibrate" process will instruct the operator to set the airflow via a (virtual) slider on
the screen to the 3 alarm velocities of: low down flow, high down flow, & low exhaust (representing
low inflow). These values will then be used by the set-up software to derive an operating condition
for the cabinet such that no alarms will be activated. When the data is written to the controller, the
values will be stored & used until the cabinet is next calibrated or adjusted. The operating
condition of the cabinet can be manually adjusted if desired.

5.1.6 Low Down flow Alarm setting:


The value of velocity the instruction specifies to set will be the value retrieved from the
configuration file. With the anemometer at the position defined in 5.1.4, adjust the fan to give the
value of 0.25 m/s (BS EN12469: 2000). Adjustment is made by dragging the slider next to “Main
fan” speed slider on the calibrate screen. Alternatively, for small increments, the bar either side of
the slider may be clicked, or for the smallest adjustment, either direction arrow may be clicked.
When the correct speed has been obtained as indicated on the anemometer, although the
software waits until sensor values are stable, it is advisable to observe the sensor values until the
numbers are steady before clicking “Continue”, note the sensor values, fan value and fan voltage
(This is achieved by using a multi-meter set to AC voltage and placing the test probes to
connector “PL9” pins 1 and 2. The connector is marked PL9 and numbered “1” and “6” on each
end.) Record the noted values on the Set-up record sheet.

NOTE: Check that the correct probe is connected to correct connector on the PCB by either
placing your hand over or a cover over the Probe inside the working area. Check that the correct
sensor reading moves significantly. These probes are connected to “PL2” which is the “Exhaust”
probe and “PL3” which is the “Down flow” probe. Should the wrong sensor reading move, reverse
connection on the PCB and exchange sleeves on the cables and then "Cancel" the calibration and
restart from “5.1.4”.

5.1.7 High Down flow alarm setting:


A new dialogue box will now appear, the value of velocity the instruction specifies to set will be the
value retrieved from the configuration file. With the anemometer at the position defined in 5.1.4,
adjust the fan to give the value of 0.50 m/s (BS EN12469: 2000). Adjust the fan to give the value
of 0.5 m/s. should the voltage to the fan exceed 195 V ac, it is recommended to NOT to set above
this figure. Dragging the slider next to Main fan speed title on the sub screen makes adjustment.
Alternatively, for small increments, the bar either side of the slider may be clicked, or for the
smallest adjustment, either direction arrow may be clicked. When the correct speed has been
obtained as indicated on the anemometer, although the software waits until sensor values are
stable; it is advisable to observe the sensor values until the numbers are steady before clicking
Continue, ”, note the sensor values, fan value and fan voltage (This is achieved by using a multi-
meter set to AC voltage and placing the test probes to connector “PL9” pins 1 and 2. The
connector is marked PL9 and numbered “1” and “6” on each end.) Record the noted values on
the Set-up record sheet.

Then click on the Continue button. A new dialog box will appear, “Please wait for Stabilization”.
Wait until this clears, if this fails to clear within 10 minutes, there is a problem with the flow sensor,
as it is not stabilizing.

5.1.8: Average Down Flow Sensor Evaluation


Note the down flow sensor values recorded in 5.1.6 & 5.1.7 add these two values together and
then divide the result by two and record this value.

5.1.9: Exhaust Calibration Procedure


The exhaust calibration process may now be started. Ensure that the front screen is in the normal
operating position and with the exhaust fan slider now set to 3250

5.1.10 Mean Inflow Velocity Determination:


Using a 100mm vane anemometer, carry out a survey of the inflow velocity and note all readings
and determine the average inflow velocity, this should be no less than 0.40 m/s. Once the average
inflow has been calculated the location of the point closest to the average inflow should be used
for all further inflow measurement.

5.1.11 Low Inflow alarm setting:


Note: Ducted Units
In environments that are proven to giving poor KI discus (containment) results, i.e. those with high
external turbulence, it may be acceptable to increase the inflow from 0.5 m/sec to circa 0.55 m/sec
to give an added factor of safety.

With the anemometer still at the average inflow point determined in 5.1.10, close the front screen
down onto the head as shown below

A new dialogue box will now appear, the value of velocity the instruction specifies to set will be the
value retrieved from the configuration file, the value of 0.40 m/s (79 ft/min) when using software
V2.40 & the value of 0.50 m/s (98 ft/min) when using software V2.44. With the anemometer at the
position defined in 5.1.10, adjust the fan to give 0.40 m/s (79 ft/min) when using software V2.40 by
dragging the slider next to Main Fan speed title on the screen makes the adjustment or Adjust the
fan to give 0.50 m/s (98 ft/min) when using software V2.44 by dragging the slider next to Exhaust
Fan speed title on the screen makes the adjustment.

