5.1 INTRODUCTION TO POLARIMETRY ............................................................................................................................................................................ 19

5.1.1 Effects on Polarimetry Measurings ........................................................................................................................................................ 23

5.1 Introduction to Polarimetry

Polarimeters utilize so called polarized light. The human eye can not detect, whether light is polarized or not, and therefore
it is somewhat difficult to understand polarimetry. Two polarizing filters, which look like simple grey filters at first sight,
show an interesting effect if you hold them one after the other against a light source. In that case, no light passes through
the combination at one special position of the two filters to each other, while it gets bright again if one of the filters is turned
by 90°. Physicists say, that there is only one plane of polarization that can pass through a polarizing filter. If the planes of
the two filters are perpendicular to each other, the light from the first one cannot pass the second one, and that’s why it
gets dark. Some materials like quartz or sucrose solutions turn the plane of polarization. They are called optically active.
What polarimeters do is detecting the change in the plane of polarization induced by optically active samples. In the typical
design of a polarimeter, two polarizing filters are in a position where no light passes through. If an optically active sample
is brought between those filters, light passes again. The dark position can be found again by turning one of the filters - and
thus we know the rotation induced by the sample.

19
S+H VariPol Theory VariPol User Manual – Version 1 / Revision 1

A very simple polarimeter consists of at least these parts:

Figure 1: Polarimeter build-up

In practice this simple design will not function well, as it is impossible to find the darkest position with the desired accuracy.
That’s why Faraday modulators are used in our automatic polarimeters. Faraday modulators make use of the effect, that
some materials get optically active in a magnetic field. If a rod of such a material is brought into a coil, which is connected
to alternating currency, the rod will have an alternating optical activity. Such a modulator, positioned in between two
polarizing filters, will thus induce a periodically changing intensity of the light passing through. And with this signal the final
position of the polarizing filter can be found easily. The method of Faraday modulation is very accurate and non sensitive to
errors.

20
VariPol User Manual – Version 1 / Revision 1 Theory S+H VariPol

In case of diluted samples, which are to be measured in the instrument, the measured angle depends on

sample

concentration

length of the tube

temperature

colour of light (wavelength)

The appropriate formula is named after the French physicist Jean B. Biot, who lived in the 19 th century. BIOT's law states:

c with
 
20
D 10000
l

c : concentration (in g / 100 cm3 solution)

: rotation (in angular degree)

 20D : specific rotation

l : length of the tube (in mm)

21
S+H VariPol Theory VariPol User Manual – Version 1 / Revision 1

The specific rotation depends on the temperature and the wavelength of the light. For some samples this value depends
on the concentration also. Please be aware, that the concentration is given in g / 100 cm³ or g / 100ml, but not in g / 100 g.

Let’s have a look at an example. The specific rotation of aqueous solutions of sucrose at 20.0 °C and a wavelength of 589.44
nm (that is the centre of the two yellow lines of the sodium lamp in vacuo) is given by the ICUMSA (the International
Commission for Uniform Methods of Sugar Analysis) as

 20D = 66.588 ± 0.002

So, if 26.000 g of sucrose are dissolved in pure water to 100 cm³, the rotation in a tube of 200 mm will be

 = 34.626° ± 0.001°

as you can verify by putting in the numbers into BIOT's formula above. This solution is called the Normal Sucrose Solution
and its rotation is 100.00 °Z (sugar degrees). The International Sugar Scale (ISS) is linearly divided, e.g. a rotation of 17.313°
equals to a reading of 50.00 °Z.

22
VariPol User Manual – Version 1 / Revision 1 Theory S+H VariPol

Effects on Polarimetry Measurings

In the following there are shown some effects that could influence polarimetric measurements. Based on these effects
some false measurements could appear after the instrument is delivered. In consequence of that, please calibrate the
instrument before first use.

We recommend to calibrate a polarimeter with a certified quartz plate every time before you use it.

