Test title: Liver Function Tests
The purpose of the experiment:
The quantitative determination ALP
of Alkaline Phosphatase Total Activity in solution
The basis of the test: Colorimetric determination of the ALP activity as follows:
p-Nitrophenyl phosphate (Reagent) p-Ntrophenoxide + Pi
[Test/Standard sample]
The results are based on the total absorbance per unit of time.
Test procedure:
ALP [samples] (20 µL) + pNPP [reagent] (1000 µL) <--> pNP [Dyed sample]
Mix and insert the cuvette into the spectrophotometer at 405 nm against air blank.
Read inidial absorbance after 1 minute and record absorbances at 1 minute intervals thereafter for 3
minutes.
Calculate the average absorbance per minute and multipy by factor 2.757 for results in IU/L.
Test result:
Ct= 606.5 IU/L
