Experiment Title: DETERMINATION of PARACETAMOL FROM SERUM

SAMPLE with SPECTROPHOTOMETRIC ASSAY

Introduction: Paracetamol is a safe and effective analgesic and antipyretic drug.

It is also one of the rare drugs that may cause death by liver necrosis in
overdose cases. Paracetamol is a good alternative of aspirin as it has few
adverse effects. It has no anti-inflammatory effect bu it can be used in
combination with anti-inflammatory drugs to increase their analgesic effect.
Toxicity is observed at minimum, 140 mg/ kg of dose( 10 g for a 70 kg adult).
Nevertheless, toxic dose changes according to the person’s glutathione levels.
That’s why the received dose and toxicity are not strongly related. In general,
liver damage is not observed less than 15 g of doses. Therapeutic dose is 10-20
µ/mL for adults and 7 µ/mL for children

Materials: Hydrochloric acid, urine, distilled water,o-cresole solution,

Ammonium hydroxide

Method: 0.5 mL hydrochloric acid is added to 0.5 mL of urine and heated 10

minutes at 100 °C. Then, 10 ml distilled water, 1 ml 1% o-cresole solution and


4 ml 4M ammonium hydroxide to 1 drop of mixtures are added respectively
Raw and Processed Data: Blue color represents paracetamol presence

Conclusion: Cresole- Ammonia test is very sensitive and positive result is


observed at therapeutic doses. Even a few days after high doses of intake, the
molecule itself and conjugated metabolites may also be determined with this
experiment. In case of a positive result, blood paracetamol levels should be
analyzed