Wavelength Effects

Let's have a closer look at the wavelength effect on the optical activity of sucrose solutions. There are four basic wavelengths
in use in polarimeters of the sugar industry. For these wavelengths the following table gives the specific rotation  20
D
and
the rotation  of the above defined Normal Sucrose Solution in a tube of 200 mm:

Description Wavelength  20D 

mercury green 546.23 nm 78.4178 40.777°

sodium yellow 589.44 nm 66.5885 34.626°

HeNe laser 632.99 nm 57.2144 29.751°

NIR 882.60 nm 28.5306 14.836°

23
S+H VariPol Theory VariPol User Manual – Version 1 / Revision 1

For further details on the International Sugar Scale, ISS, refer to "Specification and Standard SPS-1 (1998): Polarimetry and
the International Sugar Scale" in the ICUMSA Methods Book.
In this example you can see influence of the wavelength of the light on the rotation, which is called Optical Rotation
Dispersion (ORD). A change of 0.03 nm will lead to a change in the rotation of about 0.01%. This is a very high influence!
That’s why light sources with a small bandwidth have to be used in polarimetry. The typical choice is a halogen lamp plus
an interference filter. The Purity Analyzer is equipped with high power LED’s. To filter out the side bands of their spectrum,
the LED’s are combined with interference filters.

Temperature Effects

Another influence is that of the temperature. The reading of Quartz plates increases with temperature:

Reading(T) = Reading(20.0°C) (1.0 + 0.000144 (T - 20.0))

A plate, which shows 40.000° at 20 °C, will thus show 40.006° at 21 °C and 40.029° at 25 °C.

24
VariPol User Manual – Version 1 / Revision 1 Theory S+H VariPol

In difference to that, the rotation of solutions will decrease with temperature.

For sucrose solutions for example the effect is

Correct Reading (  ) = Reading (  )  (1.0 - 0.000471  (T - 20.0))

39.906° = 40.000° (1.0 - 0.000471 (25°C - 20°C))

A solution, which shows 40.000° Reading(  ), will thus show 39.981° at 21 °C and 39.906° at 25 °C.
Please be aware, that there are three temperature effects on diluted samples. First of all, the flask, which is used to fill up
to 100 cm³, is calibrated for 20 °C. Also the length of the tube is valid for 20 °C. And last but not least: the optical activity of
the sample is influenced by the temperature. The formula given above for sucrose solutions is only taking the last effect
into account.

Tube Length

The rotation is proportional to the length of the tube. Usually , in Pharma applications 100 mm tubes are used, in food
applications 200 mm tubes.

It is possible to enter a temperature coefficient which compensates the change of the tube length with temperature.

28
VariPol User Manual – Version 1 / Revision 1 VariPol S+H VariPol

All samples have to be filled in a sample tube and this tube has to be placed in the sample room before measurement.
SuH offers a wide range of tubes for the use with the VariPol.

Variations are possible for:

- product touching material
- product volume
- filling mechanism
- tube length
For the VariPol we designed the VariTubes. These are metal tubes optimized for the best temperature control performance,
a standard low sample volume and Luer-connection for easy filling and fast work flow.
When they are connected via cable to the instrument, they automatically identify themselves and transmit the sample
temperature.
Other tube models are also possible.

29
S+H VariPol VariPol VariPol User Manual – Version 1 / Revision 1

Before filling a tube, confirm that the volume and product touching materials are suitable for your application.

Before using a tube, please confirm if there is no damage to the tube. You can do so by zeroing the instrument. Then
measure the empty tube in three different positions (in relations with its rotational axis). If the values are all zero (with
respect to the repeatability of the instrument) then you have a good indication of a properly functioning sample tube.

If the values differ, please contact your local SuH representative.

If you fill the tube with a new sample or use hand filling, make sure that the tube is dry and clean.

Before filling the sample tube, check:

 the glass windows of the sample cell are installed
 the hoses are connected correctly
 the hose connections are tight
 the waste hose leads into the waste container
 the waste container’s volume is large enough for the number of samples
(Some points may not be applicable depending on your type of tube.)

30
VariPol User Manual – Version 1 / Revision 1 VariPol S+H VariPol

If you refill a sample using the flow through tube, make sure that you passed enough new sample to replace the old sample.

Check that no air bubbles have formed in the light path.

Check that no solid particles float in the sample that could interfere with the light path.

To clean the tubes, open the cover glass caps on both sides.
Be careful: once unscrewed, the cover glasses themselves come lose.

Rinse the tubes thoroughly with pure water.

Follow this by a quick rinse with Ethanol or similar pure alcohol.
To finish, dry the tube with filtered pressurized air.

Rinse the cover glasses also with water and alcohol and dry them with a soft paper tissue.
Make sure that all visible residues have been removed.

31
S+H VariPol VariPol VariPol User Manual – Version 1 / Revision 1

The VariPol is equipped with integrated temperature control. To fully utilize this function, you have to use the VariTube
sample tubes. Further, you have to connect the cable delivered with the tube to the tube and to the designated slot at the
back of the instrument.

