*When “RBC detector clog” error occurs, ignore it. (Press “Reset Alarm.”)
Enter "100" in "Target", press “Calc”, "Save", and then press "Continue".
Repeat until the RBC Clogs value is within "100±1".
When within the specification, press "Close".

(2) On the following screen, confirm whether the Blank value is within the specified range (5000 ±
200). If it is within the specified range, click "Close". If it is out of specified range, adjust the value
according to the following procedure.

When Gain value is decreased,

1) Enter a numeric value (0 to 255) in "Blank Gain". Then select magnification x1, x2 or x4 by
pressing button “▼”.
2) Press "Save".
3) Repeat steps 1) and 2) until "Blank" value is within the specified range (5000 ± 200).
4) After adjustment, press "Close".

(2) On the following screen, confirm that the Blank value is within the specified range (5000 ± 200). If
the value is within the specified range, click "Close". If it is out of specified range, adjust the value
according to the following procedure.

1) Enter a numeric value (0 to 255) in "Blank Gain" and select x1 or x2 by pressing ▼ button.
2) Press "Save". ("Blank" value is updated)
3) Repeat steps 1) and 2) until "Blank" value is within the specified range (5000 ± 200).(If Blank
value can not be adjusted within the specified range, please perform step (3) below for further
adjustment.)
4) After adjustment, press "Close".
(3) If Blank value can not be adjusted within the specified range, when selecting magnification x1 or x2
above, please perform the procedure below for further adjustment.
1) Enter a numeric value (0 to 196) in "Blank Gain" and select x4 by pressing ▼ button.
2) Press "Save". ("Blank" value is updated)
3) Repeat steps 1) and 2) until "Blank" value is within the specified range (5000 ± 200).
4) After adjustment, press "Close".

Remove the checkmark from "Use aspiration sensor", perform blank analysis (CBC mode), and then
perform blood aspiration sensor blank adjustment again.
*Because there is a possibility that span adjustment caused the previously adjusted blank conversion to
change,
Repeat until the Blank value is within the specified range (5000±200).
After performing adjustment, restore the check mark to "Use aspiration sensor".
(5) Enter each assy sheet value into “Target and press “Calc” button.
* For detailed procedure of each parameter, refer to following procedures.
(6) After entering all values, press “Save” button.
(8) After measurement is completed, press “Sensitivity” to confirm that all parameters are within assay
sheet targets. Also confirm that scattergrams are normal.

(3) Enter the value of XN CAL ASSAY SHEET in "Target", and press "Calc".

(4) Repeat analysis. When the values are within the specifications, close the screen.

(7) Check the HGB blank level is 5000 +/- 200 by refering “4.3 HGB background level adjustment”.

1) Before performing calibration, confirm that the mode setting is "Normal".
(Procedure: Select "Service" -> "Service / Factory Tools" -> "Test" -> "Mode" in
order) If it is not "Normal", set it to "Normal".
2) In case that XN-550 is in the sampler mode, switch to the manual mode.
(The mode cannot be switched in the sampler mode.)
* If an error occurs during analysis, stop calibrator calibration, clear the error, and repeat the proce-
dure from the beginning.
* If the analysis results do not satisfy the criteria below, repeat analysis.
Select the items that you want to calibrate by checking or unchecking the checkboxes. A checkbox
cannot be checked when the criteria below are not met.
1) 80% ≤ New correction factor ≤ 120%
2) New correction factor -Current correction factor ≤ ±5%
3) Distribution range ≤ Maximum range
4) Allowable relative error ≤ Relative error ≤ Maximum allowable relative error
If you want to change an automatically calculated correction value, select the "Modify" checkbox
and manually enter the new correction value.
(4) Check the value in the [Calibrator Calibration] (execution) dialog box, and then execute calibration
with [OK].
The WBC target is WBC-D for the XN-CAL displayed value. 2016018
2. Analysis
(1) Press "Explorer" at the top left of the menu
screen.
Calculation equation:
"New correction value (%)" ="Display value" ÷ "Average value of 5 tests" × Current correction value (%)
Entry example : 98.65% ->Round off the 2nd decimal digit and enter 987
(1) In the menu screen, press "Service"
-> "Service settings"
-> "Calibration".

2. Perform analysis
(1) Press "Explorer" at the top left of the menu

(2) Perform 5 tests using sufficiently mixed XN
CAL.
(Remove the XN CAL cap before analysis.)

2. Verify whether the deference between the mean for RBC-He and the target value in the assay
sheet is within ±0.5pg.
3. If the mean for RBC-He exceed the range above, perform the calibration by “Calilbration RBC-
He”.
4. After performing the calibration, run XN CAL again (repeat step 1 - 2).
5. Verify that the data are within the above range. If it fails, repeat the steps until it falls within the
range.
(for RET-He)
6. Verify whether the deference between the Mean for RET-He and the target value on the assay
sheet is within ±1.0pg.
7. If the Mean for RET-He exceeds the range, perform the calibration by “Calibration RET-He”.
8. After performing the calibration, run XN CAL again (repeat step 1, 6).
9. Verify that the data are within the above range. If it fails, repeat the steps until it falls within the
range.
The adjustment below must always be performed after the laser diode is replaced.
In addition, the optical axis must be adjusted after the laser diode is replaced.
(Refer to 4.9 Optical Axis Adjustment)

(2) Use tape to hold the cover switch on FCM BLOCK_ASSY NO.23 in the ON position.
(2) Use the ceramic flathead screwdriver to turn VR1 counterclockwise about 10 times.
(3) Select "Service Menu" -> "Service/Factory Tools" -> "Test"-> "Laser Power", and turn ON the laser.
(4) Place the laser power meter diagonally with respect to the optical axis, and turn VR1 with the
ceramic flathead screwdriver to adjust the laser power output value to the target value.
(5) When you have completed adjustment, press "OK" and turn OFF the laser.
(2) Attach the board cover and FCM BLOCK_ASSY NO. 23 cover.
Caution:
When adjusting the laser power, always wear laser safety goggles (wavelength: 642 nm).

