1CHAPTER 4 ADJUSTMENT

4.1 Pipette and Piercer Position Adjustment


In the menu screen, select "Service
-> "Service/Factory Tools"
-> "Mechanical Adjustment"
-> "Piercer/Pipette Adjustment"

4.1.1 Initial Position

(1) Select "Init. Position".


Press "UP" or "DOWN" until the position of
the piercer (pipette) tip is aligned with the
bottom of the rinse cup.
(2) After completing adjustment, press "Save".

4.1.2 RBC/HGB Chamber position
(1) Remove the cover from Chamber_ASSY NO.149.
(2) Select "RBC/HGB Chamber".
Press "Front" or "Back" to move the position of the piercer tip (needle tip) to the center of the
chamber.
Press "UP" or "DOWN" until the position of the piercer tip (needle tip) is aligned with the top of the
chamber.

(3)) After completing adjustment, press "Save".

4.1.3 WBC chamber position
(1) Select "WBC Chamber".
Press "Front" or "Back" to move the position of the piercer tip (needle tip) to the center of the
chamber.
Press "UP" or "DOWN" until the position of the piercer tip (needle tip) is aligned with the top of the
chamber lid.

(2) When you have completed adjustment, press "Save" and then "Close" to close.

4.1.4 Aspiration position (XN-550 and XN-450 only)
(1) Select "Aspiration Position" (CP type only). Press "UP" or "DOWN" until the position of the piercer
tip is aligned with the top (lower part) of the sample tube holder - (white). (Sample tube holder will
not move automatically. Move the sample tube holder manually as needed.) Record the up/down
pulse. [AJ]

4.2 AD Offset Adjustment [AC]
Perform this sequence after replacing one of the following items.
- PCB No. 30013 (Main PCB)
- FCM Detector
- RBC/PLT Detector
- HDD (Only if mmb backup file cannot be obtained from the former HDD)
"A/D convert Auto" needs to be set as shown below, if the mmb backup file is not available.

4.3 RBC clog level adjustment


Press "Service"
-> "Service/Factory Tools"
-> "Sensitivity Adjustment"
-> "RBC Clogs"
in the menu screen.

*When “RBC detector clog” error occurs, ignore it. (Press “Reset Alarm.”)

Enter "100" in "Target", press “Calc”, "Save", and then press "Continue".
Repeat until the RBC Clogs value is within "100±1".
When within the specification, press "Close".

4.4 HGB background level adjustment [BL]
* After turning on the power, wait about 10 minutes to stabilize the LED light intensity, then
perform the following procedures.

(1) In the menu screen, press "Service"


-> "Service/Factory Tools"
-> "Sensitivity Adjustment"
-> "Sensitivity" and select "HGB" with Channel ▼.

(2) On the following screen, confirm whether the Blank value is within the specified range (5000 ±
200). If it is within the specified range, click "Close". If it is out of specified range, adjust the value
according to the following procedure.

When Gain value is decreased,
the Blank value increases.

1) Enter a numeric value (0 to 255) in "Blank Gain". Then select magnification x1, x2 or x4 by
pressing button “▼”.
2) Press "Save".
3) Repeat steps 1) and 2) until "Blank" value is within the specified range (5000 ± 200).
4) After adjustment, press "Close".

4.5 Blood Aspiration Sensor Adjustment

4.5.1 Blood aspiration sensor blank level adjustment [BL]


* After turning on the power, wait about 10 minutes to stabilize the LED light intensity, then

(1) In the menu screen, press "Service"


-> "Service/Factory Tools"
-> "Sensitivity Adjustment"
-> "Sensitivity" and select "Aspiration Sensor" with Channel ▼.

(2) On the following screen, confirm that the Blank value is within the specified range (5000 ± 200). If
the value is within the specified range, click "Close". If it is out of specified range, adjust the value
according to the following procedure.

