RAPID SLIDE AGGLUTINATION TEST

INSTRUCTIONS:

I. MATERIALS:
1. Illustrate all the materials and reagents used in the Test. DOWNLOADED
PHOTOS are NOT ALLOWED.

2. Digital drawings are accepted. Make sure to label your drawings and color
appropriately the reagents.
II. PROCEDURE:
1. Illustrate steps 1 -3 in the procedure showing the Reaction Slide with the
doubling dilution of the patient’s serum. Indicate the volume of serum sample
that should be added to each of the reaction circle. Please include the Negative
Control.

2. Illustrate step 4 in the procedure.


III. Study Questions:
1. Differentiate Rapid Slide agglutination from the Tube agglutination Test as to
the sensitivity of the test.
The tube agglutination test, is more sensitive than that of the rapid slide
agglutination test due to the longer time the antigen and antibody can react
compared to the rapid slide agglutination test. On the other than, the rapid slide
agglutination test is more specific as not it uses Salmonella typhi as its antigen
source while the tube agglutination test uses Salmonella spp.

2. Give the temperature of reactivity and time of incubation of this test.


a. Temperature of Reactivity: Room Temperature
b. Time of Incubation: 1 minute
3. What should be the titer for the H and O antibodies that indicate active
typhoid fever?
a. Anti-H/ HAb: >80
b. Anti-O/ OAb: >50
4. Give the Genus and species of the bacterium associated with this test.
a. Salmonella typhi
IV. Reading of Result/Observation:
1. Draw, Read, Give the Titer of the H and O antibodies and Interpret the result
assigned to your group.

“O” Titer – 20
“H” Titer – 320
Interpretation: The patient might be a carrier, had a previous typhoid infection or
previous immunization.

VI. Conclusion
In this exercise we did the second method of Bacterial Agglutination test called “Rapid Slide
Agglutination test”. Here, the H and O antibody titers were discussed, determined, and
interpreted. This exercised explained the different materials and procedures needed for
proper interpretation. This exercise also discussed and provided to us the possible errors
that can affect interpretation and results.