• Hex wrench (1.5 mm, 2.5 mm)

• Jig_ASSY NO. 407 (P/N: AP026516)
(2) Use tape to hold the cover switch on FCM BLOCK_ASSY NO.23 in the ON position.
<Fluorescent latex>
Dispense about 2 drops of PS-FLUORED-L2830-4 into 10 mL of CELLPACK DCL. [AA] [AK]
• The undiluted latex solution tends to coagulate. Shake well before dispensing.
• The solvent can only be used on the day it is dispensed.
• Do not add additional undiluted latex solution.
• Since florescence intensity of LATEX PS-FLUORED-L2830-4 will be reduced when exposed to
light, store at 2 - 8 degrees C in a dark place. Do not freeze latex. [AA] [AK]
• After diluting, pour the liquid into a bottle or container that is shielded from light and use that
container.
(2) Set VOLTS/DIV on the oscilloscope to CH1 FSC: 200 mV/DIV, CH2 SFL:20 mV/DIV.
(4) Set both CH1 (FSC) and CH2 (SFL) to AC. (Refer to steps [1] to [4] below.)
(9) Close the "Optical Axis Adjustment (Rough)" screen with "Close".
(2) Select "Service/Factory Tools" -> "Test" -> "Laser Power", and turn ON the laser.
(4) Move the adjustment screw to adjust the CH1 baseline (DC amount) to its lowest point.
(6) Press "Close" in the "Laser Power" screen to turn OFF the laser.
(2) Select "Service" -> "Service/Factory Tools" -> "Optical Axis Adjustment" -> "Rough".
(3) Dispense 1 mL of forward scattering latex [1] into the reaction unit (center chamber of the 3
chambers) and press the analysis start button.
(3) Make sure that the analysis results are within the range below.
(It is acceptable if the FSC distribution does not have a single peak.)
(6) Make sure that the analysis results are within the ranges below. In addition, the histogram must be
shown in SSC section at the same time. [BO]
(2) Attach the board cover and FCM BLOCK_ASSY NO. 23 cover.
(Before attaching the board cover, remove Jig_ASSY NO. 407, and connect the original wiring to J12.)
(2) Put Sample Adapter onto the sampler adapter holder and close it.
(Only for adjusting Hand height and adapter position.)
(5) Confirm that the tube is in the center of the adapter by touching "Up" and "Down.“ [BE]
(6) Confirm the tube is smoothly caught and released by touching "Release" and "Touch." [BE]

Confirm that the "40" sensor becomes white
while light shield plate moving to left.
Tighten two screws at sensor becomes from

4.10.9 Adjusting Sample Tube Holder Position (Micro tube aspiration position)
(1) Select Menu Screen "Service"
-> "Service /Factory Tools"
-> "Mechanical Adjustment"
-> Select "Tube Holder Adjustment"
(4) Adjust sample tube holder position so that the gap between the center of the micro tube holder
and the front surface of the Sample tube clamp is 2 mm by touching “Front” and “Back”.

4.11.1 Adjusting Sample Tube Holder Position (Micro tube aspiration position)
(1) Select Menu Screen "Service"
-> "Service /Factory Tools"
-> "Mechanical Adjustment"
-> Select "Tube Holder Adjustment"
(3) Adjust sample tube holder position so that the gap between the center of the micro tube holder
and the front surface of the Sample tube clamp is 2 mm by touching “Front” and “Back”.
Observe
When the piercer operation count exceeds 120,000, [Piercer replacement is required.] appears in the
[Help] dialog box. Continuing to use the old piercer will cause tip wear and may result in breakage or
other problems.
A replacement piercer is not included with the instrument. Contact your local Sysmex representative to
purchase a replacement piercer.
(1) Prepare a metal plate, screws, a new piercer and tie wraps for replacement.
The metal plate, screws and tie wraps are provided with a new piecer.

(9) Use nippers to cut the tie wrap holding the tube protruding from the top of the piercer, and remove
the tube.
Caution!
When cutting the tie wrap, take care not to damage the tube. Tube
damage may affect analysis results.
Information
(11) Remove the screw from the right side of the rinse cup (under the piercer) with a Phillips head
screwdriver.
(14) Remove the screws (x2) from the piercer and screw (x1) from the rinse cup with a Phillips head
screwdriver.
(17) Loosely fasten the rinse cup with the screw (x1), and tighten the screws (x2) on the piercer with a
Phillips head screwdriver.
Keep the screw on the rinse cup loose.
(18) Tighten the screw that was loosely tightened in step 17 with a Phillips head screwdriver.
Tighten the screw while lifting the rinse cup so that there is no gap (A).
(21) Tighten the screw on the right side of the rinse cup with a Phillips head screwdriver, and attach 2
tubes to the rinse cup.
(26) After the replacement, if any trouble occurs, please refer to [4.1 Pipette and Piercer Position
Adjustment].