(3) If Blank value can not be adjusted within the specified range, when selecting magnification x1 or x2
above, please perform the procedure below for further adjustment.
1) Enter a numeric value (0 to 196) in "Blank Gain" and select x4 by pressing ▼ button.
2) Press "Save". ("Blank" value is updated)
3) Repeat steps 1) and 2) until "Blank" value is within the specified range (5000 ± 200).
4) After adjustment, press "Close".

4.5.2 Blood Aspiration Sensor Adjustment
(1) Always adjust the blood aspiration sensor blank level before performing this procedure.
(2) In the menu screen, press "Service"
-> "Service/Factory Tools"
-> "Sensitivity Adjustment"
-> "Aspiration Sensor".

(3) Analyze sufficiently mixed XN-CHECK Level 2.


(4) Enter "13000" in "Target" and click "Calc".
(5) When the new value appears in "Span", click "Save".
Repeat analysis until the analysis result (Sample-Blank) is "13000 ±500".
(6) After adjusting the value to within the range, click "Close" to close.

Remove the checkmark from "Use aspiration sensor", perform blank analysis (CBC mode), and then
perform blood aspiration sensor blank adjustment again.
*Because there is a possibility that span adjustment caused the previously adjusted blank conversion to
change,

(7) In the menu screen, press "Service"


-> "Service/Factory Tools"
-> "Sensitivity Adjustment"
-> "Sensitivity,"
and select "Aspiration Sensor" with Channel ▼.

Repeat until the Blank value is within the specified range (5000±200).

After performing adjustment, restore the check mark to "Use aspiration sensor".

4.6 Sensitivity Adjustment

a. Adjust sensitivity after registering reagent information.


b. Aspiration sensor and HGB blank adjustment must be completed before sensitivity adjustment.

(1) In the menu screen, press "Service"


-> "Service/Factory Tools"
-> "Test"
-> "Mode".
Select the "Analysis Mode : Calibrator" checkbox.
Select the "Continuous : OFF" checkbox.
-> Press "OK".

(2) Press "Back" until you return to the menu screen.

(3) Analyze sufficently mixed XN CAL.

(4) Press “Sensitivity from “Service.”

(5) Enter each assy sheet value into “Target and press “Calc” button.
* For detailed procedure of each parameter, refer to following procedures.
(6) After entering all values, press “Save” button.

(7) Analyze sufficently mixed XN CAL.

(8) After measurement is completed, press “Sensitivity” to confirm that all parameters are within assay
sheet targets. Also confirm that scattergrams are normal.

4.6.1 WDF sensitivity adjustment


(1) In the “Sensitivity”, set "Channel " to "WDF".
(2) Enter the values of XN CAL ASSAY SHEET in"Target" (WDF-X, Y, Z), and press "Calc".
The Gain value changes.

4.6.2 RET Sensitivity Adjustment

(2) Enter the values of XN CAL ASSAY SHEET in "Target" (RET-RBC-X, Y, Z), and press "Calc".

4.6.3 RBC/PLT Sensitivity Adjustment
(1) In the "Sensitivity" screen, set "Channel " to "RBC/PLT".

(2) Enter the values of XN CAL ASSAY SHEET in "Target" (MCV, MPV) , and press "Calc".
The Gain value changes. Press "Save" to save the new value.

4.6.4 HGB Sensitivity Adjustment
Before performing this step, adjust HGB blank level adjust first.

(1) In the "Sensitivity" screen, set "Channel " to "HGB".

(2) Analyze sufficiently mixed XN CAL.

(3) Enter the value of XN CAL ASSAY SHEET in "Target", and press "Calc".
The Gain value changes. Press "Save" to save the new value.

(4) Repeat analysis. When the values are within the specifications, close the screen.

HGB : Value of XN CAL ASSAY SHEET (for XN-L series) ±0.2g/dL

(5) After adjusting the value, close with "Close".

(6) Press "Back" until you return to the menu screen.