When using software V2.44 use the slider next to Main Fan speed title on the screen to set the
down flow sensor value to the value determined in 5.1.8 you will have to balance the internal fan
to maintain this value while adjusting the exhaust fan to set the required inflow value.

Alternatively, for small increments, the bar either side of the slider may be clicked, or for the
smallest adjustment, either direction arrow may be clicked. When the correct speed has been
obtained as indicated on the anemometer, although the software waits until sensor values are
stable, it is advisable to observe the sensor values until the numbers are steady before clicking
Continue. Note the sensor values, fan value and fan voltage (This is achieved by using a
multimeter set to AC voltage and placing the test probes to connector “PL9” pins 1 and 2 - No
remote Exhaust fan fitted or “PL9” pins 5 and 6 - With remote Exhaust fan fitted. The connector is
marked PL9 and numbered “1” and “6” on each end.) Record the values noted on the Set-up
record sheet,

Then click on the Continue button. A new dialog box will appear, “Please wait for Stabilization”,
Wait until this clears, if this fails to clear within 10 minutes, there is a problem with the flow sensor,
as it is not stabilizing – seek further technical advice if this problem continues.

After the 3 alarm points have been set, a dialogue box will offer the choice either to Continue
which will store the data to the controller's memory, or to Cancel the calibration process. If the
latter is selected then flow settings & alarm values in the controller's memory will remain
unchanged. The cabinet will now operate at the borderline of safety, as the exhaust fan is running
at the inflow alarm level

5.1.12 End of Calibration:


Clicking on Continue, after a short period, a new dialogue box will appear “ Calibration Complete”.
Press “Okay”. Should the message appear “ Write failed, try again” with two buttons “Yes” and
“No”. This message does not necessary mean that it has actually failed this is an anomaly in the
software the system is looking for all parameters to be changed and if all parameters are not
changed it thinks it has failed to write the parameter this is usually connected to the service date
routine which is not always changed when recalibrating the alarm set points. Press “NO”.
Calibration is now complete. The exhaust fan speed can now be increased (+200 for all units up to
1.5 meter & +300 for 1.8 meter units) to give a suitable inflow safety margin for containment above
the low inflow alarm this value then can be “Written to the Controller”.

The set up of the cabinet is now complete, now “Disconnect” the Set-up program from the unit
under test and allow the unit to settle. Alarms may sound during this period; this is not unusual as
the unit is settling. If alarms continuously sound, then the unit has NOT been set correctly and it
will require to be re-calibrated again. Return back to the beginning of “5.1.1” and start again. If unit
does settle and no alarms sounds, leave for 5 to 10 minutes switch unit off then on then leave for
a further 10 minutes and then take final readings of all airflows, down flow and exhaust and record
these on the set-up record sheet and re-connect the set-up program and record final sensor
readings, Main & Exhaust fan slider value plus also note fan voltage. (This is achieved by using a
multimeter set to AC voltage and placing the test probes to connector “PL9” pins 1 and 2. The
connector is marked PL9 and numbered “1” and “6” on each end.) Record these on the set-up
sheet.

5.1.13 Additional airflow set up functions exhaust fan set-up: (SE Units Only)
NOTE: this only applies to exhaust adapter that has a fan fitted. If a ducted exhaust system or
secondary exhaust filter is fitted to the cabinet, then inevitably the external fan will require setting
to an appropriate speed. This speed is typically 3500 on the exhaust fan slider. This fan will
operate at a constant speed irrespective of changes in airflow.

The speed is set by use of the exhaust fan slider on the set-up screen. To set-up the exhaust fan
proceed as follows:

