CULTURE MEDIA

 An artificial preparation which contains all the


essential elements and nutrients needed by
the microorganisms to grow.
Characteristics:
 Must give satisfactory and rapid growth from
single inoculum.
 Should be reasonably cheap and easily
reproducible
 Should enable to demonstrate all
characteristics in which we are intrested.
CLASSIFICATION OF CULTURE

1. CONSISTENCY 1) Broth

REQUIREMENT 1) Simple media
2) Enriched media

B) ANEROBIC
SIMPLE MEDIA

 It is the type of media which are used to grow microorganisms that do not
have special nutritional requirements. It fulfills basic requirement of bacteria.
 Also called basal medium.

 EXAMPLES
1. Nutrient broth
2. nutrient media.

 Uses
- Preparation of enriched media.
- To maintain stock cultures of control strains of bacteria.
- For subculturing pathogens.
DIFFERENTIAL MEDIA

 It is the type of medium which has substance incorporated in it enabling


it to bring out different characteristics of bacteria and thus helping to
distinguish between them.
 EXAMPLES:
1. Blood agar differentiate al pha hemolysis and beta hemolysis. It is
due to the production of hemolysin by the bacteria.
2. MacConkey’s agar differentiate lactose fermenter and non-lactose
fermenter.
ENRICHED MEDIA
 Certain organisms can’t grow on the ordinary media . They require
growth promoting ingredients such as blood, serum, glucose, egg,
Vitamins, yeast extract etc. The media containing ingredients
enhance their growth.
 EXAMPLES:
1. Blood agar
Composition:
- Melted agar
- Sterile defibrinated blood: 5-10 %
- Beef extract
- Peptone water
- NaCl

2. Chocolate agar.
Composition same as blood agar
SELECTIVE MEDIA
These are the culture media which allows the growth of one type
of bacteria and inhibit the growth of another type of bacteria e.g.
1. MacConkeys media
-It allows the growth of G-ve bacteria and inhibit growth of G+ve bacteria
2. CLED
-It is used for the urinary tract pathogens / used for the microorganisms of UTI
- Urine sample is cultured only on CLED
- Composition:
Agar, Beef extract, Bromothymol blue, Casein enzymatic hydrosylate, L-
cystine, Lactose, Peptic digest of a normal tissue
3- L J media
- Egg based media
- Used for the mycobacterium
- Sputum specimen used for the L J medium
- 4-6 wks take by M.T.B to grow (slow growing )
