CALIBRATING AN

EYEPIECE GRATICULE
Procedure
1. Move the objective lens to the lowest
power (X4, X5, X10 or X15).
2. Place the stage micrometer on to the
stage, making sure that it is in the slide
holder correctly.
3. Using the low-
power
objective,
focus the
microscope on
the stage
micrometer.
4. Rotate the
eyepiece and
move the slide to
superimpose the
scales of the
eyepiece graticule
and the stage
micrometer .
5. Note the smallest
division on your
stage micrometer.
Here it is equal to
100 m.
5. Count the number of stage divisions
equal to a certain number of eyepiece
graticule divisions.
6. Calculate the distance between each
eyepiece division.

Sample calculation
Compare the two Scales

____ eyepiece divisions = ____ *stage divisions

____ stage divisions = _____ mm
1 stage division = ____mm
____ *stage divisions = ____mm
____ eyepiece divisions = ____mm
1 eyepiece division = ____ mm or ____m
Sample calculation
Compare the two Scales

7 eyepiece divisions = 40 *stage divisions

100 stage divisions = 1 mm
1 stage division = 0.01mm
40 *stage divisions = 0.4 mm
7 eyepiece divisions = 0.4 mm
1 eyepiece division =0.4/7= 0.57 mm or 570 m
7. Repeat for X40 objective

X40 magnification
____ eyepiece divisions = ____ *stage divisions
____ stage divisions = ___mm
1 stage division = ____mm
____ *stage divisions = ____mm
____ eyepiece divisions = ____mm
1 eyepiece division = ____ mm or ____m

CALIBRATE THE EYEPIECE FOR THREE OBJECTIVE
LENSES
(x4, x10, x40) (answers in next
slide)

Example 1 - Objective magnification X4.

Suppose the full length of the reticle scale
covered 25 divisions of the stage micrometer.
Example 2 - Objective magnification X10.
The reticle scale now covers 10 divisions of
the stage scale.
Example 3 - Objective magnification X40.
The full length of the stage scale only spans
the first 40 divisions of the reticle scale.
Answers
EXAMPLE 1
The full length of the reticle scale is equivalent to (25 x
0.1mm) = 2.5mm long.
For an eyepiece reticle with 100 divisions, each division will
measure 25m at the stage for this magnification
EXAMPLE 2
The the full length of the reticle scale is equivalent to (10 x
0.1mm) = 1mm long Magnification
Each division will measure 10m at the stage for this
magnification
EXAMPLE 3
The the first 40 divisions of the reticle scale is 0.1mm long.
So each reticle division is equivalent to (0.1mm / 40) = 2.5m.
For an eyepiece reticle with 100 divisions the full length of the
reticle scale equals 250m (0.25mm) at the stage for this
magnification.
In summary you can apply these
conversion factors to state what each
division of the eyepiece reticle is
measuring for a selected magnification.
X4
1 division = 25m
X10
1 division = 10m
X40
1 division = 2.5m
