DRUG –DRUG interactions based on

alteration in pharmacokinetics of
the drug
( METABOLISM—EXCRETIONS)
ROLL NO- 16
NAME- DEVANG GANGWANI
FINAL YEAR BPHARM ( 2018 )
DEFINATION:-
A drug interaction is a situation in which a
substance (usually another drug) affects
the activity of a drug when both are
administered together.

 Effects can either be:-1) synergistic

2) antagonistic
PHARMACOKINETIC INTERACTIONS
ARE CLASSIFIED AS:-
 ABSORPTION INTERACTIONS

 DISTRIBUTION INTERACTONS

 METABOLISM INTERACTIONS

 EXCRETION INTERACTIONS
METABOLISM( BIOTRANSFORMATION)
INTERACTIONS
 Are those where the metabolism of the object drug is
altered.
Mechanism of metabolism interactions include:-

1)ENZYME INDUCTION ( INCREASE IN ENYME HEPATIC ACTIVITY

THAT RESULTS IN GREATER METABOLISM OF THE DRUG )

2) ENZYME INHIBITION ( DECREASES IN ENZYME HEPATIC

ACTIVITY THAT RESULTS IN REDUCED METABOLISM OF THE DRUG)
EXAMPLES OF ENZYME INDUCTION AND ENZYME
INHIBITION:-
EXCRETION INTERACTIONS:-
 Where the excretion pattern of the object drug is
altered .

Major mechanism of excretion interaction are:-

1) ALTERATION IN RENAL BLOOD FLOW

2) ALTERATION OF URINE PH
3) COMPETITION FOR ACTIVE SECRETIONS
4) FORCED DIURESIS
