Introduction

Polyherbal formulations have several advantages, including:

stimulating physiological functions.
7-Nephroprotective effect: Polyherbal formulations can preserve renal damage due to their
antidiabetic and antioxidant actions.
Factor affecting polyherbal formulation:
1-Imperfect preparation Selection of the herbs should be done carefully based
on the required synergistic effect required to be produced.
2-Drug adulteration Adulteration is the practice of substituting the original crude
drug with a inferior property substance which sows less or spoiled therapeutic
effect
3-Faulty collection Some errors are made in herbal adulteration as a result of
the negligence displayed by collectors and suppliers of herbal products. It is
important to gather the appropriate sections of the genuine plant, while
refraining from collecting the less valuable parts.
4-Substiution with exhausted drugs Substitution with exhausted drugs: In this
type, the same drug is admixed but does not have any therapeutically active
constituents because these are already extracted out. This practice is more
common in volatile essential oils that contain drugs such as fennel, clove,
coriander, and caraway.

Advantages of polyherbal formulation:


While herbal medicines are widely used and considered safe, they are often used
in combination, sourced from various plants that may have their own species,
growing conditions, and biologically active components.
The main advantage of botanicals, compared to conventional single-component
drugs, lies in the presence of multiple active compounds. These compounds can
work synergistically, producing a potentiating effect that cannot be achieved by a
single compound alone.
Polyherbal formulations contain pharmacological agents derived from plants.
These agents can exert synergistic, potentiating, agonistic, and antagonistic
actions due to their associated diverse active principles. These principles work
together in a dynamic way, maximizing therapeutic efficacy while minimizing
side effects.
Synergism can occur through two mechanisms: pharmacodynamics and
pharmacokinetics. Pharmacokinetic synergism refers to the ability of one herb to
facilitate the absorption, distribution, metabolism, and elimination of other herbs.
(Ponrasu T, 2020)[11] On the other hand, pharmacodynamic synergism studies
the combined effect of active constituents with similar therapeutic activities that
target the same receptors or physiological systems. By acting on multiple targets
simultaneously, a combination of herbal remedies can effectively alleviate the
symptoms and complications caused by diseases.
This is particularly helpful considering that diseases often have multiple factors
and complications, leading to both visible and invisible symptoms. Polyherbalism
offers several benefits that are lacking in single herbal formulations. For instance,
it allows for better therapeutic effects with a lower dose of the herbal preparation,
thereby reducing the risk of side effects.Additionally, polyherbal formulations
provide improved convenience for patients by eliminating the need to take
multiple different herbal formulations simultaneously. This indirectly leads to
better compliance and overall therapeutic effects. Considering the multiple types
of molecules present in polyherbal formulations, they can effectively target
disease complications through different mechanisms, providing a comprehensive
therapy for the condition.

Material and Method:


Collection and authentication The medicinal plants used in the study were
purchased from the local market of Khari Baoli, New Delhi, India. Fresh and
shade dried roots of all selected plants have been authenticated by Botanical
Survey of India, the Northern regional .
Preparation of extract
The collected roots of the polyherbal plant were cleaned, dried under shade and
crushed by a mechanical grinder. The powdered roots of each selected plants
(1000 g) were poured in distilled water until the reach of complete exhaustion.
Successive extracts were separately filtered by using Whatman filter paper and
concentrated at an appropriate temperature (40 °C) on a rotary evaporator and
dried under freeze drier. The percentage yield was found between 20-25% (w/w)
respectively, and the final dried powder was stored in a closed container at a cool
place [15].
Characterization of powder
The biologically potent polyherbal extract powder with a stand for varied physical
properties and micromeritic properties. Extract powdered are heterogeneous
because it was composed of individual particles of different sizes and shapes
randomly interspersed with air spaces and becomes more complicated with
polyherbal [16, 17]. Measurements were carried out in triplicate for each
formulation and presented as the average±standard deviation (SD).
Angle of repose
The flow properties of thoroughly mixed all polyherbal extract powder in the
formulation were determined by calculating the angle of repose by the fixed
height method. A funnel with 10 mm in diameter of the bottom was fixed at the
height of 2 cm over the plain and smooth surface. About 10 gm of a thoroughly
mixed sample was slowly passed beside the wall of the funnel until the tip of the
pile formed and touches the bottom of the funnel. A rough circle drowned around
the pile base, and the radius of the powder cone was measured [18]. The angle of
repose was calculated by the average radius using the following formula given as

Drug content uniformity test
From each formulation, 20 tablets were taken, weighed and thoroughly triturated.
The sufficient powder equivalent to 250 mg of the drug was accurately weighed
and diluted in 300 ml of 0.1N HCl for 10 min with vigorous shaking. Further, this
prepared mixture was diluted with 0.1N HCl to produce 400 ml and then filtered.
10 ml of this filtrate was diluted to 100 ml with distilled water, and the absorbance
was measured at an appropriate wavelength, i.e. Lower Hausner’s ratio-Better
flowability, Higher Hausner’s ratio Poor flowability [18, 19]. Preparation of
polyherbal dispersible tablet max 200-400 nm using UV spectrophotometer [21].
Disintegration time
From each formulation 6 polyherbal dispersible tablets were randomly selected
to determine the disintegration time. The acidic buffer (pH 1.2) used as
disintegration medium and temperature was maintained at 37±0.5 °C. The
average disintegration time of six tablets was noted down for calculation [22].
Dispersion time In vitro dispersion time of polyherbal dispersible tablet was
measured by dropping the tablet in a beaker containing 100 ml of normal water.
Two tablets from each formulation were randomly selected and in vitro dispersion
time was determined. A smooth dispersion was produced, which passes through
a sieve screen with a nominal mesh aperture of 710 µm [23, 24]. In vitro
dissolution study The dissolution testing of polyherbal medicines become
difficult to oversee authority requirements due to widely varying constituents.
The ingredients of polyherbal medicinal products often cover a mixture of
multiple herbal constituents; dissolution method development is much more
complex than for defined single constituent [25]. Stability

Limitation:
Despite these advantages, there are limitations to polyherbal formulations. The
combination of various constituents may lead to chemical incompatibilities,
resulting in instability of the formulation. Therefore, careful consideration is
required when combining different plants with their constituents to ensure
optimal activity. In India, where most Ayurvedic polyherbal formulations are
manufactured and exported, the regulation of Ayurvedic herbal preparation
manufacturing is somewhat less stringent. Despite the existence of the Drugs and
Cosmetic Act, which aims to control the manufacture and quality control of herbal
formulations, toxicity studies and clinical trials are not mandatory for patent
applications or manufacturing licenses granted to Ayurvedic herbal product
manufacturers. polyherbal formulations provide numerous advantages over
single herb remedies. They offer the potential for synergistic effects, lower doses,
improved convenience, and a comprehensive approach to disease treatment.
However, caution must be exercised to ensure chemical compatibility and
stability. Although regulations in India may be more lenient, it is important to
uphold good clinical practice, toxicity studies, and clinical trials to ensure the
safety and efficacy

Conclusion
In spite of the fact that polyherbal detailing is generally utilized in many pieces
of the world, however, the scientific proof is as yet deficient. Numerous
homegrown treatments are yet under in-vivo assessment and have not been
assessed by clinical preliminaries. Besides, well-being assessments, for example,
toxicological investigations have not been performed. Here is a need of time
to assess polyherbal detailing utilizing scientific strategies like clinical
preliminary, conceivable bioactive mixtures, and systems of activity for the
future world.
