Micro tubule Associated Protein Tau(MAPT)

Gene Accession Number: P10636

Introduction:
Tau protein is a crucial component in the human
brain ,key role in stabilizing micro tubules, which are essential for
maintaining structure of nerve cells. Abnormal aggregation of tau is
associated with neuron degenerative disorders including Alziheimer’s
disease.

Function:
It provides structural support and facillitates intracellular
transport , essential component of cytoskeleton.
Primary function is to stabilize micro tubules with in nerve cells,
contributing to the proper function of neurons.

Homology Modeling Summary:

Using Swiss Model, template of high quality is generated with a GMQE
score of 0.15 and QMEAND Global score is 0.44. The selected template
7p6e.1.E. (E591-698) shows 99.32 % sequence identity and 58%
coverage.
BIOINFORMATICS PROTEIN 3D MODELING ASSIGNMENT

Methodology :
1.Sequence Retrieval:
Acquired MAPT amino acid sequences( UniProt : P10636).
2.Homology Modeling:
Utilized Swiss Model with template search against PDB.
3.Template Selection:
Choose template 7p6e.1.E.( SMTL Chain id: ABCDEFGHIJ
PDB Chain id: ABCDEFIJQR).
4. Model Building:
Generate a monomeric model for the selected template 7p6e.1.E
(From Human cell).
5. Structure Assessment:
Achieve a relivable model of GMQE score o.15 and QMEANDISCO Global
score 0.44.
6. Template Evaluation:
The choosen template shows 99.32% seq identity and 58% complete
coverage.

Model results:
-Model 01-7p6e.1.E monomer.
-GMQE:0.15
-QMEANDISCO Global score: 0.44

CONCLUSION:
The template 7p6e.1.E. is AGD Type 2 Tau Filament which
shows how abnormal aggregation of Tau protein cause neuron
degenerative disease Argyrophilic Grain disease. The homology model
