Single Dose Administration and
Continuous and Multiple Dose Administration

Single Dose Administration occurs when the next dose of the same drug is administered
only after the drug of the previous dose is completely eliminated from the body. Multiple Dose
Administration is when a drug is given repeatedly at intervals shorter those required to
completely eliminate the drug of the previously given dose.

1. A physician wants to administer an anesthetic agent at a rate of 1.5mg/hr by IV infusion. The


elimination rate constant 0.095 hr –1 and the volume of distribution (one compartment) is 9 L.
What loading dose should be recommended if the doctor wants the drug level to reach 20
µg/100mL immediately?

2. What is the concentration of a drug 6 hours after administration of a loading dose of 10 mg


and simultaneous infusion at 2mg/hr (the drug has a t1/2 of 3 hr and a volume of distribution of
10 L)?

3. A fifty-year old male patient weighing 235 pounds was given an oral dose of Doxycycline
(0.5mg/kg) every twelve hours for one week. From the pharmacokinetic profile of the drug, it is
about 90% bioavailable and has an apparent volume of distribution of of 1.8L/kg. The
elimination half life of the drug is 24 hours. The absorption rate constant is 1.0 1/hr. From this