To change or enable the temperature control please see the chapter 9.4.5 “Temperature Widget”.

Please note that if the temperature control is enabled (which is the standard configuration) and the sample tube not
connected to the instrument, then the instrument will use the temperature of the sample room as a reference. In this case,
there can be a deviation between the real temperature of the sample and the measured one.
Therefore, we advise that if no temperature sensor is connected to the instrument, the temperature control should be
disabled to allow for more reliable results.

32
VariPol User Manual – Version 1 / Revision 1 VariPol S+H VariPol

We advise to set the instrument before each measurement or each batch measurement.

After turning on the instrument, wait at least 15 minutes for the optical system to stabilize.

To set the instrument zero, make sure that the sample room and sample tube are clean and free of any sample residues.
Place a sample tube filled with pure water in the sample room.
Close the sample room cover.
Then execute the “Setting Zero” by pushing the button on the display.

33
S+H VariPol VariPol VariPol User Manual – Version 1 / Revision 1

We advise that a calibration should only be executed by a trained technician, certified by SuH.
To calibrate the instrument, you need a SuH manufactured quartz control plate and a hexagon wrench key size M2.

Make sure that the test certificate of the quartz plate is not expired.
Set the instrument zero with an empty and closed sample room.
Insert the quartz plate and connect it to the temperature control.

Perform a measurement and compare the result to the quartz plate certificate.
If the result is within the accepted tolerances, no action is required.
If the values differ from said tolerances, then the instrument has to be mechanically adjusted.

To do so, insert the wrench key into the calibration slots at the left side of the instrument. To calibrate the wavelength
589nm use the lower slot for any additional the upper slot. Make sure the wrench key enters into the screw head.

If the values were too low, turn the key slightly clockwise. If they were too high, turn the key slightly counter clockwise.
Execute measurements after each adjustment.
Continue with small adjustments until the measurement values show the specified quartz plate value.
Then remove the wrench key and seal the calibration slot with SuH sealing tape to avoid unauthorized access.

34
VariPol User Manual – Version 1 / Revision 1 VariPol S+H VariPol

To execute a measurement, at first select the appropriate method. The instrument comes with a selection of pre-configured
methods. In addition, you can design methods as per your specifications.
For more information about Methods, see chapter 9.3. “What is a Method?”

In either method, the instrument will measure the optical rotation of the sample. Further, the instrument can calculate the
concentration or the specific optical rotation, given that the user provides the necessary inputs.

For more details on how to start a Simple Measurement, see chapter 9.6.3.4. “Simple Measurement”.

Aquisys 3 is the software that controls the instruments of the VariFamily and makes the measurement results visible and
sharable. Thanks to the intuitive operation, it guides the user through the measurements – right from the start through
individual configurations and settings until showing results as well as saving them and printing reports.

The Aquisys 3 software offers almost unlimited freedom of configuration. To mention but a few, you can define the layout
of the display elements (Widgets). You determine sequences of measurement routines and allocate them to a button in the
task bar.

To get an overview of the structure of Aquisys 3, have a look at the following picture with description.

S+H VariPol Aquisys 3 VariPol User Manual – Version 1 / Revision 1

This section describes the content and functionality of Methods in Aquisys 3.

In the context of Aquisys 3, a Method is a collection of settings to perform either a single measurement or batch
measurements in a pre-defined way, e.g. from zeroing the device to temperature control settings and starting the
measurement, saving results up to printing reports and more.
A Method reflects a systematic approach, designed to eliminate bias thanks to clearly defined steps. It is a systematic
process of achieving certain ends with accuracy and efficiency, usually in an ordered sequence of fixed steps.
Aquisys 3 comes with pre-installed methods, depending on your type of instrument. There is always a standard method
factory set (different for each type of instrument, e.g. a standard method for a polarimeter or a refractometer).
In addition, methods can be copied and modified or freely created from the scratch.
We strongly recommend to copy a pre-installed method and adjust it to your individual needs. See chapter 1.3.2 Create a
Method for more detailed information.

To give you a first insight, the Standard Method of the Refractometer is described as an example:

In general, most measurements are made at a standard temperature of 20°C.

Set Zero Button

By tapping/clicking on this button, the instrument will be set to zero (reset) – a confirmation dialog with instructions to
clean the sample chamber and fill in distilled water will appear.
Alternatively, you can skip this step by confirming using the “Skip step” button.
To assure accurate results, the instrument should be zeroed often. We recommend to activate this procedure before
every measurement.