(7) Check the HGB blank level is 5000 +/- 200 by refering “4.3 HGB background level adjustment”.

4.7 Calibration

4.7.1 Whole blood mode (calibrator calibration)

Note: [BM]
1) Before performing calibration, confirm that the mode setting is "Normal".
(Procedure: Select "Service" -> "Service / Factory Tools" -> "Test" -> "Mode" in
order) If it is not "Normal", set it to "Normal".
2) In case that XN-550 is in the sampler mode, switch to the manual mode.
(The mode cannot be switched in the sampler mode.)

1. Display the [Calibrator calibration] dialog box.


(1) In the menu screen, press "Calibration" -> "Calibrator Calibration".

[Calibrator Calibration] (analysis) dialog box

(2) Start manual analysis of sufficiently mixed XN CAL.


Calibrator calibration is performed in manual analysis.
• Discretes to be analyzed are set automatically and cannot be changed.
• With RET analysis license: CBC + DIFF + RET
• Without RET analysis license: CBC + DIFF

* If an error occurs during analysis, stop calibrator calibration, clear the error, and repeat the proce-
dure from the beginning.

2. Manual analysis procedure [AJ]


(1) XN-550 and XN-450
• Place a vial in the sample holder.
• Press the manual analysis start switch.
• Repeat analyzing the calibrator 11 times. Mix the calibrator properly while analyzing.

(2) XN-350
• Insert the pipette until it touches the bottom of the vial, and press the manual analysis start
switch.
• Repeat analysis (total of 11 times)

Mix as needed while performing analysis.

* If the analysis results do not satisfy the criteria below, repeat analysis.

• All analysis results are normal.


• All calibration parameters are the [Limit (%)] value or less.
In the [Calibrator Calibration] (analysis) dialog box, touch the analysis numbers of analysis results
that must be repeated, and with the numbers in the selected state, execute manual analysis again.

(3) When analysis ends, press [OK].


(XN-550/XN-450 only) Remove the vial.

Select the items that you want to calibrate by checking or unchecking the checkboxes. A checkbox
cannot be checked when the criteria below are not met.
1) 80% ≤ New correction factor ≤ 120%
2) New correction factor -Current correction factor ≤ ±5%
3) Distribution range ≤ Maximum range
4) Allowable relative error ≤ Relative error ≤ Maximum allowable relative error

If you want to change an automatically calculated correction value, select the "Modify" checkbox
and manually enter the new correction value.

(4) Check the value in the [Calibrator Calibration] (execution) dialog box, and then execute calibration
with [OK].

(5) Check calibration
After calibration, analyze XN CAL, and make sure the result is within the acceptable limit.
* If any parameters are outside the limit, repeat adjustment.
* "Acceptable limit" is the displayed value multiplied by "Acceptable limit (%)".
* Analysis must be performed in calibrator mode.

The WBC target is WBC-D for the XN-CAL displayed value. 2016018

4.7.2 Dilution mode


1. Instrument settings

(1) Press "Change Mode" at the bottom right of the
menu screen.

The analysis mode selection screen opens.


Select the "Dilution" checkbox, and then close
the screen with "OK".

(2) In the menu screen,


press "Service"
-> "Service/Factory Tools"
-> "Test"
-> "Mode".
Select the "Analysis Mode
: Calibrator" checkbox.
Select the "Continuous
: OFF"checkbox.
-> Press "OK".

(3) Press "Back" until you return to the menu


screen.

* Make sure that the instrument is in manual mode


(4) Press "Manual" at the bottom right of the menu
screen.

The manual analysis screen opens.


(5) Enter any sample number.
(6) Discreet: CBC + DIFF + RET
(7) and then close the screen with "OK".

2. Analysis
(1) Press "Explorer" at the top left of the menu
screen.

(2) Perform 5 tests using a x7 dilution of XN CAL
(500μl) with CELLPACK (3ml).
(Remove the cap from the sample tube before
analysis.)

* Mix the solution before each test.

