Precautions during Nucleic Acid Extraction for NGS Work

Precautions to be taken during DNA isolation:

1. Make sure that the vials and tips are DNase free.
2. Avoid vortexing during DNA extraction.
3. Make sure that the extracted DNA is treated with RNase to remove the RNA.
4. It is good to use the columns to purify the DNA. The columns will remove the salts
presents in the DNA.
5. Ship the DNA samples on sufficient Dry ice.

Precautions to be taken during RNA isolation:

Handling RNA asks for special precautions, RNases are everywhere!

1. Always wear gloves when handling RNA and make sure that your bench, pipets, tips
and solutions are RNase-free!
2. Use liquid Nitrogen to grind the tissues while you extract RNA.
3. Freeze and thaw your RNA samples as little as possible. It is better to keep samples
on ice for a few hours then to re-freeze them in between.
4. Always snap freeze RNA samples in liquid nitrogen and keep them at –80° C.
5. Ship the RNA samples on sufficient dry ice.