Set-up the cabinet with the exhaust system removed as described in “5.1.1” onwards
Connect a Digital manometer, which has a resolution that is capable of reading 1 Pa to sample
port 'B' located on the side of the cabinet. Zero the Manometer before proceeding. Establish a
communication link between computer & controller by clicking on Connect. Note the flow sensor
values displayed, check that these are similar to figures obtained in “5.18” & “5.1.10” to within ± 2
units. If the figures are not within ± 2, disconnect and allow the unit to settle and then re-connect.
If the unit fails to settle then the unit will require re-calibrating. Fit the exhaust components to the
cabinet while it is still operating and the unit is still “Connect” to the PC. Adjust the exhaust fan
slider, until the previously recorded flow sensor values are re-established or the manometer reads
zero, or slightly negative pressure, 0 Pa to –5 Pa. Observe the measuring device (sensor reading
or manometer) for a couple of minutes to ensure the condition are stable. Click on Write, if no
other operation is to be performed, then Disconnect. After Disconnecting the Set-up program from
the unit under test, allow the unit to settle for at least 5 to 10 minutes. Alarms may sound during
this period; this is not unusual as the unit is settling. If alarms continuously sound, then the unit
has NOT been set correctly and it will require to be re-calibrated again. Return back to the
beginning of “5.1.1” and start again. If unit does settle and no alarm sounds, after 10 minutes, re-
“Connect” the set-up program and record final sensor readings, final exhaust fan slider & main fan
slider number plus also note fans voltage. (This is achieved by using a multimeter set to AC
voltage and placing the test probes to connector “PL9” pins 1 and 2 for the Main Fan and pins 5
and 6 for the Exhaust fan.). Record these on the set-up sheet. Setting up of the exhaust fan is
now complete & the cable linking computer & controller can be removed.

5.2.0
Place the 100mm vane anemometer in the spot position nearest to the location of the mean down
flow reading obtained from section “5.1.4”

5.2.1: Low Down flow alarm test procedure:


To check the Low Down flow Alarm and the Control system, on completion of “Calibration” and
recording the final airflow pattern, place the 100mm vane anemometer in a position close to the
mean airflow as determined in section “5.1.4” Use sheets of paper, place them one by one over
the intake grill, and observe the Anemometer display. After a couple of sheets, the airflow might
fall slightly but the control system should compensate for this. Place some more sheets and the
airflow will fall further and this time, the control system will be unable to compensate. Eventually
the airflow will fall below the Low Alarm Set point and the LCD display shall display “Low Down
flow” and may display “Low Exhaust” or their language equivalent. The alarm shall sound.
Remove all the sheets and allow the system to restore itself back to “Safe”. Please note that it
may take some time to recover due to the severity of this test.

5.2.2: High Down flow alarm test procedure:


To check the “High Down flow” alarm. Reconnect the BSC Set-up program to the unit being tested
& note original fan slider value settings. Slide the Main Fan speed controller slider to maximum
and immediately disconnect the BSC Set-up program, the unit fan should be running at full speed
and the alarm should sound along with the message on the LCD display “High Down flow” or the
language equivalent. On completion, re-connect the Set-up program and reset the Main Fan
speed control slider, to its original position and the “Disconnect” the Set-up program. Allow the unit
to settle.

5.2.3: Low Inflow alarm test procedure:


If re-circulating (non-exhaust fan assisted units) B.S.C proceed to “5.2.4”: only. If
a Ducted (exhaust fan assisted units) B.S.C proceed to “5.2.5”:

5.2.4:
Proceed to block the front aperture and if required also the rear liner grills as well of the BSC until
the airflow alarm sounds, note the following readings at this point record these on the set-up
sheet.

* Reading can be taken from exhaust filter outlet when unit is not ducted if required.

5.2.5:
Place the 100mm vane anemometer in the spot position nearest to the mean inflow reading.

Disconnect the exhaust fan electrical supply and note the actual indicated low inflow velocity
reading when the low inflow alarm sounds record these on the set-up sheet.

* Reading can only be taken from front aperture when unit is ducted.
** 0,40m/s on non-exhaust fan assisted units and 0,50m/s on exhaust fan-assisted units

6.0 Responsibilities
I. The project / area manager employed by BIOQUELL (UK) Limited. Is responsible for
ensuring all permanent, temporary and contract staff involved in the activity of set up,
calibration and testing of airflow alarms on the “SE” & “ABS” range of Biological Safety
Cabinets do observe compliance with all documented procedures of BIOQUELL (UK)
Limited

II. The individual service engineer or service department team member is responsible for the
tasks required of them as agreed by the project manager/area manager

III. The service management is responsible for the general administration of the activity of
set up, calibration and testing of airflow alarms on the “SE” & “ABS” range of Biological
Safety Cabinets of BIOQUELL (UK) Limited.

IV. The individuals line manager is responsible for ensuring that his staff are trained to carry
out the tasks that they are employed for in connection with the business activities of
BIOQUELL (UK) Limited.

V. The line manager is responsible for sanctioning training & ensuring accurate details of
training are maintained in the individual’s personal file.