3. Checking the correction value


Calculate the correction value and make sure it is within the adjustment range below.

Calculation equation:
"New correction value (%)" ="Display value" ÷ "Average value of 5 tests" × Current correction value (%)

Parameters: RBC_PD_CAL, PLT_PD_CAL, HGB_PD_CAL, WDF_PD_CAL, RET_PD_CAL,
RBCO_PD_CAL, PLTO_PD_CAL

Enter the correction values in the parameters of the CAL file.

Entry example : 98.65% ->Round off the 2nd decimal digit and enter 987
(1) In the menu screen, press "Service"
-> "Service settings"
-> "Calibration".

4.7.3 Body fluid mode


1. Instrument settings
(1) Press "Change Mode" at the bottom right of the
menu screen.

The analysis mode selection screen opens.


Select the "Dilution" checkbox, and then close
the screen with "OK".

(2) In the menu screen,


press "Service"
-> "Service/Factory Tools"
-> "Test"
-> "Mode".

Select the "Analysis Mode

(3) Press "Back" until you return to the menu screen.

* Make sure that the instrument is in manual mode.

(5) Enter any sample number.

(6) Close the screen with "OK".

2. Perform analysis
(1) Press "Explorer" at the top left of the menu
screen.
(2) Perform 5 tests using sufficiently mixed XN
CAL.
(Remove the XN CAL cap before analysis.)

* Mix the solution before each test.

3. Check the correction value


Calculate the correction value and make sure it is within the adjustment range below.

4.7.4 Calibration for RBC-He and RET-He at Customer side
Note: Before the calibration for RBC-He and RET-He, calibration for RBC-Y, HGB, and RBC has to
have been completed.
1. Run XN CAL five times consecutively, and calculate mean for RBC-He and RET-He.

(for RBC-He)
2. Verify whether the deference between the mean for RBC-He and the target value in the assay
sheet is within ±0.5pg.
3. If the mean for RBC-He exceed the range above, perform the calibration by “Calilbration RBC-
He”.
4. After performing the calibration, run XN CAL again (repeat step 1 - 2).
5. Verify that the data are within the above range. If it fails, repeat the steps until it falls within the
range.

(for RET-He)
6. Verify whether the deference between the Mean for RET-He and the target value on the assay
sheet is within ±1.0pg.
7. If the Mean for RET-He exceeds the range, perform the calibration by “Calibration RET-He”.
8. After performing the calibration, run XN CAL again (repeat step 1, 6).
9. Verify that the data are within the above range. If it fails, repeat the steps until it falls within the
range.

4.8 Laser Power Adjustment
Caution: When adjusting the laser power, always wear laser safety goggles (wavelength: 642 nm).

The adjustment below must always be performed after the laser diode is replaced.
In addition, the optical axis must be adjusted after the laser diode is replaced.
(Refer to 4.9 Optical Axis Adjustment)

4.8.1 Removing the covers


(1) Loosen the screws on the board cover and FCM BLOCK_ASSY NO.23 cover, and remove the
covers.

(2) Use tape to hold the cover switch on FCM BLOCK_ASSY NO.23 in the ON position.

4.8.2 Laser power adjustment
(1) Set the laser power meter to 633 nm.

(2) Use the ceramic flathead screwdriver to turn VR1 counterclockwise about 10 times.

(3) Select "Service Menu" -> "Service/Factory Tools" -> "Test"-> "Laser Power", and turn ON the laser.

(4) Place the laser power meter diagonally with respect to the optical axis, and turn VR1 with the
ceramic flathead screwdriver to adjust the laser power output value to the target value.

(5) When you have completed adjustment, press "OK" and turn OFF the laser.

