Impregnation

 Impregnation is replacement process of clearing


agent from the tissue and replacing it with
paraffin.
 Clearing agent remaining can be crystallized in a
tissue so that when cut with a microtome tissue
will tear.
 Impregnation technique :
 Paraffin/ paraplast I : 2 jam;
 Paraffin/ paraplast II : 1 jam;
 Paraffin/ paraplast III : 2 jam.
Preparation:
•Microtome knive
•Microtome blade (disposable)
• Place the knife on the microtome with a
certain angle.
• Place the blocks of paraffin to be cut on the
holder by using a hot spatula.
• Place the following block holder in place in
microtome preparations.
•Prepare coated slides:
•Albumin (putih telur);
•Gelatin;
•SuperFrost
•Poly-L-Lysine
•Waterbath contain hot water (adjust
with paraffin boiling points)

Sectioning Technique
 thickness + 5-10 micrometer
 Adjust the distance of preparations that are held by
the holder in the direction the blade as close as
possible;
 Move the rotor (rotation) on microtome rhythmically,
Dispose of paraffin ribbons without tissue;
After a piece of tissue, cut into blocks preparations
carefully;
 move carefully with tool, from water in the waterbath;
 After the paraffin ribbon terkembang well, stick to the
glass object paraffin;
 Keep glass objects contain pieces of paraffin and tissue
until the time to stain
