OLANZAPINE

OLANZAPINE
• is a thienobenzodiazepine derivative

• structurally similar to clozapine

• effective in treating schizophrenia and acute manic episodes, and in


preventing recurrence of bipolar disorders.

• Can be used in combination with fluoxetine (Symbyax) for treatment-


resistant depression.

• Not incidated as monotherapy treatment for treatment-resistant


depression.
PHARMACOLOGY
• 85% of oral OLZ dose is absorbed in GI tract and 40% is
inactivated by first-pass hepatic metabolism, with oral
bioavailability of about 60%.

• Peak plasma concentration reached in 5 hours

• Mean half-life of 31 hours (21-54 hours) in healthy individuals.

• Approximtely 93% bound to plasma proteins, mainly albumin.

• It is given in once-daily dosing.

• Available in 2.5, 5, 7.5, 10, 15, and 20mg oral and Zydis
form (ODT).

• Initial dosage for treatment of psychosis is usually 5 or


10 mg.

• For acute mania, 10 or 15mg are given once daily.


DRUG
INTERACTIONS

• Fluvoxamine (Luvox) and Cimetidine (Tagamet) increase,


whereas cabamazepine and phenytoin decrease serum
concentrations of olanzapine.

• Ethanol increaess olanzapine absorption by more than 25%,


which leads to increase sedation.

• OLZ has little effect on the metabolism of other drugs.


SIDE EFFECTS