Print Simple Report

Aquisys 3 offers great flexibility in printing by using the standardized CUPS technology.
CUPS is a modular and open-source printing system that uses the Internet Printing Protocol (IPP) as the basis for managing
printers, print requests, and print queues. CUPS supports network printer browsing and PostScript Printer Description-
based printing options. CUPS also provides a common printing interface across a local network.

To print a report, tap/click on the [Print Report] button and select if you want to print on your Vari device or a local printer.

Please note that the [Simple Print] and [Simple Report] functions only access and consequently display data of the last 50
measurements.
Further, the underlying data storage will be reset when you:
- use a sequence that refreshes the short term data storage
- restart the instrument
- change the method
- change instrument configurations
- access the instrument via a new device
In this case, the data will be available on your current platform but not on the new device.
- install an update

58
VariPol User Manual – Version 1 / Revision 1 Aquisys 3 S+H VariPol

We strongly recommend to work on the basis of an existing method and copy it - for adjusting it to your individual needs.
You will just make modifications instead of starting from the scratch. This way, you will reach precise results with your new
individual method in a fraction of time compared to setting up a completely new method.

For the majority of applications and especially in the on-site lab starting phase of the instrument, the Standard method
seems to be the most suitable method to copy and adjust to your individual needs. If you have already a variety of specific
methods created in your lab, one of them may be your first choice to copy.

To copy a method, do the following:

1. Tap on the MENU button and go to CONFIGURATION.
2. Choose Methods. A list of all methods available on the instrument is shown.
3. Tap or click on the [COPY] button.
4. Mark the method you want to copy, e.g. Standard. Confirm by tapping on the CONFIRM button.
5. Type the new name for the new Method into the text field and confirm.
6. Confirm with the [Confirm] button.

For an illustration of these steps, see next page.

68
VariPol User Manual – Version 1 / Revision 1 Aquisys 3 S+H VariPol

In Aquisys 3, a Widget is a control element in the user interface that visualises the measurement data and allows small
configurations. A widget helps you to easily monitor both the general instrument behavior as well as the status, settings
and results of the measurements.

A widget is the way to display a (measuring) value or information in a way that you want. You can define its size, content
and how to interact with it.

The layout of widgets on the Home screen is always associated to the (activated) Method, i.e. for different methods, your
Home screen display will have a specific layout if you customized it to your individual requirements.

This section describes the nature of widgets and how to install common and often used functions as widgets.

In general there are three types of widgets:

Traffic Light Widget

Live Data Widget

Temperature Widget

For each widget, you can individually define its size, content and how to interact with it.
69
S+H VariPol Aquisys 3 VariPol User Manual – Version 1 / Revision 1

Note: You can only add a widget to self-created methods and not to pre-installed methods. To save time and efforts, we
recommend to copy an existing method and add widgets to it or delete widgets (as described in the following paragraphs).

Fig.: Example of Widgets of the Standard Method placed on the Home Screen (VariRef)

70
VariPol User Manual – Version 1 / Revision 1 Aquisys 3 S+H VariPol

In Aquisys 3, there is a 3x3 grid, so that you have 9 rectangular spaces for placing widgets on the display.

You have up to 5 pages with 9 spaces for widgets each.

To control the appearance of a widget, you have several layout options – horizontally and vertically.

For detailed instructions on how to place widgets on the Home screen, refer to chapter Placement of Widgets

71
S+H VariPol Aquisys 3 VariPol User Manual – Version 1 / Revision 1

In general there are three types of widgets:

 Traffic Light Widget
 Live Data Widget
 Temperature Widget

Apart from the General category widgets, there are instrument specific widgets depending on the type and configuration
of your instrument.

The menu will guide you through the widget creation process.

72
VariPol User Manual – Version 1 / Revision 1 Aquisys 3 S+H VariPol

A Traffic Light Widget is first choice if you want to get instant visual information by red/green color display if a certain value
or status of the instrument is within defined thresholds.

The color settings of the Traffic Light Widget are pre-defined by Aquisys 3, just as known from a traffic light:

Green: Value is within the thresholds you defined for your widget

Red: Value is outside the thresholds you defined for your widget

Please note that only an admin user can install updates.

If it is required to install an update, please make sure you have a stable internet connection or the update file already on a
USB stick connected with the instrument.
The instrument will then automatically recognize if there is a new update ready for installation.
It is possible that the instrument shows available updates but you are not able to install them. In this case, there are two
possibilities:
1. You do not have administrator rights on the instrument.
2. The Update is not included in the package you purchased.