• Since florescence intensity of LATEX PS-FLUORED-L2830-4 will be reduced when exposed to
light, store at 2 - 8 degrees C in a dark place. Do not freeze latex. [AA] [AK]
• After diluting, pour the liquid into a bottle or container that is shielded from light and use that

4.9.4 Oscilloscope preparations
(1) Connect the oscilloscope probe to the test points below on the main board.

(4) Set both CH1 (FSC) and CH2 (SFL) to AC. (Refer to steps [1] to [4] below.)

4.9.5 Adjustment of front and rear directions of light emitter


(1) Set the oscilloscope to CH2 (SFL).
(2) Select "Service" -> "Service/Factory Tools" -> "Optical Axis Adjustment" -> "Rough".
(3) Check the Gain values of FSC, SFL, and SSC. If a value requires adjustment, enter the number
and click "Send".

4.9.8 Adjustment of front scattering receiver


(1) Change CH1 (FSC) of the oscilloscope to DC.(Refer to steps [1] and [2] below.)

(2) Select "Service/Factory Tools" -> "Test" -> "Laser Power", and turn ON the laser.

(3) Fixing screws: Loosen 2 screws.

(4) Move the adjustment screw to adjust the CH1 baseline (DC amount) to its lowest point.

* If too far out of position, the DC amount will be reduced.


When moving the adjustment screw, the waveform will change in the order High -> Low -> High.
Move to the position where the center is lowest.

(5) Fixing screws: Tighten 2 screws. (Torque: 10 cN-m)

(6) Press "Close" in the "Laser Power" screen to turn OFF the laser.

4.12 Replacing the piercer, and Adjustment

[Help] dialog box. Continuing to use the old piercer will cause tip wear and may result in breakage or

A replacement piercer is not included with the instrument. Contact your local Sysmex representative to
purchase a replacement piercer.

(1) Prepare a metal plate, screws, a new piercer and tie wraps for replacement.
The metal plate, screws and tie wraps are provided with a new piecer.

(2) Touch the [Maintenance] icon in the [Menu] screen.


The [Maintenance] menu screen appears.

(3) Touch the [Exchange] icon.


The [Exchange] dialog box appears.

(4) Touch [Replace Piercer].

The piercer cycle count is reset, the piercer moves to the replacement position, and the instrument

(9) Use nippers to cut the tie wrap holding the tube protruding from the top of the piercer, and remove

Caution!
When cutting the tie wrap, take care not to damage the tube. Tube
damage may affect analysis results.

(10) Remove the tubes (x2) from the rinse cup at the bottom of the piercer.

(11) Remove the screw from the right side of the rinse cup (under the piercer) with a Phillips head
screwdriver.

(12) Install the replacement metal plate.


Place so that the slit in the metal plate is aligned with the center hole in the rinse cup and the upper
hole in the piercer.

(13) Tighten the screws (x2) on the metal plate with a Phillips head screwdriver.
Tighten the screw on the lower part first, and then the upper part.

(14) Remove the screws (x2) from the piercer and screw (x1) from the rinse cup with a Phillips head
screwdriver.

(15) Remove the piercer.


Dispose of the piercer that has been removed in accordance with local laws and regulations.

(16) Install the new piercer.

(17) Loosely fasten the rinse cup with the screw (x1), and tighten the screws (x2) on the piercer with a
Phillips head screwdriver.
Keep the screw on the rinse cup loose.

(18) Tighten the screw that was loosely tightened in step 17 with a Phillips head screwdriver.
Tighten the screw while lifting the rinse cup so that there is no gap (A).

(19) Remove the screws (x2) from the metal plate with a Phillips head screwdriver.

(20) Remove the metal plate.

(21) Tighten the screw on the right side of the rinse cup with a Phillips head screwdriver, and attach 2
tubes to the rinse cup.

(22) Attach the tube above the piercer and fasten with a tie wrap.
Be careful not to kink the tube when binding the tie wrap. Cut off the excess tie wrap.

(23) Close the right cover and lock.

(24) Turn ON the main power switch.

(26) After the replacement, if any trouble occurs, please refer to [4.1 Pipette and Piercer Position
Adjustment].
