SHOT GUN SEQUENCING
 Fred sanger developed.
 By using chain termination method 100 to 1000 base
pairs can be sequenced but for shot gun seq. large
seq.of genomes can be done.
 It is a technique for determining the sequence for the
entire chromosomes and entire genomes based on
producing random fragments by finding overlapping
ends.
 It is not a unique one, after all used for breakage of
large number of DNA genomes can be produced.
STEPS
 Multiple copies of the same chromosome are
prepared.
 Huge DNA are broken in smaller fragments
1kb.
 By mechanical shearing or chromosomal
shearing.
 1kb fragments are cloned in to plasmids
 Seq them individually by dideoxy or pyrroxy
method
 A computer combines the fragment pieces.