Network

Two or more computers connected to each other with hardware (network adapters) and networking software to
communicate and exchange data.

Browser

The "Browser" is a computer program that allows you to view web pages. There are several browsers. On the computer
they are usually there from the beginning, for example the Internet Explorer (Windows) or the Safari (Mac). Tablet PCs and
smartphones also have their own browser, which you do not have to download yourself.

For almost all mentioned devices you get free Internet browsers: for example Firefox, Opera and Chrome.

161
S+H VariPol Useful Information VariPol User Manual – Version 1 / Revision 1

IP-Address

An IP address is assigned to every computer on an Ethernet network. Like the street address for your home, an IP address
identifies network computers. It helps traffic flow between computers because each one has its own IP address.

An IP address is formatted as a series of four values separated by periods: 192.168.0.1

Each value ranges from 0 through 255. Each period is just a period.

For your home or office network, the IP address is most likely assigned by the router, using something called DHCP. The
router keeps track of every computer’s NIC and maps an IP address to the NIC every time the computer joins the network.
In the end, all computers have a unique IP address, and the world is safe for local networking.

The IP address assigned to your PC on a network is a local address. Similar IP addresses are used on the Internet to identify
domains and other resources. Those are Internet IP addresses, separate from your local address.

The router is assigned an IP address by your Internet service provider (ISP). That IP address is an Internet IP address. It’s
shared by all PCs on your network. Local IP addresses start with 192.168 and 10.0.

The IP addresses discussed here are IPv4 addresses. Because the number of unique IP addresses is limited, a second
standard, IPv6, has been established. The IPv6 standard allows for many more addresses, which will help accommodate
future growth of the Internet.

162
VariPol User Manual – Version 1 / Revision 1 Useful Information S+H VariPol

If the router doesn’t assign an IP address, one must be configured manually. No two computers on the network can have
the same IP address.

IP is often prefixed by the acronym TCP, as in TCP/IP. The TCP part stands for Transfer Control Protocol: It’s simply a set of
rules for transmitting information on a network. Technically, TCP/IP refers to the methods and engineering as opposed to a
specific address or value.

TCP/IP

(English abbreviation: Transmission Control Protocol / Internet Protocol) This is a network protocol that controls the
communication of a computer with the Internet. The protocol divides the information that is sent to the Internet into smaller
packages and provides them with certain markings. This allows the packets to reach their destination and the TCP / IP
protocol can identify them there.

DHCP

A number that identifies a computer’s location on the Internet. On a modern Ethernet network, with a robust router as
overseer, IP addresses are assigned to each PC as it joins the network. The process, which is automatic, is referred to as
Dynamic Host Configuration Protocol, or DHCP.

S+H VariPol Useful Information VariPol User Manual – Version 1 / Revision 1

There’s nothing to set up: Your router’s configuration program most likely contains a page that directs the router to use
DHCP to set IP addresses. You can turn off the option, but there’s no reason to do so.

There are two parts to DHCP: the client and the host. The host exists on your router, which assigns IP addresses. The client
is on your computer. It asks the host for an IP addresses and then assigns that address to the network connection.

When your computer isn’t connected to a network, its DHCP client program assigns a random IP address to the computer.
That IP address is useless because the PC isn’t connected to a network. But the random IP address confuses some users
because it appears as though the computer has a legitimate IP address. It does not.

The best way to test a network connection is to ping the router. You can configure the router to specify which range of IP
addresses to use. Again, you have no reason to change the DHCP settings for a small, home or office network.

LAN/Ethernet

Multiple computers connected as a network in one general location. A "Local Area Network" is a local area network where
computers are connected. A home network in which multiple computers access each other to an Internet connection is
usually a LAN. A connection can be made by cable or wireless (WLAN)

Server

A computer that provides services for other computers (called workstations or clients) on a network. Also called a host.

17.2 Slow Reactivity of Display

Please check if the transport protection film has been removed from the screen.

17.3 Problems with Connection to Network

If you can’t connect to the instrument with a secondary device in your own network, please check:

1. Does the instrument show an IP address?

a) If so, check the correct connection of the Ethernet cable at the instrument and the socket.
b) If not, confirm with your system administrator if the network requires additional steps to authorize
new IP addresses or even automatically provides IP addresses to new instruments.

2. If so, check with the system administrator if the instrument is in the same sub network of your local network and
no additional restrictions are in place.

3. If those steps did not solve the issue, please contact your local SuH representative.
